Which way for drug-mediated immunosuppression?

The basic immunosuppressive protocol involves the use of multiple drugs, each directed at a discrete site in the T-cell activation cascade. The prevailing paradigm regarding the mechanisms of action of immunosuppressants is that they function to prevent allograft rejection by preventing cell activation, proliferation and/or cytokine production. A new hypothesis is that some of the immunosuppressants might function by stimulating the expression of immunosuppressive molecules and/or cells.

Human 17 beta-hydroxysteroid dehydrogenase: overproduction using a baculovirus expression system and characterization.

Estrogenic 17 beta-hydroxysteroid dehydrogenase (17 beta-HSD) plays a pivotal role in the synthesis of estrogens. We overproduced human placental estrogenic 17 beta-HSD using a baculovirus expression system for the study of the enzyme mechanism. A cDNA encoding the entire open reading frame of human 17 beta-HSD was inserted into the genome of Autographa californica nuclear polyhedrosis virus and expressed in Spodoptera frugiperda (Sf9) insect cells. Metabolic labeling and Western blot analysis using polyclonal antibodies raised against native human 17 beta-HSD indicated that a molecule with an apparent mass of 35 kDa was maximally expressed 60 h after infection. At that time interval, intracellular 17 beta-HSD activity reached 0.26 U/mg of protein in crude homogenate, about 70 times the level measured in human placenta. Purification of recombinant 17 beta-HSD was achieved by a single affinity fast liquid protein chromatography step yielding 24 mg of purified 17 beta-HSD protein per liter of suspension culture, with a specific activity of about 8 mumol/min/mg of protein for conversion of estradiol into estrone, at pH 9.2. In addition, the recombinant protein purified from infected Sf9 cells was assembled as a dimer with molecular mass and specific activity identical to those of the enzyme purified directly from placenta. The present data show that the baculovirus expression system can provide active 17 beta-HSD that is functionally identical to its natural counter-part and easy to purify in quantities suitable for its physico-chemical studies.

The influence of salt on the structure and energetics of supercoiled DNA.

We present a detailed computational study of the influence of salt on the configurations, energies, and dynamics of supercoiled DNA. A potential function that includes both elastic and electrostatic energy components is employed. Specifically, the electrostatic term, with salt-dependent coefficients, is modeled after Stigter's pioneering work on the effective diameter of DNA as a function of salt concentration. Because an effective charge per unit length is used, the electrostatic formulation does not require explicit modeling of phosphates and can be used to study long DNAs at any desired resolution of charge. With explicit consideration of the electrostatic energy, an elastic bending constant corresponding to the nonelectrostatic part of the bending contribution to the persistence length is used. We show, for a series of salt concentrations ranging from 0.005 to 1.0 M sodium, how configurations and energies of supercoiled DNA (1000 and 3000 base pairs) change dramatically with the simulated salt environment. At high salt, the DNA adopts highly compact and bent interwound states, with the bending energy dominating over the other components, and the electrostatic energy playing a minor role in comparison to the bending and twisting terms. At low salt, the DNA supercoils are much more open and loosely interwound, and the electrostatic components are dominant. Over the range of three decades of salt examined, the electrostatic energy changes by a factor of 10. The buckling transition between the circle and figure-8 is highly sensitive to salt concentration: this transition is delayed as salt concentration decreases, with a particularly sharp increase below 0.1 M. For example, for a bending-to-twisting force constant ratio of A/C = 1.5, the linking number difference (delta LK) corresponding to equal energies for the circle and figure-8 increases from 2.1 to 3.25 as salt decreases from 1.0 to 0.005 M. We also present in detail a family of three-lobed supercoiled DNA configurations that are predicted by elasticity theory to be stable at low delta Lk. To our knowledge, such three-dimensional structures have not been previously presented in connection with DNA supercoiling. These branched forms have a higher bending energy than the corresponding interwound configurations at the same delta Lk but, especially at low salt, this bending energy difference is relatively small in comparison with the total energy, which is dominated by the electrostatic contributions. Significantly, the electrostatic energies of the three-lobed and (straight) interwound forms are comparable at each salt environment. We show how the three-lobed configurations change slowly with ALk, resulting in branched interwound forms at higher salt. In longer chains, the branched forms are highly interwound, with bent arms. At low salt, the branched supercoils are asymmetric, with a longer interwound stem and two shorter arms. From molecular dynamics simulations we observe differences in the motions of the DNA as a function of salt. At high salt, the supercoiled chain is quite compact but fairly rigid, whereas at low salt the DNA is loosely coiled but more dynamic. Especially notable at low salt are the large-scale opening and closing of the chain as a whole and the rapid "slithering"of individual residues past one another. Toroidal forms are not detected under these conditions. However, the overall features of the open, loose supercoils found at low salt are more similar to those of toroidal than interwound configurations. Indeed,simulated x-ray scattering profiles reveal the same trends observed experimentally and are consistent with a change from closed to open forms as salt is decreased. Like the minimization studies, the dynamics reveal a critical point near 0.1 M associated with the collapse of loose to tight supercoils. Near this physiological concentration, enhanced flexibility of the DNA is noted. The collective observations suggest a potential regulatory role for salt on supercoiled DNA function, not only for closed circular DNA,but also for linear DNA with small looped regions.

