The silver-staining technique--a tool for characterizing lymphocyte populations in mammalian peripheral blood.

Nucleolus organizer regions (NORs) of the peripheral blood lymphocytes (PBL) of 9 mammalian species were analyzed by means of a silver-staining procedure. Species-specific NOR patterns were demonstrated. The number of NOR chromosomes was positively correlated with the nucleolar coefficient, and negatively correlated with the relative frequencies of uninucleolar cells in PBL interphase.

Lectin-induced lymphocyte stimulation in pigs--a kinetic study of NORs.

Early reactions of rRNA-cistrons in lectin-stimulated PBL cultures of domestic pigs are shown. Interphases showing 3 or more clusters of stained areas were attributed mainly to cells after first division. Clear differences are shown between PHA- and ConA-induced NOR-expression patterns compared to the patterns in PW-stimulated cultures.

Spontaneous rosette formation of pig and guinea pig thymocytes and peripheral blood lymphocytes: influence of proteases, oxidising and reducing agents, cytochalasin B and carbohydrates.

Spontaneous rosette formation (SRF) of pig and guinea pig thymocytes (Th) and thymus derived peripheral blood lymphocytes (PTL) was tested under different experimental conditions. Treatment with different proteases (trypsin, chymotrypsin, protease type VII) showed that SRF of pig Th and PTL is inhibited to a much higher extent than that of guinea pig Th and PTL. Cytochalasin B (CB) inhibited SRF of PTL of both species to a higher degree as compared to Th. Different carbohydrates neither influenced SRF of Th nor of PTL in both species indicating that simple carbohydrates are not the recognized component of SRF receptors. Treatment with oxidising and reducing agents did not influence SRF.

[The diagnostic value of computerized tomography in mediastinal diseases depending on their localization].

The diagnostic ranking and importance of computerised tomography was examined in 116 patients with abnormal findings in the mediastinal region, and was compared with conventional, non-invasive x-ray examinations. This method also yielded significant CT-specific additional information in the anterior mediastinum in 74.4% of the cases, in the mesomediastinum in 69.1%, in the posterior mediastinum in 79.2% in the upper thoracic aperture in 94.4% and in the paracardial region in 100% of the cases. The hilar region is an exception; in 68.8% of the cases, both methods were rated equal, whereas in 28.1% of the cases assessment via the CT method was even inferior. The specific additional information furnished by the CT method justifies a wider application of CT in solving the following problems concerning the mediastinum. -- clarification of a suspected but not yet established space-occupying growth in the mediastinum, before using invasive diagnostic methods such as mediastinoscopy and angiography; -- staging of an already identified primary mediastinal tumor of malignant lymphoma; -- in case of suspected changes in the large mediastinal vessels before employing angiography; -- on-target in the following diseases: bronchogenic carcinoma (because of improved pretherapeutic staging according to the TNM system), myasthenia gravis and identification of thymoma, hyperparathyroidism with suspected dystopic parathyroid glands.

[The influence of fractionation on the survival of patients following primary irradiation of bronchial carcinomas (author's transl)].

153 patients with a local carcinoma limited to the thoracic region underwent irradiation with weekly doses of 1 x 5.0 Gy TD, 2 x 5.0 Gy TD, 4 x 2.5 or 5 x 2.0 Gy TD up to a total dose of generally 60.0 Gy TD. Collectives comparable in view of stages and histology did not show significant differences in 3-year survival rates, though a tendency toward low doses was recognizable.

Detection of nucleolus organizer regions (NOR) in the chromosomes of the domestic pig (Sus scrofa domestica L.).

In the domestic pig (Sus scrofa domestica L.) nucleolar organizer regions (NOR) were detected by a combined silver-Giemsa method (Ag-G). The main sites of NORs are the secondary constrictions of chromosomes 8 and 10. Sometimes an additional NOR was observed near the centromere of 1 homologue of chromosome 11. Association of NORs were seen only between chromosomes 10 and at a very low frequency.

[Validity of scintigraphic and endocrinological methods in classifying autonomous adenomas of the thyroid (author's transl)].

The validity of various scintigraphic and endocrinological methods was assessed in order to classify confirmed autonomous adenomas (AA) and to differentiate AA from euthyroid and hyperthyroid goitres. Methods used included: Measurement of 131J-uptake 6, 24 and 48 hours after 131J-application, before and after suppression by T3, thyroid scanning, count-rate measurement in the AA and the paranodular tissue, and determination of T4. All patients received 131J-therapy (40,000 rads). In 8 out of 30 scintigraphically compensated autonomous adenomas (CAA) there was no TSH response to TRH. 12 cases showed a subnormal TSH response and 10 patients had a normal TSH response. T4-values were increased in only 2 cases. Out of 73 decompensated autonomous adenomas (DAA), 66 cases (90%) had no TSH response and 7 cases showed only a subnormal response. In 48% of DAA T4-levels turned out to be in the hyperthyroid range. Both, scintigraphic and endocrinological tests were found to agree in most cases of DAA. However, the diagnosis of CAA could only be proved on the basis of a scintigraphically confirmed suppressive effect of T3 on the paranodular tissue. Thus, in the individual case, separation of CAA from DAA turned out to be impossible using the results of TRH-test alone.
