[Studies on anordrin injection for termination of early pregnancy in mice].

In order to increase the effectiveness on the termination of early pregnancy, anordrin injection was prepared. A nonaqueous solvent was selected as the parenteral solvent which was not irritating and toxic. The security test showed the injection was also not irritating and toxic. After the injection was heated for 76 h in 95 +/- 2 degrees C water or stored for 3 mon at 37 degrees C, the content of anordrin showed no change. The activation energy of the injection was 68.1 kJ/mol determined by thermogravimeter analysis. This also showed that the injection was steady. Results of termination of early pregnancy in mice showed that a single dose of anordrin injection (3 mg/kg) intraperitoneally was more effective than a single dose of anordrin tablet (3 mg/kg) orally (P less than 0.01). Histological studies showed that embryo, decidual cells and nutrient cells were markedly degenerated or dead when treated with the injection (3 mg/kg) intraperitoneally.

[Morphological observation of termination of early pregnancy with anordrin in mice].

Twenty four mice were divided randomly into experimental and control groups. There were 10 animals in the control group. In experimental group, anordrin (3 mg/kg) was injected into the peritoneal cavity to terminate early pregnancy. The control group was treated with normal saline. Pathological changes in the decidual cells, blastocysts (including giant cells), corpora lutea and embryo were observed. It was found that the animals in the control group were developing normally. In all the experimental groups decidual cells and blastocysts showed degeneration and necrosis. The granulosa lutein cells also showed degeneration. The dead embryo was embedded in the necrotic decidual cells and blastocysts. The mechanism concerned may involve a combination of various factors. The degeneration of granulosa lutein cells may be the most important of the factors.

[Hypertensive putaminal hemorrhage with intraventricular hemorrhage--50 clinical cases and 6 pathoanatomy cases].

50 cases of hypertensive putaminal hemorrhage (HPH) complicated by intraventricular hemorrhage (IVH) were studied by the vertical position typing method with comparison between findings CT results and patho-anatomy of 6 cases. HPH with IVH were typed into five types: IV type, 2V type, IV+3V type, 2V+3V type, total V type. Research the relation between the three ways intruding verticle and position of HPH. Study the average of survive period in HPH with IVH and prognosis. The prognosis of IV+3V type is the best and 2V type os the worse.

[Study on dissolution rate of injection of microencapsulated compound megestrol acetate].

Dissolution rate of megestrol acetate in the injection of microencapsulated compound megestrol acetate was determined by the first derivative spectrum amplitude method. The experimental results reveal that t50 of the microencapsulated sample (I) is about 60.16 d, while the broken microencapsulated sample (II) is about 15.89 d and the unmicroencapsulated sample (III) about 15.87 d. The difference is regarded as of obvious significance (P less than 0.01). The values of the dissolution rate were linear with the parameters of rabbits and women both in vivo.

[Studies on coprecipitate of anordrin-PVP].

The coprecipitate of anordrin (AD)-PVP was prepared by solvent method. The DSC (differential scanning calorimetry) revealed that AD did not have crystalline structure in coprecipitates of 1:7-1:9. X-ray diffraction spectrum of 1:8 coprecipitate (COPPT) showed no crystalline structure of AD. The dissolution rate of AD was about 38 times higher for 1:8 COPPT than pure AD. Activation energies determined by DTG (derivative thermogravimetry) were 182.8 and 133.4 kJ/mol, respectively, showing that the 1:8 COPPT is much more stable than pure AD in thermal degradation. The experimental results showed that anti-implantation effect of the 1:8 COPPT was much better than that of AD tablets: the number of implantation sites of mice administrated 1:8 COPPT (5 mg/kg) was 0.2 (P less than 0.01) and that of mice administrated AD tablets (10.6 mg/kg) was 0.9 (P less than 0.05); effective dose of 1:8 COPPT was less than half of that of AD tablets.

Study of lung flukes from Philippines: --a preliminary report.

In this paper taxonomic study on paragonimus including their metacercariae obtained from the crabs collected from Sorsogon, Luzon, Philippines and adult worms and ova obtained through animal feeding, with light microscopy and scanning electron microscopy revealed that there may exist two or more species of lung flukes in Philippines. The occurrence of Semisulcospira libertina in addition to Antemelania dactylus (Lea) as an additional snail host is mentioned for the first time. The chromosome Karyotyping of the adult worms revealed that they belong to the diploid pattern. Electronic scanning showed that the cuticular spines of the Paragonimus from Sorsogon look like "pineapple ears" both in distribution and in appearance, a new finding that has not been recorded before. In the lungs of a white rat killed 290 days after infection, two adult worms were obtained within the solitary cyst. These 2 adult worms measured 4.5 x 2.5 mm and 4.5 x 3.0 mm respectively. Their unusually small size and somewhat round shape suggest that they may be of a different species from those hitherto reported in the Philippines. Clearcut evidence is presented for the first time, demonstrating the occurrence of the phenomenon of cross fertilization (allogamy) of lung flukes.

Ineffective diastolic filling in tachycardia-induced cardiomyopathy with total pulsus alternans.