A nested case-control study of oesophageal and stomach cancers in the Linxian nutrition intervention trial.

BACKGROUND: Rates of oesophageal/gastric cardia cancer in Linxian, a rural county in north central China, are among the world's highest, but the risk factors are not well understood. METHODS: A nested case-control study of oesophageal and stomach cancers was conducted within a cohort of 29,584 adults who participated in a randomized nutrition intervention trial. Information on participant characteristics collected during interviews before the trial began was compared between individuals who subsequently developed cancers of the oesophagus (N = 640) or stomach (N = 539), mainly cardia, and individually matched controls (control/case ratio = 5). Analyses were performed separately for oesophageal and stomach cancers using conditional logistic regression. RESULTS: For oesophageal cancer, tobacco smoking was associated with a significantly elevated risk, with a twofold increase among long-term smokers. Alcohol consumption was uncommon and not related to risk. High consumption of eggs or fresh vegetables was associated with 20% reductions in risk, and risk significantly declined as pre-trial body mass index (BMI), an indicator of long-term nutritional status, increased. No increases in risk were associated with intake of pickled vegetables or mouldy foods, although consumption levels at the start of the trial were low. Excess risks of 40-80% were found among individuals who had reported a history of cancer, notably of the oesophagus and stomach, in parents or sibs. For stomach cancer, only low BMI was significantly associated with elevated risk. CONCLUSIONS: This study indicates that several risk factors for oesophageal and stomach cancers in Linxian, including smoking, nutritional deficiency, and familial cancer occurrence, resemble those in other areas of the world and contribute partly to the remarkably elevated rates in this area of China.

Amino acid sequence similarity of the VP7 protein of human rotavirus HCR3 to that of canine and feline rotaviruses.

The sequence of the VP7 gene of human rotavirus strain HCR3 was determined and its predicted amino acid sequence was compared with that of other rotavirus strains. The VP7 gene is 1062 nucleotides long and contains a single open reading frame of 981 nucleotides capable of encoding a protein of 326 amino acids. The VP7 amino acid sequence similarity of strain HCR3 to those of various human and animal G3 serotypes ranged from 88.7 to 99.4%, and from 60.4 to 88.3% to strains representing each of the other 13 G serotypes. Alignment of four variable regions [VR4, VR5(A), VR7(B) and VR8(C)] of HCR3 with those of G3 strains of different host species showed that HCR3 possesses a sequence almost identical to that of canine rotaviruses and feline rotavirus strain CAT97 in all four regions. A considerable divergence in regions VR4, VR7(B) and VR8(C) was found with strains of human, mouse, monkey, horse and rabbit rotaviruses. This observation together with results of our previous study on VP4 indicated that human rotavirus HCR3 is genetically more closely related to animal rotaviruses than to other human rotaviruses.

Induction of IL-1 beta, IL-6, TNF-alpha, GM-CSF and G-CSF in human macrophages by smooth transparent and smooth opaque colonial variants of Mycobacterium avium.

Both smooth transparent (SmT) and smooth domed-opaque (SmD) colonial variants were obtained from a strain of Mycobacterium avium isolated from a patient with AIDS. The two variants showed similar biochemical characteristics but SmT bacteria proliferated better than SmD bacteria inside human macrophages and were much less capable than the SmD variant of inducing the release of IL-1 beta, IL-6, TNF-alpha, GM-CSF and G-CSF, after incubation for either 3 or 6 days. As cytokines are important extracellular signals for immune cells, the lack of induction observed in SmT-infected macrophages may be one of the pathogenic mechanisms of M. avium.

Association of rotating shiftwork with preterm births and low birth weight among never smoking women textile workers in China.

1035 married women workers in three modern textile mills in Anhui, China were surveyed to investigate the association of rotating shiftwork with low birth weight and preterm birth in 1992. Information on reproductive health, occupational exposure history, and other covariates including age at pregnancy, time and duration of leave from job since pregnancy, and mill location was obtained by trained nurses with a standardised questionnaire. This analysis was limited to 845 women (887 live births), who were middle or high school graduates, never smokers, and non-alcohol drinkers. About 72% of the women worked an eight day cycle with shift changes every two days throughout pregnancy. Mean gestational age was 38.8 and 39.0 weeks for shift and regular schedule workers, respectively. Multiple linear regression was used to adjust for confounding factors including maternal age at pregnancy, order of live birth, mill location, job title, occupational exposure to dust/gases/fumes, stress, carrying and lifting of heavy loads, working in a squat position, time and duration of leave from the job since pregnancy, and indoor coal combustion for heating. The adjusted difference in gestational age associated with rotating shifts was statistically significant (beta = -0.44 (SE 0.20) weeks.) Mean birth weights were 3248 g and 3338 g for rotating shift workers and regular schedule workers respectively. The estimated effect of rotating shiftwork on birth weight was -79 (SE 42) g. When the analysis was restricted to first order live births or to production workers, the estimated effects of rotating shiftwork on both gestational age and birth weight were significant. The proportions of preterm birth (<37 weeks) and low birth weight (<2500 g), respectively, were 20% and 9% for shift workers and 15% and 6% for regular schedule workers. The adjusted odds ratio of shiftwork was 2.0 (95% CI) 1.1-3.4) for preterm birth and 2.1 (95% CI 1.1-4.1) for low birth weight. This association remained significant when the analysis was restricted to production workers or first order live births.

