DNA-liposome complexes transduction of herpes simplex virus thymidine kinase renders human tongue cancer cell line sensitive to ganciclovir in vitro.

OBJECTIVE: To evaluate, in vitro, the cytotoxicity of the herpes simplex virus thymidine kinase (HSV1-tk) gene and ganciclovir (GCV) treatment, a widely used prodrug/suicide gene therapy, in human tongue cancer cell line Tbp-TL. METHODS: Retrovirus expressing plasmid-liposome complex transduction was used. After treatment with G418 for 3 weeks, the integrated HSV1-tk gene was detected by PCR test with the primers specifically designed for HSV1-tk cDNA; HSV1-tk mRNA was detected with in situ hybridization method. In vitro cytotoxicity effect of HSV-GCV on Tbp-TL was detected with MTT and trypan blue exclusion test. RESULTS: In vitro experiments demonstrated dose- and time-dependent cell death by transduction of the HSV1-tk gene followed by GCV treatment. The IC50 (the concentration required to elicit 50% growth inhibition) shifted from 1000 micrograms/mL in non-transduced Tbp-TL cells to 1.1 micrograms/mL in Tbp-TL/TK cells transduced with HSV1-tk gene, with the therapeutic index of 1000. Treatment with GCV at a dose of 10 micrograms/mL for 5 days led to complete cell death in HSV1-tk transduced tumor cells. When mixed with tk positive cells, the non-transduced cells (both the parent Tbp-TL cells as well as another non-transduced cell, the human muco-epidermoid carcinoma cell line M3SP2) were also killed by a degree associated with the percentage of tk positive cells. Seventy to 90% of the mixed cells were killed with only 10% to 20% tk positive cells involved. CONCLUSION: These results suggest that the HSV-tk/GCV approach to human tongue cancer cell line may be efficacious, with a wide therapeutic range, and that it exerts a bystander effect in vitro.

[Anatomical study and clinical application of a leg flap pedicle-included with cutaneous nerve and its concomitant vessels].

OBJECTIVE: To investigate the blood supply patterns and the clinical liability of a leg flap pedicle-included with cutaneous nerve and its concomitant vessels. METHODS: Fresh cadaver legs with thirty-two in infants and two in adults were anatomically examined after the intravenous injection of the red Chlorinated Poly Vingl Choride (CPVC). Five patients with the soft tissue defects were selected for the treatment with the flap pedicle-included with the cutaneous nerve and its concomitant vessels. RESULTS: Four main cutaneous nerves were found in the leg after they perforated the deep fascia out. They were companioned with their concomitant vessels with different blood-supply pateeerns, which the upper part of the leg was in an axial pattern and the lower part was in a "chain-type anastomosing" pattern. Following the above-mentioned findings, five cases were successfully treated with this led flap. CONCLUSIONS: The leg flap should be designed along the cutaneous nerve and its concomitant vessels. When the flap is applied in the area of blood supply with "chain-type anastomosing" pattern, the deep fascia should also be included in the flap.

Azathioprine and methylprednisolone: prevention of chronic cerebral vasospasm in dogs.

OBJECTIVE: To study the preventive effects of single use of azathioprine and its combination with methylprednisolone on chronic cerebral vasospasm (CVS). METHODS: A canine model of the "double subarachnoid hemorrhage (SAH)" was established to study the angiogram characteristics of chronic CVS. The effects of azathioprine and its combination with methylprednisolone on the prevention of chronic CVS were observed in a randomized study. The content of malondialdehyde (MDA) and pathological changes in the basilar artery (BA) wall were also analyzed. RESULTS: The results were obtained on the 7th day after subcarachnoid hemorrhage. In the azathioprine group, the BA diameter was 87% +/- 26% of the original BA diameter, and the MDA content of BA wall was 0.03 +/- 0.01 nmol/mg tissue. In the azathioprine plus methylprednisolone group, it was 93% +/- 20% (BA diameter) and 0.02 +/- 0.01 nmol/mg tissue (MDA content). In the control group, the BA diameter was 53% +/- 19% and MDA content was 0.11 +/- 0.05 nmol/mg tissue. Pathological examination confirmed the reduction in BA wall damage in the experimental groups. CONCLUSION: Azathioprine has a preventive effect on chronic CVS following SAH. It may suppress the production of free radicals and reduce damage to the BA wall. Combined use of azathioprine with methylprednisolone allows lower dosage and less complication compared with a single agent, and results in a better outcome.

