A new insight into the role of "old" chemotactic peptide receptors FPR and FPRL1: down-regulation of chemokine receptors CCR5 and CXCR4.

N-formyl peptides, such as fMet-Leu-Phe (fMLP), are some of the first identified and most potent chemoattractants for phagocytic leukocytes. In addition to the bacterial peptide fMLP and the putative endogenously produced formylated peptides, we recently identified a number of other novel peptide agonists that selectively activate the prototype formyl peptide receptor (FPR) and/or its variant FPRL1. These agonists include several synthetic peptide domains derived from the envelope proteins of the human immunodeficiency virus type 1 (HIV-1) and intact human acute phase serum protein serum amyloid A. The activation of FPR and/or FPRL1 in monocytes by these agonists resulted in increased cell migration, calcium mobilisation and the heterologous down-regulation of the expression and function of chemokine receptors, notably CCR5 and CXCR4, two crucial fusion co-receptors for HIV-1. This down-regulation of CCR5 by FPR and FPRL1 agonists was associated with a rapid serine phosphorylation of CCR5. The desensitisation of CCR5 by FPR or FPRL1 agonists, not only inhibited its biological function induced by chemokine ligands, but also interfered with its capacity to act as a fusion co-receptor for monocyte tropic HIV-1. Thus, heterologous desensitisation by FPR and FPRL1 may play an important role in orchestrating the host innate immune responses which generate multiple chemotactic stimulants. Furthermore, the understanding of the structural and biochemical basis of FPR/FPRL1 activation may lead to the development of novel immunoregulatory and anti-HIV agents that emulate the process of heterologous desensitisation.

Tumor tissue recycling--a new combination treatment for solid tumors: experimental and preliminary clinical research.

Although H&H combination treatment (high active TIL and high sensitive drugs) as previously described to solid tumor patients is more efficient than single-agent treatments such as TIL adoptive immunotherapy, it has a short-term efficiency for the immune response to metastatic cancers. Currently, a new version of the combination of active and adoptive immune response was established. TILs, tumor vaccines and high sensitive drugs. The experimental results demonstrated that TILs from 49 cases (65%) were more than 1,000 expansion-fold and only 6 cases (24%) less than 500 fold. The peaks of TIL 3H-TdR cytotoxicity test from 35 of 64 TIL specimens were kept from 40 to 56 days. TIL phenotypes studied here indicated CD3 80 +/- 21%, CD4 37 +/- 21%, CD8 44 +/- 18% and HLA DR 69 +/- 24% after IL2 induction, in contrast, to CD3 20 +/- 12%, CD4 10 +/- 7%, CD8 11 +/- 3% and HLA DR 30 +/- 16% before induction. Thirty two of 75 cases were assayed using the chemosensitivity test. The distribution of positive rates for the chemosensitivity test were slightly different in different tumors regarding tissues in liver, lung, ovary, breast, and melanoma, 2 cases with melanoma all showed negative results. The results of a tumor vaccine using TNF-alpha gene transduction demonstrated that the expression of HLA Class I and HLA Class II were dramatically increased 87% and 43%. After implanting tumor cells via transduced TNF-alpha retroviral vector into 6-12 week old BALB/c nude mice, only one subject of nine nude mice had a tumor weight of 2.67 g, but the control group all displayed tumors with a weight of 3.24 +/- 0.56 g. Preliminarily clinical trials also showed that the new version was obviously a promising combination.

Cardiomyocyte aging is gender-dependent: the local IGF-1-IGF-1R system.

To determine whether insulin-like growth factor 1 (IGF-1) and its receptor (IGF-1R) are implicated in the aging process of the heart, and if their impact differs in the two genders, the expression of IGF-1, and extracellular alpha-subunit and transmembrane beta-subunit of IGF-1R was measured in left ventricular myocytes isolated from male and female Fischer 344 rats at 3, 8, 12, 16, and 26 months after birth. Additionally, the extent of myocardial damage in both sexes was evaluated in rats at 3 and 26 months by confocal microscopy. Finally, ventricular hemodynamics was assessed in the closed-chest preparation. From 3 to 26 months, aging was characterized by an 83%, 84% decrease and disappearance in the quantity of IGF-1, IGF-1Ralpha and IGF-1Rbeta in male myocytes. Corresponding changes in female myocytes were 40%, 28% and 43%. These molecular modifications at the myocyte level were coupled with tissue injury, consisting of multiple foci of replacement fibrosis across the left ventricular wall. However, myocardial fibrosis in females was 76% and 77% significantly less than in the young and old male heart, respectively. These multiple age-associated events were accompanied by cardiac decompensation in the senescent male rat, while modest indices of ventricular dysfunction were detected in old female rats. In conclusion, the enhanced IGF-1-IGF-1R system in female myocytes may condition the favorable outcome of age in this gender.

