We present a general model for the evolution of chromosome-specific satellite DNA subfamilies. Sus scrofa domestica has a bimodal karyotype with two autosomal subsets of 12 meta-/submetacentric (Mc) and 6 acrocentric (Ac) chromosome types (Mc and Ac "subgenomes"). We show that the centromeric heterochromatin is characterised by two distinct satellite DNA families designed Mc1 and Ac2. Mc1 is a diverse satellite family of the Mc subgenome of which certain members with a 100 bp repeat unit are found to occur at the pericentromeric regions of each Mc autosome, while others are chromosome-specific, e.g. clone Mc pAv1.5, a higher order repeat variant, which hybridises specifically to chromosome 1. Ac2 is a homogeneous satellite occurring at the subterminal pericentromeric regions of all Ac autosomes. DNA sequence analyses showed that all clones investigated are built up from a 14 bp repeat unit which is highly conserved. In situ hybridisation to meiotic pachytene nuclei revealed a distinct spatial arrangement of the Ac2 centromeric satellite.
To study whether specific DNA sequences are associated with nuclear membranes, residual DNA was extracted from DNase-treated nuclear envelopes prepared from erythrocytes of adult chickens (Gallus domesticus). This DNA was then blunt-end ligated into a bacterial plasmid vector. DNA blot analysis and nucleotide sequence determination revealed that approximately 30% of the cloned fragments consisted of different multiples of a 41-42 bp tandemly repeated, partially symmetrical sequence. In situ hybridization to chicken chromosomes demonstrated that the sequence was located primarily on microchromosomes, although some hybridization was also observed to macrochromosomes 7 and 8. Digestion of chicken DNA with any of a number of restriction enzymes did not completely reduce the intensity of a high molecular weight band to which the repeated sequence hybridized. These results, along with those obtained from in situ hybridization, suggested that many copies of this sequence are organized into large tandem arrays, and are not dispersed in many shorter repetitive blocks throughout the chicken genome. Although the repetitive sequence constituted approximately 10% of the chicken genome, it did not hybridize to quail or turkey DNA.
The efficacy and safety of a vaccination during anaesthesia was examined in 20 puppies from 8 litters of one kennel. The animals were vaccinated in a state of reflex-free anaesthesia (neuroleptanalgesia in combination with halothane, nitrous oxide and oxygen). 20 puppies from the same litters were used as controls. A parvovirus living vaccine (Canimed) and a rabies vaccine from inactivated viruses (Rabisin) were used for the vaccinations. The titers of neutralizing antibodies against rabies virus were significantly lower in the group of anaesthetized animals compared to the control group on the 10th and 20th day p.vacc. The average titers of antibodies against parvovirus of both groups, however, do not allow any statistically significant statements. It is nevertheless remarkable that 6 of the anaesthetized puppies (three different litters) showed no increase in antibodies. The results of the investigations lead to the recommendation not to vaccinate anaesthetized puppies. The risk of reducing the efficacy of vaccinations with vaccines from inactivated agents is greater than with living vaccines.
Kennel-specific oral E. coli vaccines were tested for efficacy and side effects at breeding and boarding kennels with severe diarrhoea problems. Oral vaccines contained heat inactivated E. coli bacteria from specific kennels and were given once daily for 14 days. Oral vaccines were administered directly orally to puppies while for adult dogs vaccines were mixed with daily food rations. In breeding kennels with 4 to 10 week old puppies suffering from non-fatal diarrhoea, the oral immunization led to a decrease in morbidity from 86.5% to 0%. In kennels with some cases of fatal diarrhoea the rate of morbidity decreased from 45% to 21% and the mortality rate from 25% to 10.3% after using the vaccination. By vaccinating adult dogs in boarding kennels the morbidity rate dropped from 83.5% to 6.5% and the mortality rate from 4.1% to 0.5%. The kennel specific oral E. coli vaccine was found to be free of side effects. No adverse effects were observed in either puppies or adult dogs.
60 patients with suspected lesion of the rotator cuff were examined by digital subtraction arthrography and the results were compared to ultrasound. The dynamic study of digital subtraction arthrography visualised in 22 of 23 complete or incomplete cuff tears the exact location of the extravasation of the contrast medium. All diagnoses were confirmed by surgery. Problems of conventional arthrography were not seen by the digital method.
31 patients with advanced renal cell carcinoma underwent abdominal MRI which were examined by an independent experienced radiologist and compared with the intraoperative findings. 12 of 13 vena caval tumor thrombi were diagnosed correctly, 1 small tumor thrombus in a case with large retroperitoneal lymph nodes compressing the vena cava could not be detected. The extension of the tumor in the vena cava could exactly be described in topographic relation to liver, diaphragm and right atrium which is important for planning the operative procedure.
An eight-year-old female cat developed a skin neurofibroma. The cytogenetic evaluation of the tumour cells showed the presence of a high percentage (16.4 per cent) of trisomic cells. The trisomy concerned chromosome number D2.
The results of a prospective follow-up study comprising 52 patients with central tumours of the lung submitted to endoluminal irradiation are reported. Additional external irradiation was possible in 30 patients (group A), but not in 22 patients (group B). Satisfactory immediate results were observed, together with a one-year survival rate of 39.3% (group A) and 12.5% (group B).
