Topography of cones and rods in the tree shrew retina.

The topographical distribution of cones and rods in the tree shrew retina was analysed quantitatively in whole-mounted retinae and horizontal semithin sections stained with cresyl violet or toluidine blue. The outer nuclear layer consists of a single layer of photoreceptor nuclei with the rod nuclei slightly displaced towards the outer plexiform layer. This facilitated quantification of the photoreceptor populations. The density of cones ranges from 12,000/mm2 in the peripheral retina to a maximum of 36,000/mm2 in the inferior retina. Unlike ganglion cell density, the density of cones does not peak in the temporal retina. Rod density, between 500/mm2 and 3,500/mm2, also peaks in the inferior retina, but not in the same region as cone density. Rods constitute from 1 to 14% of the photoreceptor population, depending on retinal location, and have a local minimum at the central area. Amongst the cones a regularly arrayed subpopulation of presumed blue-sensitive cones is distinguished by its special staining properties. These cones constitute between 4 and 10% of the cone population depending on retinal location. A second, irregularly spaced, subpopulation of possibly pathological cones is also described.

Analysis of guinea pig leukocyte antigens using interspecies T cell hybrids.

In the guinea pig system, there are no T lymphocyte tumors available. In addition, in this species the production of T cell lines in vitro for analysis of T cell markers or function proved to be difficult. As an alternative approach, guinea pig X mouse and guinea pig X rat T cell hybridomas were obtained by fusion of guinea pig T cells with HAT-sensitive mouse or rat thymomas. In ten fusions a total of 578 hybrids were produced and kept in culture for periods ranging from weeks up to several months. The highest yield was obtained when the Mb C 12 line was used as tumor parent. BW 5147 and the rat thymoma W/Fu(C58NT)D gave slightly lower fusion efficiencies. The yield depended also on the pretreatment of the parental cells. Activation by MLR proved to be most effective. A number of interspecies hybrids expressed guinea pig T cell differentiation antigens as detected by a cell ELISA. The positive hybrids were recloned several times and exhibited a stable expression of these markers, even after continuous culture for more than 2 months. Western blot analysis was used to confirm antigen expression at the protein level by comparing the hybrids with both parental cells. The hybrids expressed the proteins of the guinea pig lineage. No reaction was observed with the murine or rat thymomas. Therefore, the application of interspecies T cell hybrids provides an alternative for studies of guinea pig T cells. The cells are easy to grow and to clone and may be stored frozen in liquid nitrogen. The hybrids should permit analysis at the clonal level also for functional studies.

[Oxoferin and sodium chlorite--a comparison].

Oxoferin, a preparation approved for wound treatment, has been subjected to an in vitro analysis. 1. Oxoferin produces methaemoglobin even if diluted 500 fold, and leads to additional alterations if added in high concentrations to red cells. Sodium chlorite (NaClO2, 15 mM) is equivalent to undiluted Oxoferin. 2. Oxoferin in fiftyfold dilution damages fibroblasts in cell culture slowly and persistently. This dilution is equieffective with a 200 microM solution of sodium chlorite. Oxoferin and sodium chlorite also damage vascular endothelial cells. 3. The oxidation equivalent of Oxyoferin is 12.3 mM sodium chlorite. Our data indicate that Oxoferin may be equated essentially with aqueous sodium chlorite.

Problems in genetic counseling in a family with an "atypical" centronuclear myopathy.

Genetic counseling often deals with a rare disease the inheritance of which is not clearly established or in which genetic heterogeneity is reported. In addition, relevant parameters such as penetrance, gene frequency, and mutation rate may not be available. In this situation, establishing the risk may be very difficult. An example is presented in which Bayesian risk calculation proved to be of great help in providing precise risk estimates in a family in which an "atypical" centronuclear myopathy was segregating.

Opsin- and S-antigen-like immunoreactions in photoreceptors of the tree shrew retina.

In the tree shrew retina individual rod and cone photoreceptors can be readily identified and quantified because their perikarya are arranged in a single layer. This retina is therefore an ideal system for testing the specificity of photoreceptor-directed antibodies. Here we describe the staining properties of polyclonal antibodies against (rhod)opsin and retinal S-antigen in the tree shrew retina. The (rhod)opsin antibody exclusively and completely labelled the rod population. The antibody against S-antigen also labelled all rods and, in addition, a regularly arrayed subpopulation of cones, which we argue to be the blue-sensitive cones. In the context of our findings, the labelling of pinealocytes with these antibodies is discussed.

Hot spot of recombination within DXS164 in the Duchenne muscular dystrophy gene.

The DMD gene, which spans more than 2,000 kbp, has been assigned to band Xp21 of the X chromosome. Two subclones (PERT 87-1 and PERT 87-15) of the intragenic locus DXS164 physically are separated by approximately 60 kbp. Linkage studies were done in 49 informative DMD families by using the LINKAGE program. Crossing-over between the loci studied occurred in four families. A recombination rate of 4% (support interval [Zmax-1] 1%-10%), which was 54 (support interval 14-135-fold) times higher than expected, was found with a maximum lod score of 13.50. These data suggest a hot spot for recombination within DXS164.

