Human platelet monoamine oxidase activity in iron-deficiency anaemia.

1. The activity of monoamine oxidase, when assayed with four substrates, was significantly lowered in platelets prepared from the blood of patients with iron-deficiency anaemia. 2. Treatment with oral iron preparations restored platelet monoamine oxidase activity to normal in those patients whose serum iron concentrations also returned to normal. 3. Platelet monoamine oxidase activity remained low if treatment failed to restore serum iron concentration to within normal limits.

Adult

Reversal of UDP-galactose 4-epimerase deficiency of human leukocytes in culture.

Stimulation with phytohemagglutinin of the leukocytes from six of the seven known individuals with UDP-galactose 4-epimerase (= UDP-glucose 4-epimerase; EC 5.1.3.2) deficiency consistently resulted in the appearance of epimerase activity in the cultured cells. A long-term lymphoblast culture derived from one proband also contained an active epimerase enzyme. A comparison of the properties of this enzyme with those of epimerase produced by control lymphoblast lines revealed comparable Km values for UDP-galactose and NAD and identical behavior on polyacrylamide electrophoresis. However, a difference in the NAD requirement for heat stability at 40 degree provided some evidence for a structural defect in this enzyme. Possible explanations for the appearance of UDP-galactose 4-epimerase activity in stimulated lymphocytes include an increased rate of synthesis of a mutant enzyme and a derepression of an epimerase locus during lymphocyte transformation.

Carbohydrate Epimerases

The role of colostrum in glomerulonephritis of Finnish Landrace lambs: effect of cross-fostering newly born Finnish Landrace and Cheviot lambs.

Newly-born Finnish Landrace and Cheviot lambs were cross-fostered to determine the role of colostrum in the development of mesangiocapillary glomerulonephritis of the Finnish Landrace breed. Severe glomerulonephritis occurred in four Finnish Landrace lambs reared by their own dams. One of six siblings of affected lambs, all reared by Cheviot foster mothers, developed a milder focal form of the disease. No glomerulonephritis occurred in three Cheviot lambs reared by Finnish Landrace foster mothers whose own naturally reared lambs died of the disease. An artificially reared colostrum-free Finnish Landrace lamb became severely affected, though its naturally reared sibling had no renal disease in sequential biopsies. It is concluded that colostrum has no primary role in the development of the disease.

Animals

Normal blood-gas values in lambs during neonatal development and in adult sheep.

Arterial oxygen (PaO2) and carbon dioxide (PaCO2) tensions and pH were determined in 20 lambs during development from two weeks to five months of age and in 34 adult sheep. Throughout the period the lambs' mean PaCO2 (33.1 mm Hg) and pH (7.46) values were similar to those determined in the adult sheep (PaCO232.1 mm Hg; pH 7.48). The lamb's mean PaO2 increased from 63.3 mm Hg at two weeks of age to about 80 mm Hg at eight weeks and by 20 weeks was similar to the mean PaO2 (88.0 mm Hg) of adult sheep.