[Experimental study on prefabricating iliac grafts using composite bone].

OBJECTIVE: To investigate the feasibility of prefabricating a specified shape autograft capable of transfer using coral and type I collagen as a carrier for recombinant human bone morphogenetic protein-2 (rhBMP-2). METHODS: In this study, the composite of rhBMP-2, coral and type I collagen was made certain shape to prefabricate vascularized osteomuscular autograft capable of microvascular free tissue transfer and autogenous bone graft with certain shape and titanium implant in it. The composite was implanted in the iliac area in dog with the titanium implant at the same time. After 3 months and 4 and a half months of implantation, the composites were studied with gross measurement, X-ray, and histological examinations. RESULTS: After 3 months, composited bone was turned to bone tissue, and the shape of iliac bone was changed with implant in it, bone interface was seen between new bone and implant. And new bone was matured after 4 and a half months. CONCLUSION: Coral and type I collagen are effective carrier for rhBMP-2 to prefabricate vascular osteomuscular autograft with certain shape. The use of rhBMP-2 for tissue engineered microvascular free bone flaps has an unlimited potential and adds a new dimension to maxillofacial reconstruction.

[Effects of hexamethylene bisacetamide on in vitro adhesion, motility and invasion of human mucoepidermoid carcinoma cell line MEC-1].

OBJECTIVE: To investigate the effects of hexamethylene bisacetamide (HMBA) on the in vitro adhesion, motility and invasion of human mucoepidermoid carcinoma cell line MEC-1. METHODS: MEC-1 cells were induced with 2 mmol/L of HMBA in vitro, the adhesion of MEC-1 cells to the reconstituted basement membrane Matrigel was investigated with MTT colorimetric assay; the motility and invasion of MEC-1 cells were assayed using biopore membrane Millicell-PCF inserts culture system. RESULTS: The adhesion of MEC-1 cells treated with HMBA for 30 min or 90 min reduced by 26.47% and 31.13%, respectively; the motility and invasion of MEC-1 cells treated with HMBA reduced by 63.49% and 67.65%, respectively. CONCLUSIONS: HMBA has anti-invasion effects on human mucoepidermoid carcinoma cells in vitro.

[Correlation of intrauterine growth retardation with endocrine parameters].

OBJECTIVE: To investigate the relationship between intrauterine growth retardation (IUGR)and endocrine parameters and to assess the effects of the main endocrine factors on IUGR. METHODS: The concentrations of growth hormone (GH), insulin, triiodothyronine (T(3)), tetraiodothyronine (T(4)), and thyroid-stimulating hormone (TSH) were measured in maternal serum, umbilical blood and amniotic fluid. The samples were collected from 23 term pregnancies with IUGR, and 42 normal pregnancies served as control. Growth hormone and insulin were measured by radioimmunoassay, and T(3), T(4) and TSH by microradioimmunoassay. RESULTS: The concentrations of GH, insulin and T(4) in umbilical blood were lower in IUGR group than that in the normal group (IUGR: GH 4.63 microg/L vs 7.01 microg/L, insulin 10.68 mIU/L vs 31.44 mIU/L, T(4) 0.68 mg/L vs 1.07 mg/L. P < 0.05, 0.05, 0.05), the TSH level in umbilical blood was higher in IUGR than that in the control group (10.84 mIU/L vs 5.74 mIU/L. P < 0.01). The concentration of GH in maternal serum and insulin in amniotic fluid in IUGR group were also lower than that in normal group (GH 1.77 microg/L vs 2.74 microg/L, P < 0.01; insulin 5.84 mIU/L vs 15.64 mIU/L, P < 0.01). CONCLUSIONS: This study confirms previous observation that insufficiency of GH might be a pathogenesis of IUGR. The decrease of GH and insulin might compromise the basic metabolism of the fetus. Besides, the change of early hypothyrosis of fetus with IUGR might benefit them to live in unfavorable environment.

[Craniovertebral decompression and posterior fossa reconstruction treatment of Chiari syringomyelia complex].