[Percutaneous catheter embolization of the of the internal iliac artery in life-threatening gynecological bleeding].

16 patients with life-endangering hemorrhages caused by gynecologic malignancies have been treated with the percutaneous transcatheter embolisation. In general for embolisation of the internal iliac artery gelfoam was used. Life-endangering bleeding could be stopped in all cases, only 3 mild persistent bleedings and another recurrence of hemorrhage could be controlled conservatively. Major-complications did not occur. The significant advantage of catheter embolisation in controversy to surgical treatment is discussed.

[The iodine status in the puerperium and in newborn infants].

In this paper we report about the actual supply of iodine of women in childbred determining iodine levels in serum and milk of the mothers. No significant differences were found between women without, with treated or with untreated goitre. In early childbred and 4 weeks after birth the iodine in milk amounts to about the half of the maternal serum level and does not further increase with continuation of nursing.

Characterization of pre-rRNA components in ribosomal precursor particles from macronuclei of Tetrahymena thermophila.

Ribosomal precursor particles were extracted from purified macronuclei of Tetrahymena thermophila and separated in sucrose gradients. The RNA components of major particle fractions were isolated and analyzed by Northern blot hybridization using cloned rDNA fragments. A tentative scheme of preribosome maturation was established based on the RNA constituents and corresponding processing steps occurring in the different particle classes. The primary transcript of ribosomal genes, the unspliced precursor rRNA, was found in some experiments in the upper region (less than 40S) of sucrose gradients run for short times. This is in accordance with earlier results by others indicating that slowly sedimenting ribonucleoprotein (RNP) structures may exist as a transitory stage of preribosome formation. Usually, however, unspliced pre-rRNA was only found in 80S preribosomes, where splicing occurred as indicated by the presence of splice intermediates and products only in this fraction. In addition, the further processing of spliced pre-rRNA at three major sites in variable temporal order took place in the 80S preribosomes, i.e., (i) the cleavage at or near the 5'end of the 17S rRNA sequence, (ii) the central cleavage in the internal transcribed spacer (ITS2) between the 5.8S and 26S rRNA sequence, and (iii) the cleavage in the ITS1 at or near the 3' end of the 17S rRNA sequence. Only the latter event was found to result more or less immediately in the division of the 80S preribosomes into separate precursors (p40S and 60S) of the small and large ribosomal subunits. If the alternative pre-rRNA cleavage site in the ITS2 was used first the 80S preribosomes retained their integrity. The conversion of the p40S precursors into nuclear 40S subribosomal particles was correlated with the processing of pre-17S rRNA into 17S rRNA. In the 60S ribosomal precursor particles the processing of pre-26S rRNA, including the formation of precursors (ITS and 7S RNA) to 5.8S rRNA, occurred. A substantial proportion of 26S rRNA molecules isolated from these particles already contained the central hidden break as indicated by the presence of 26S rRNA alpha- and beta-subfragments. The major pre-rRNA processing by-products, IVS and ETS RNA, were partly associated with preribosomes and partly present as free RNAs in the supernatant of sucrose gradients. This indicates that they are liberated and degraded mainly outside the particles in which they are formed. In contrast, the initiation fragment (IF), a small promoter-proximal transcript, was exclusively associated with large particles.(ABSTRACT TRUNCATED AT 400 WORDS)

Ordered activation of the Ig lambda locus in Abelson B cell lines.

Derivatives of the mu-producing Abelson line P8 have been analyzed for L chain gene rearrangements. Two of seven clones studied assembled their V lambda genes while growing in culture. V lambda gene rearrangements occurred only in those Abelson subclones that either were rearranging or had rearranged their recombining sequence (RS) element on both Ig kappa alleles. Our data suggest that (a) RS rearrangements are preferentially initiated in kappa- pre-B cells; and (b) the deletion or inactivation of sequences lying between J kappa and RS is a requirement for the activation of the Ig lambda locus.

Synergistic antiplatelet and antithrombotic effects of a prostacyclin analogue (iloprost) combined with a thromboxane antagonist (sulotroban) in guinea pigs and rats.

The stable PGI2-analogue iloprost and the TXA2-receptor antagonist sulotroban were investigated for possible cooperative effects on platelet function and experimental thrombus formation in guinea pigs and rats. Iloprost and sulotroban inhibit intravascular platelet aggregation in guinea pigs and rats induced by the stable endoperoxide U 46.619 and collagen, with iloprost being the more potent and (for collagen) more efficacious drug. Combinations of both compounds show synergistic or additive effects on in vivo platelet function. Thrombus formation in rats induced by vascular damage is strongly reduced by combining doses of iloprost and sulotroban (BM 13.177) which given alone are ineffective. These results suggest a cooperative enhancement of antiplatelet and antithrombotic effects for combinations of iloprost and sulotroban. In view of disadvantages of currently used platelet inhibitors this cooperativity may offer a new approach in antiplatelet therapy.
