Effect of Paxirasol aerosol on the clinical condition and objective functional respiratory parameters of obstructive respiratory tract disease patients.

Paxirasol aerosol applied in daily 3 x 5-puff doses was well tolerated by the examined patients. In the course of the 21-day therapy the laboratory findings did not change-haemopoetic or liver injury was not observed. On two-thirds of the patients the amount of sputum decreased in response to Paxirasol aerosol but the symptoms accompanying expectoration did not change notably. Among the objective respiratory parameters FEV1 values increased and Rtot values decreased. These changes may be observed during the first third of therapy. There was a continuous and linear increase in FEV values characteristic of respiratory pressure conditions. This phenomenon is not characteristic alone of the changes of values of obstructive diseases patients. The authors recommend the use of Paxirasol aerosol for the treatment of respiratory tract disease patients.

DNA distribution pattern and comparison of quantitative data in breast tumours.

Aspiration cytology smears from 24 breast alterations were subjected to cytophotometry and TV image analysis. The smears were grouped according to histological diagnosis and cytological pattern. Cell cycle parameters and DNA indices were calculated from the decomposition of the DNA histograms. Based on quantitative morphometric features the TV image analyser distinguished the benign cells from the malignant ones. In benign cases its diagnostic accuracy is of 80%. Cytological Grades 1, 2 and 3 are also identifiable by this system. Cytophotometric measurements were also performed with 22 invasive breast cancers with known 5-year survival. The cell cycle parameters were related to the survival data. From the point of view of survival only the ratio of the S plus G2 phase cells of all the parameters examined proved to be significant.

[DNA cytometry and morphometry, based on TV image analysis, in the diagnosis of gastric carcinoma].

Cytological smears from gastric biopsies were studied by two types of TAS. Böcking's algorithm and a great number of form factors were used to separate the intact cells from the tumourous ones. The numerically expressed density features and morphometric parameters are able to indicate the possibility of malignant transformation even in dysplasia. These values become much higher in tumours and lead to unequivocal diagnosis. Both types of TAS render valuable help in assessing cases not reliably diagnosed by routine staining procedures.

Endocervical cytology: characterization of the cells by image analyzer.

Authors report on 359 patient's endocervical smears taken by cytobrush. Stromby's endocervical brush did not cause bleeding disturbance or preventing colposcopic examination. The smears contained numerous, well evaluable squamous and endocervical cells. Out of seven cytologically positive cases three proved to be negative by colposcopy. Histology, however, confirmed the cytological diagnosis. Cytology was followed by histology in 42 cases. Cytological and histological diagnoses were compared, there was one false negative. For the more precise characterization of endocervical cells 13 features were measured by a Robotron type image analyzer and the results were statistically analyzed. The respective cell groups can be separated from each other by morphometric and densitometric parameters. Results of this study have produced a data base that can be extended further on. It may give help in case of differential diagnostic problems since it is capable of distinguishing certain cell groups, consequently these data can be incorporated into a diagnostic program.

Fibronectin determination in pregnancy.

The levels of fibronectin were determined by immunoturbidimetric assay in two populations: (a) plasma of healthy nonpregnant and pregnant women, and in amniotic fluid of healthy pregnant females; (b) plasma of umbilical cord blood of healthy newborns and of newborns with sepsis. Fibronectin concentrations of amniotic fluid showed a significant decrease during pregnancy, but the changes of plasma fibronectin levels were not significant in this period. In newborn sepsis, the levels of plasma fibronectin were significantly decreased. We did not find a significant difference between the fibronectin concentration of umbilical cord blood of premature infants compared to mature infants.

Ultrastructural comparative investigations on the hypophysary cells in fresh-water teleosts under natural conditions. The glycoproteic thyreotrope cells.