We report a case of total pulse alternans in a patient with paroxysmal ectopic atrial tachycardia and echocardiographic findings obtained before and after radiofrequency catheter ablation (RFCA) that terminated the arrhythmia. The patient was a 27-year-old man with history of paroxysmal palpitations with worsening episodic dizziness, chest tightness, and dyspnea. Electrocardiography (ECG) showed atrial tachycardia at 160 to 170 beats/min while the simultaneous pulse was in the 80s beats/min. Echocardiogram showed that aortic and mitral valves opened with alternating excursions and outflow velocities. Furthermore, despite similar ventricular wall thickening during systole of consecutive cardiac cycles, there was alternating mitral valve opening during diastole of the same cycles, providing direct evidence that ineffective diastolic filling and mitral valve opening may play a role in the pathogenesis of pulse alternans. Repeat ECG and echocardiography after the successful RFCA showed normal sinus rhythm and normal opening excursion and the velocity across the aortic and mitral valves.

Coronary artery stenoses: a phantom study using contrast enhanced three-dimensional electron beam tomography.

This paper evaluated the accuracy of electron beam tomographic angiography (EBA) with conventional coronary arteriography (CCA) using four graded artificial stenoses in a postmortem swine coronary phantom model. The sensitivity, specificity, and accuracy of EBA for diagnosing significant stenosis (> or =50% stenosis) were 94.3%, 96.7%, and 95.8%, respectively. The diagnostic accuracy of EBA had no significant difference with CCA (chi(2)=0.0162; P>.05). EBA three-dimensional (3D) procedures had high interobserver reproducibility (k=.92-.95, P>.05). Maximum intensity projection (MIP) was the most sensitive and curved planar reformation (CPR) was the most accurate 3D procedure for quantitatively identifying coronary stenosis. EBA yields promising results concerning the visualization of coronary artery stenosis with high accuracy for stenoses >50%.

Ciprofloxacin-loaded bovine serum albumin microspheres: preparation and drug-release in vitro.

Ciprofloxacin albumin microspheres were prepared by the spray drying technique, with bovine serum albumin as the natural biodegradable wall material. The spherical microspheres, flowed well, were organic solvent free and in the size range 1-5 microm. The drug release from the microspheres could be retarded by further thermal denaturation. The sustained-release microspheres were suitable for dry powder inhaled lung drug delivery systems.

Long-acting delivery microspheres of levo-norgestrol-poly(3-hydroxybutyrate): their preparation, characterization and contraceptive tests on mice.

The preparative technology for sustained release drug delivery microspheres of levo-norgestrol-poly(3-hydroxybutyrate) was optimized based on the in-liquid-drying method. The formation of the drug microspheres was confirmed with differential thermal analysis. The appearance, particle size and distribution, residual CHCl3, drug content, drug release characteristics in vitro, stability and anticonceptive effect on mice of the microspheres were all examined. The average particle size was 64 microm with over 90% of the microspheres being in the range of 28.7-85.8 microm. The residual CHCl3 was lower than 0.001%. The drug release behaviour in vitro could be described by the Higuchi equation and the drug release t1/2 was prolonged by 1.8 times, compared with the original drug LNG. The microspheres were stable for 3 months and showed significant sustained release and anticonceptive effect in mice, and lower toxicity compared with the original drug.

Nonphospholipid vesicles of Carboplatin for lung targeting.

Carboplatin (CPt) was incorporated into nonphospholipid vesicles (NPLV) by the method of membrane dispersion and its content determined spectrophotometrically. The particle size was between 2.0-10.0 microm with a mean value of 3.26 microm and a span of 0.55; and the drug content was 3.965 mg/ml. Dynamic dialysis was used for in vitro release of CPt and the results indicated that the drug release t(1/2) was increased eight times, compared with the original drug. These parameters had little change after lyophilization. When tested with mice, the body distribution parameters showed obviously lung targeting characteristics. The S-180 lung neoplasm models were established by intravenous cancer cells in mice and the number of pulmonary nodules examined for evaluation of the therapeutic efficacy. Antitumor effect was increased and differed significantly (p <.05) compared with the original drug of the same dose.

Coronary artery motion in electron beam tomography.

PURPOSE: The purpose of this work was to evaluate coronary artery motion characteristics and determine optimal electron beam tomography (EBT) scan time during the cardiac cycle to image the coronary arteries. METHOD: This study evaluated the movement of coronary arteries in 20 EBT cine studies, at rest and during stress, obtained for evaluating coronary artery disease. The proximal, middle, and distal segments of each coronary artery were measured at multiple times during the cardiac cycle. The motion distance (mm) and velocity (mm/s) of each segment of the coronary arteries were then measured to establish the motion that occurs in the x and y axes during different times in the cardiac cycle. RESULTS: Coronary artery velocity ranged from 22.4 to 108.6 mm/s. The least motion (and slowest speed) occurred between 30-50 and 40-60% of the R-R interval at rest and stress, respectively. The right coronary artery moved the greatest in the x and y planes (highest speed and spatial change), followed in decreasing order by the circumflex, left main, and left anterior descending arteries. The phase of the cardiac cycle with the greatest coronary artery motion was between 0 and 20% of the R-R interval. CONCLUSION: Coronary artery motion varies greatly throughout the cardiac cycle. To minimize cardiac motion during tomographic imaging of the coronary arteries, we recommend 40-50% R-R interval as an electrocardiographic trigger time and avoiding the use of image acquisition times of >100 ms.