OBJECTIVE: To study the results of Chiari malformation/syringomyelia (CM-SM) complex treated by craniovertebral decompression and posterior fossa reconstruction (PFR). METHODS: 37 patients of CM-SM complex were treated surgically from 1994 to 1999. All patients underwent craniovertebral decompression and posterior fossa reconstruction. The procedure consisted of suboccipital craniectomy and laminectomy of C1 (when necessary C2), exploration and plugging of the obex, and resection of arachnoid adherence. A wide dural graft was used to reconstruct the cisterna magna artificially. Syrinx shunt was not performed. RESULTS: Follow-up for 0.5 to 4.5 years showed that 31 patients (83.8%) had their symptoms improved, 5 (13.5%) stabilized, but 1 (2.7%) deteriorated. Magnetic resonance was used to evaluate the morphological results. The shrinkage of the syrinx, upward migration of the hindbrain, the formation of an artificial cisterna magna were observed. CONCLUSIONS: Craniovertebral decompression and posterior fossa reconstruction in restoring the craniovertebral junction are recommended in the treatment of CM-SM complex.

[Relationship between tumor suppressor gene p16 and Rb and early diagnosis of lung cancers].

OBJECTIVES: To study the incidences and mechanisms of tumor suppressor gene p16 and Rb inactivation in lung cancers, and to investigate their relation to biological characters, clinical pathological diagnosis and gene classification diagnosis of lung cancers. METHODS: Immunohistochemistry, double in situ hybridization, PCR, PCR-SSCP and sequencing were applied to study the changes of p16 and Rb genes in 106 lung cancers and 23 lung tissues of benign diseases. RESULTS: Total expression rates of p16 and Rb proteins and mRNAs in lung cancers were lower than those in normal lung tissues and benign lung diseases, and they were specially related to histological type, metastasis and clinical stage of lung cancers. In stage I and II lung cancers, the obvious inactivation of tumor suppressor gene p16 or Rb was examined (32.6% or 28.3%); p16 inactivation was detected mainly in non-small cell lung cancers, and Rb inactivation mainly in small cell lung cancers. There were three mechanisms of homozygous deletions, methylations and mutations for p16 gene inactivation. The deletion rate of p16 exon1 and/or exon2 was 25.8%, mainly took place in p16 protein negative cases of non-small cell lung cancers. 15 cases (16.9%) took part in methylations on SmaI sites of CpG island of p16 gene. PCR-SSCP and sequencing showed that 9 cases had p16 gene mutations. CONCLUSIONS: p16 and Rb genes may play important roles in genesis and progression of lung cancers. Inactivation of p16 or Rb gene may be is an early link of lung canceration, that is very important for early diagnosis of lung cancers. A new gene classification model for lung cancer diagnosis would be set up based on the research of p16 and Rb genes.

Relations of tumor suppressor gene p16 and RB to early diagnosis of lung cancer.

OBJECTIVES: To study the incidence and mechanism of the tumor suppressor gene p16 and RB inactivation in lung cancer, and to investigate their relations to biological characteristics, clinical pathological diagnosis and gene classification. METHODS: Immunohistochemistry, double in situ hybridization, PCR, PCR-SSCP, and sequencing were used to study the changes in the p16 and RB genes in 106 patients with lung cancer. The specimen of normal lung tissue from the patients with lung cancer and specimens of the 23 patients with benign lung diseases were studied in contrast to those of the same patients. RESULTS: The total expression rate of the p16 and RB genes in the lung cancer tissues of the 106 patients was lower than that in normal lung tissues and tissues of benign lung diseases and was specially related to histological type, metastasis, and clinical stage of lung cancer. In stage I and II lung cancer, the obvious inactivation rate of the tumor suppressor gene p16 or RB was 32.6% or 28.3%. The p16 inactivation appeared largely in non-small cell lung cancer (50.1%), and the RB inactivation largely in small cell lung cancer (88.2%). Homozygous deletion, methylation, and mutation were three mechanisms for the p16 gene inactivation. The deletion rate of the p16 exon 1 and/or exon 2 was 25.8% in the patients with non-small cell lung cancer. Fifteen patients (16.9%) showed methylation at the SmaI sites of CpG island of the p16 gene. PCR-SSCP and sequencing revealed p16 gene mutation in 9 of the 106 patients with lung cancer. CONCLUSIONS: The p16 and RB genes may play an important role in genesis and progression of lung cancer. The inactivation of the p16 or RB gene may be an early sign of lung canceration, that is important for the early diagnosis. A new gene classification model for lung cancer diagnosis would be set up based on the research of the p16 and RB genes.

[Photoreceptor apoptosis and p53 gene expression in inherited retinal degeneration of RSC rats].