The structural und ultrastructural characteristics of the glycoproteic hypophysary cells - considered as TSH producers - were studied with optical and electron microscopical means in 7 species of freshwater teleosts: Leuciscus cephalus L., Barbus meridionalis petényi H., Chondrostoma nasus L., Gobio gobio carpathicus Vlad., Phoxinus phoxinus L., Nemachylus barbatulus L. and Cobitis taenia L., during the reproductive period and in the period of sexual rest. These cells are present in a relatively reduced number in the hypophysis, forming islets of 2...3 cells, each. They are of small dimensions (the maximal diameter being 7 micron) having polyhedral form with prolongations or an ovoidal form. The cellular nucleus is of large dimensions (to 5 micron) often deformed or segmented. The cytoplasm, during the reproductive period, contains numerous moderately electrondense secretory granules. The mean diameter of the granules does not exceed 1200 A in any of the species studied. In the reproductive period the mitochondria are swollen and prolonged, the Golgi complex more developed and in some species a large number of microvesicles is spread around this complex. The researches proved the tight connection of these cells with the nerve fibres which contain or not neurosecretory peptidergic granules. The more intensive elaboration of the granules during the reproductive period is discussed.

Comparative ultrastructural study of the neuro-intermediary lobe in fresh-water teleosts under natural conditions.

A comparative study of the hypophyseal neuro-intermediary lobe (NIL)j is 8 species of fresh-water teleosts (Cyprinidae) was performed by means of light and electron microscopy. NIL is well developed in all species and consists of two types of glandular cells, three types of peptidergic nerve fibres, pituitary cells and blood cappillaries. Type A glandular cells (lead-haematoxylin staining cells) are elongated and small (8...12 micrometer/ 3...6 micrometer). They contain a large number of secretory granules with an average diameter of 160 nm. Type B cells (PAS-positive) are larger (13...16 micrometer/ 7...9 micrometer), constituting the NIL major substance. They present two variants: cells with a clear cytoplasm (light cells) and cells with a dark cytoplasm (dark cells). All the B-type cells include numerous vesicles (300...350 nm in diameter) with a slight electrondens content. In some of them a small number of intense electrondens secretory granules also appear. Type I peptidergic nerve fibres (PNF) are the most frequent; they contain numerous granules with an average diameter of 170 nm. Type II PNF are less frequent and contain granules with an average diameter of 170 nm. Type II PNF are less frequent and contain granules with an average diameter of 220 nm. The least frequent are type III PNF. Their secretory granules have an average diameter of 200 nm. The fenestrated aspect of the endothelial wall of the capillaries and the local discontinuities of the perivascular space indicate the existence of an especially tight neuro-cascular relation. The cytological "picture" of NIL is very similar in all species and characteristic for the period of the autumnal sexual repose of these animals.

[Ultrastructural modifications in the hypophyseal-adrenocortical system in rabbits during experimentally induced parodontosis (author's transl)].

During parodontosis induced experimentally by irritation of dental pulp with carbolic acid in rabbits, the participation in the pathological process of adenohypophyseal ACTH-producing cells and of adrenocortical fasciculata cells was studied. Ultrastructural alterations suggesting stimulation of ACTH and glycocorticoid synthesis were observed. The data obtained prove that via afferent pathways to the central nervous system, irritation of the dental pulp induces morphofunctional alterations in the hypothalamo-adenohypophyseal neurosecretory system, by means of functional perturbation of the cerebral cortex leads to hypothalamus (CRF) leads to adenohypophysis (ACTH) leads to adrenal cortex (glucocorticoids)axis.

Diabetic lymphangiopathy. An optical and electron microscopic study.

The lymphatic capillaries in the tegument of the dorsal side of the leg in patients with diabetes mellitus, and especially in those with clinically manifested macroangiopathy, present morphopathological modifications at the level of the endothelium and the basal membrane. Ultrastructurally, the modifications appear as dilatations of the intercellular space, dislocation or desquamation of the endothelium, an exaggerated growth of the micropinocytic vesicles, the appearance of the intracellular vacuoles and of the lysosomes. These modifications point to an increase in the endothelium permeability, which is an important phenomenon in the diabetic microangiopathy.