Characterization of p53 gene mutations in colorectal carcinomas from an Asian population.

A series of colorectal carcinomas from an Asian population in Singapore were analyzed for mutations in the tumor suppressor gene p53. Based on single-strand conformation polymorphism (PCR-SSCP) analysis and direct DNA sequencing, 15 of 38 tumors (39%) were found to contain mutations in exons 5-8 of the p53 gene. The point mutations were predominantly base transitions. Among the 10 transitions, 5 were at CpG dinucleotide sites. One-third (5/15) of the mutations were found at previously identified hotspot codons 175, 248 and 282. In one case, an insertion of a 6-base pair sequence was found. p53 mutations did not correlate with tumor histological grade, Dukes' stage, or metastases. However, tumors at the distal site showed a higher proportion of mutations than the proximal site. Further, no mutation was found in the normal mucosa adjacent to tumor site, suggesting that no germ-line mutations were present. The results were compared with those from other studies and are discussed in connection to possible etiological factors that are specific to the local population.

Effects of vitamin/mineral supplementation on the proliferation of esophageal squamous epithelium in Linxian, China.

Abnormalities of epithelial proliferation have been proposed as an early step in gastrointestinal carcinogenesis. To determine whether micronutrient supplementation may reduce squamous epithelial proliferation in the esophagus, we evaluated proliferation in subjects participating in a randomized nutrition intervention trial in Linxian, China, where esophageal cancer rates are among the highest in the world. After 30 months of intervention involving daily supplementation with multiple vitamins and minerals, an endoscopic survey was performed and squamous biopsies from 512 subjects were labeled with tritiated thymidine and autoradiographed. Analysis showed no treatment effect on the overall amount of squamous epithelial proliferation measured by the total labeling index. However, a measure of the vertical distribution of labeled cells showed lower values with supplementation: a 14% reduction in all subjects (P = 0.29), and a 29% reduction in nonsmokers (P = 0.03). These results suggest a potential modest benefit for short-term intervention with multiple vitamins and minerals on squamous epithelial cell proliferation of the esophagus in this high-risk population.

Urinary excretion of nitrosamino acids and nitrate by inhabitants of high- and low-risk areas for nasopharyngeal carcinoma in southern China.

The hypothesis that endogenous synthesis of nitrosamines from dietary precursors is a risk factor for nasopharyngeal carcinoma (NPC) in China was tested by applying the nitrosoproline (NPRO) test to subjects living in high- and low-risk districts for NPC in Zangwu county, Guangxi region, in southern China. Samples of 12-h urine were collected from 77 subjects: (a) before any treatment; (b) after ingestion of proline; and (c) after ingestion of proline together with vitamin C. NPRO, other nitrosamino acids, and nitrate were measured as indices of exposure to preformed and endogenously formed nitrosamines or their precursors. The NPRO level after proline intake was significantly increased in subjects from the high-risk area (P = 0.012) and markedly reduced after ingestion of ascorbic acid (P = 0.007), but such an effect was not seen in subjects from the low-risk area. Levels of N-nitrosothiazolidine-4-carboxylic acid and the sum of nitrosamino acids in subjects in the high-risk area were significantly reduced by ascorbic acid (P < 0.01) but were not reduced in subjects from the low-risk area. The urinary nitrate level was about twice as high in subjects from the high-risk area. In subjects from high- and low-risk areas combined, NPRO levels in any of the three dose groups were highly correlated with nitrate levels (P = 0.0001). These results demonstrate a higher potential for endogenous nitrosation in subjects living in the high-risk area of NPC and suggest the occurrence of nitrosation inhibitors in the diet consumed in the low-risk area.(ABSTRACT TRUNCATED AT 250 WORDS)

Inhibition of shear-induced platelet aggregation by Chinese herbal medicines.

Shear-induced platelet aggregation (SIPA) plays an important role in vascular diseases. In this study, SIPA was generated by a computerized cone-plate apparatus. Six compounds from Chinese herbal medicine were selected for in vitro and in vivo tests. Rabbit and rat were involved in the tests accordingly. Ligustrazine hydrochloridi appeared to be most effective for inhibition of SIPA. The inhibition rate of SIPA reached 44.2% (p < 0.01) for an in vitro concentration of 1 mg/ml, and 46.1% (p < 0.01) for an in vivo dose of 50 mg/kg.