OBJECTIVE: To investigate the gene regulation of photoreceptor apoptosis in inherited retinal degeneration. METHOD: The retinas of inherited retinal degeneration in animal models of RCS (Royal College of Surgeons) rats, ranging from 13(th) to 60(th) postnatal days in age, were studied by TdT-mediated biotin-dUTP nickend labeling (TUNEL) for apoptosis, immunohistochemistry for p53 protein, in situ hybridization and reverse transcription-polymerase chain reaction (RT-PCR) for p53 mRNA. RESULTS: The photoreceptor of RCS rats underwent apoptosis from 25(th) postnatal day, and reached the highest level at 30 - 35(th) postnatal days. At the early stage of photoreceptor apoptosis, the level of p53 protein and mRNA in photoreceptors increased significantly. RT-PCR also showed that there was more production of p53 mRNA at 25 to 35(th) postnatal days in the retinas of RCS rats. CONCLUSION: P53 gene expression increases significantly at the early stage of photoreceptor apoptosis, indicating that p53 gene might play a role in the regulation of photoreceptor apoptosis in inherited retinal degeneration.

[Umbilical cord blood stem cells transplantation from an unrelated donor into an adult with severe aplastic anemia].

OBJECTIVE: To evaluate the availability of the treatment of adult severe aplastic anemia with unrelated allo-cord blood stem cell transplantation (CBSCT). METHODS: HLA-matched unrelated cord blood transplantation has been successfully performed for an adult severe aplastic anemia patient. A unit of cord blood provided by Guangzhou Cord Blood Bank containing 1.89 x 10(7)/kg mononucleated cells, 1.8 x 10(4)/kg CFU-GM and of CD(34) positive cells was 0.009. The patient was conditioned with CTX (60 mg/kg) and anti-lymphocyte globulin (ALG, 120 mg/kg). GVHD prophylaxis consisted of both MTX and CsA. The CsA had been given for 8 months. RESULTS: The lowest ANC was 0.6 x 10(9)/L post-transplantation. The patient achieved an ANC of greater than 0.5 x 10(9)/L at 10 days, and the platelet of greater than 50.0 x 10(9)/L at day 20 after transplantation. He developed Grade 1 GVHD in the tenth month after grafting. Microsatellite DNA fingerprinting indicated a stable donor-recipient mixed chimerism, whilst the circulating red cells remained host origin. CONCLUSION: It is the first report in China on successful treatment of adult severe aplastic anemia by unrelated allo-CBSCT.

[Effect of oxidatively modified high density lipoprotein on transcriptional expression of proto c-fos and PDGF receptor genes in cultured human arterial smooth muscle cell].

The proliferation of arterial smooth muscle cell (SMC) plays an important role in the process of atherosclero-genesis. Using the establishment of primary culture and sub-culture method for human arterial SMC, we observed the effects of native HDL (N-HDL) and oxidized HDL of proto c-fos and PDGF receptor genes transcriptional expression in cultured SMC. The results showed that N-HDL had no effect on the expression of proto c-fos and PDGF receptor genes in the cultured human SMC, that OX-HDL had no effect on the expression of PDGF receptor gene in the cultured human SMC, and that OX-HDL promoted the expression of protoc-fos gene significantly (P < 0.01). These results suggest that the OX-HDL's atherogenic role is closely related to its stimulating effect on proto c-fos gene expression of SMC.

[A modified assay for phospholipids in blood serum].

An antiscorbutate reduction method for serum phospholipids assay was modified by decreasing the consumed quantity of sulphuric acid and perchloric acid to 30% of that before modification, and the quantity of serum to 40% of that before modification. The digesting time was only 50%-62% of that before modification. The coefficients of variation (CV) were 3.4% and 3.7% for two batches of assays. Recovery of phosphotidylcholine added to serum ranged form 94% to 109%, averagely 100.8%. Serum phospholipids assay of 50 normal adults by the methods before and after modification showed statistically no significant difference (2.813 +/- 0.333 mmol/L vs 2.828 +/- 0.316 mmol/L, P > 0.05), but good correlation (r = 0.991). The results indicate that the modified assay for serum phospholipids can decrease the use of serum and reagents, especially the harmful acids, and get the same result as that before modification.

[Study on vitamin E succinate inducing apoptosis of human gastric carcinoma cell].