Experimentally produced monozygotic twins, natural opposite sex blood chimeras (freemartins), and several pedigrees were used to evaluate the genetic influences on the nucleolus organizer region (NOR) patterns in cattle. In monozygotic twins, the NOR patterns of both twins are extremely similar. In chimeras, NOR patterns of genetically identical, peripheral blood lymphocytes (PBL) from the two partners resemble each other. In contrast, genetically different PBL (sib organ) differ significantly in the same environment. A high heritability of the individual NOR patterns is also demonstrated in our 23 pedigrees. In conclusion, our data demonstrate that variation in NOR expression is predominantly due to genetic factors.
Chromatin configurations of cattle oocytes from different follicle sizes were classified in three chromatin categories. Early diplotenes, dictyate-like structures, more progressed diplotenes and early diakinesis and metaphases were found. In follicle group A (0.4-0.9 mm) immature diplotenes predominated (82.3%), and a rather low maturation competence was observed (12.1%). With increasing follicle size the maturation competence was improved to 47.8% metaphases II in follicle group C (3-8 mm). Impressive CMA3 positive blocks were visualized especially in early diplotenes. Heterochromatic CMA3 positive clusters surrounded the nucleolus.
Computed tomography (CT) was performed in 142 patients thought to have an endobronchial tumor based on clinical or radiologic grounds. In 121 patients an endobronchial mass was confirmed at bronchoscopy with biopsy or at surgery. The CT scans were evaluated independently by two experienced observers (A and B). For statistical purposes the result in each single bronchus from the level of the trachea to the segmental bronchi was considered separately. A total of 361 abnormal and 1,413 normal bronchi were confirmed with bronchoscopy or surgery. Observers A and B identified 100% and 99%, respectively, of the abnormal bronchi and 97% and 96%, respectively, of the normal bronchi on CT scans. For the standard CT examination (8-mm-thick sections) a sensitivity of 94% (observer A) or 91% (observer B) and a specificity of 99% (observers A and B) were found in the diagnosis of a normal or narrowed bronchial lumen. CT proved to be a reliable method for demonstrating tumor lesions of the bronchi.
In our hospital in the years 1980 till 1986 37 displacements of the tuberositas tibiae were performed in chondropathia patellae and recurrent luxation of the patella. The follow-up examination took into consideration the radiological and clinical findings, the subjective complaints and the ability of going in for sports. These clinical experiences and pressure measurements in the femoro-patellar joint show the best results after the displacement of the tuberositas tibiae according to Blauth. Additionally we recommend an accurate indication for chondropathia patellae and recurrent luxation of the patella and accompanying surgical measures.
25 patients with an extrathoracic malignant tumor and radiologically proven lung nodules had a thoracic CT preoperatively. 111 nodules were confirmed by surgery (median: 10 mm). Histopathological examination revealed 104 metastases or sterilised metastases and 7 benign nodules. CT detected 97% of the nodules. Localisation of the nodules according to different lobes or segments was correct in 97% of nodules. CT is a very sufficient method for the detection and localisation of lung metastases.
Digital subtraction angiography (DSA) plays an important role in the management of vascular diseases of the lower extremities. A disadvantage is the lack of an automatically moving table top. We used a 1,024 x 1,024 matrix with a large-screen intensifier system and an automated "stepping" facility. In 161 examinations of the arteries of the lower extremity digital peripheral arteriography was performed with and without the subtraction technique. We compared the influence of different iodine concentrations in DA and DSA. Peripheral DA proved to be equal to peripheral DSA in the region of the pelvis, thigh and knee, with no adequate contrasting being obtained merely in the region of the lower leg arteries in about 45%. It is necessary to use contrast medium at a concentration of 300 mg I/ml. The installation of an automated "stepping" facility reduces the amount of contrast medium needed and the exposure time.
The expression of nucleolus organizer region (NOR) activity in diploid cells was investigated in a model mammalian hybrid system, the hinny (female ass x male horse), by sequential Ag-NOR and chromomycin A3/distamycin A/DAPI (CDD) staining ion lectin-stimulated peripheral blood lymphocytes. In the majority of cases we found non-expression of the horse-derived NOR chromosomes in the hinny. However, in one case there was strong NOR expression on horse-derived chromosome no. 1.
66 patients with suspected bronchial tumors were examined by conventional tomography and by CT. The results were evaluated separately by two radiologists (A and B). All diagnoses were confirmed by bronchoscopy or surgery. In evaluating individual bronchi, conventional tomography had a sensitivity of 88% (A) or 87% (B) and a specificity of 97% (A) or 95% (B); CT had a sensitivity of 95% (A and B) and a specificity of 99% (A and B). If CT is available, our experience indicates that conventional tomography is no longer indicated.
Forty patients with anterior dislocation of the shoulder were examined in order to demonstrate Hill-Sachs defects and bony Bankart lesions. All patients had conventional radiographs of the shoulder in two planes, special views according to Hermodsson and Bernageau and CT. The examinations were evaluated by three observers. CT was regarded as the gold standard. Sensitivity for demonstrating Hill-Sachs defects for the standard views was 45% and for the Hermodsson views was 76%. For demonstrating Bankart lesions, standard views had a sensitivity of 37% and the Bernageau view 64%. Computed tomography was markedly superior to the standard and the special views, both for demonstrating the lesions and the agreement between various observers.