The effects of vitamin E succinate (VES) on the growth of human gastric carcinoma SGC-7901 cell were observed in cell culture. The apoptosis of SGC-7901 cell induced by VES was studied by fluorescent staining and flow-cytometry. The results showed that VES (5, 10 and 20 mg/L) could obviously inhibit the growth of gastric carcinoma cell, and it could be the result of inducing apoptosis.

[The experimental study on the activity of rhBMP-2, coral and collagen composites inducing intramuscle bone].

OBJECTIVE: To investigate the ectopic bone induction activity of the composite of coral, collagen and rhBMP-2, and the degradation of the carriers of rhBMP-2. METHODS: The composites of rhBMP-2, coral and collagen were implanted into the muscle pouches of mice. The mice were divided into the following four groups. Group H: each composite had 2 mg rhBMP-2, Group L: each composite had 0.5 mg rhBMP-2, Group J: each composite had only type I collagen, Group C: only coral was implanted in the muscle of mouse. Each group had 20 mice. Every 5 mice in each group were killed postoperatively after 1, 2, 4 and 8 weeks. The ectopic osteoinductive activity was assessed with histology and histomorphometry. The point counting method was applied to measure the formation of new bone and cartilage and the degradation of coral in the composites. The data was analyzed statistically using t test. RESULTS: There was no obvious morbidity in any of the animals in this study, and no wound infection, bleeding, or feeding difficulties postoperatively. The samples were examined histologically. Cartilage and bone induction occurred in Group H and L. After 1 Week, new bone was observed in Group H, and cartilage formation occurred mainly in Group L. After 2 weeks, both cancellous bone and bone marrow elements were present in Group H and L. New bone formation was not uniform throughout the specimen. Part of the coral in the composites was not absorbed between 2 to 4 weeks postoperatively. At the 8th week, the coral in the composite was absorbed completely. While in Group J and C there was not any new cartilage or bone formation. According to the statistical analysis, bone formation occurred 2 weeks earlier in Group H than in Group L after operation. From 4 postoperative weeks, no difference of the bone formation rate was observed between Group H and Group L. The degradation rate of coral in Group C was the fastest among the four groups. CONCLUSION: The composites of rhBMP-2, coral and collagen are degradable bone substitutes with bone induction and conduction, and induce dosedependent and time-dependent amounts of intramuscle bone in mice during limited postoperative period. The coral in the composites could be absorbed completely.

[Treatment of chronic hypertrophic rhinitis by Nd:YAG laser under nasal endoscope].

OBJECTIVE: To evaluate the curative effect of the treatment of chronic hypertrophic rhinitis with Nd:YAG laser under nasal endoscope. METHOD: Fifty-seven patients with chronic hypertrophic rhinitis were treated with Nd:YAG laser operation under nasal endoscope at our outpatient clinic. Some factors affecting operation effect were discussed. RESULT: In all patients, 10 cases (17.5%) were cured, 16 cases (28.1%) significantly improved, 28 cases (49.1%) improved, 3 cases (5.3%) unimproved and the total effective rate was 94.7%. No complication happened. CONCLUSION: Compared with the partial middle or inferior turbinectomy and YAG laser surgery without nasal endoscope. This technique has several advantages such as less damage, less hemorrhage, clear and clean operation field, easy to operate and so on.

[Preparative GC separation and identification of methyl 2-methyl-3-furyl disulfide flavour].

On 4% FFAP + 1% H3PO4 Chromosorb W/AW-DMCS (80 mesh-100 mesh)column, the optimized separation of methyl 2-methyl-3-furyl disulfide flavour was obtained. By using automatic preparative attachment model App-5 to GC-9A, the content of methyl 2-methyl-3-furyl disulfide flavour was raised from about 77% to 99.5%. The product was identified by IR, MS and 1HNMR.

[Production, partial purification and characterization of methamidophos-degrading enzyme from methylotroph WB-1].

After cultivation for 20 h in the inorganic salt medium with methamidophos as sole carbon and nitrogen source, Methylotroph WB-1 could produce methamidophos-degrading enzyme in larger amounts. The enzyme was partially purified by sonication disruption, Tween-20 extraction, heat treatment(9 min at 60 degrees C), DEAE-cellulose 32 and CM-cellulose32 chromatography with 22.8 times purification and 78.8% recovery. Activity staining showed single violate band corresponding to that of protein staining. Optimum pH of the enzyme was 9.0; poor substrate specificity was showed. It was strongly inhibited by Hg2+, Mn2+ and Cu2+, but not by Co2+ and Zn2+; it also exhibited poor stability.