Molecular cloning and expression of Epstein-Barr virus antigens in the lambda gt11 expression vector: antibodies towards proteins from the BORF2 and BKRF4 reading frames in nasopharyngeal carcinoma patients.

An easy way to clone and screen for Epstein-Barr virus antigens significant in the diagnosis of nasopharyngeal carcinoma (NPC) has been developed. Two proteins cloned and expressed as fusion proteins in lambda gt11 have been identified to be expressed from 661 bp of the BORF2 and from 500 bp of the BNKRF4 reading frames. Western blotting studies on these proteins using serum from 16 NPC and 16 normal healthy individuals showed that 15 NPC patients have either IgG or IgA antibodies towards either protein whereas only 2 normal individuals were positive. Hence, IgG and IgA antibodies towards these antigens are of diagnostic value for NPC.

Gender difference in smoking effects on adult pulmonary function.

Data on 1,618 male and 1,669 female adults aged 40-69 yrs, from China in the Beijing Respiratory Health Study, were analysed to investigate the gender differences in effects of smoking on pulmonary function. Smoking was characterized by total smoking-years, smoking status (former, transitional and constant), smoking type (cigarette, cigar and others). The effects of smoking on height-standardized forced vital capacity (FVC) and forced expiratory volume in one second (FEV1) were assessed by multiple regressions adjusting for age, education level, use of an indoor coal stove for heating, passive smoking, occupational dust and gas/fume exposure, and residence. Prediction equations were derived from nonsmoking asymptomatic subjects. As compared to women, men had a much higher smoking prevalence (78 vs 35%) but a lower quitting rate (14 vs 23%). Female lifetime nonsmokers had greater mean percentage predicted lung function values than male lifetime nonsmokers, whilst female cigarette smokers had lower values than their male counterparts. In both sexes, the highest mean percentage predicted lung function values were found in lifetime nonsmokers, whilst the lowest values were found among former smokers, the second lowest among transitional smokers, and constant smokers actually had greater values than both former and transitional smokers. These findings were confirmed by sex-specific regression analyses. A global test on the interactions between smoking and sex was highly significant. This study suggests that adverse smoking effects on pulmonary function were greater in women than in men.(ABSTRACT TRUNCATED AT 250 WORDS)

Antithetic relationship of dietary arachidonic acid and eicosapentaenoic acid on eicosanoid production in vivo.

Eicosanoids are oxidative derivatives of arachidonic acid. When produced in excessive amounts, many are proinflammatory and/or prothrombotic agents. N-3 polyunsaturated fatty acids (PUFA) have been used to attenuate tissue arachidonic acid (AA, 20:4 n-6) levels and thus modulate eicosanoid production. However, there is growing evidence that dietary arachidonic acid may also be able to modulate eicosanoid formation by enriching tissue phospholipids with AA. Therefore, the effects of dietary AA and n-3 PUFA are in diametric opposition. This study investigates the antithetic relationship of dietary AA and eicosapentaenoic acid (EPA, 20:5 n-3) on fatty acid composition of hepatic phospholipids and eicosanoid production in vivo. Forty-nine CD-1 male mice were randomly divided into four dietary groups. Identical diets were supplemented with ethyl esters (1.5% w/w) of the following fatty acids: oleic acid (OA, 18:1 n-9), AA, EPA or AA+EPA. After 4 weeks on diet, peritoneal cells were stimulated in vivo with opsonized zymosan and the peritoneal exudates were analyzed for eicosanoid production (PGE2, 6-keto-PGF1 alpha, TXB2, LTB4, LTE4, and LTE5). Hepatic phospholipids were enriched with AA when AA was included in the diet, and EPA was enriched at the expense of AA when EPA was added to the diet. However, when AA was added to the diet containing equivalent amounts of EPA (AA+EPA), any effect EPA had on modulating hepatic phospholipid fatty acid composition was almost completely eliminated. Similar effects were observed with eicosanoid production. The pooled eicosanoid production in the AA group was 41% and 300% higher compared to the OA (control) and EPA groups, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

[Experimental and clinical study on pneumocystosis. III. Development and characterization of monoclonal antibody against Pneumocystis carinii].

BALB/c mice were repeatedly inoculated subcutaneously with purified rat source Pneumocystis carinii (P.c.) cysts. Six weeks later, the spleen cells were fused with NS1 myeloma cells, and a mouse hybridoma producing monoclonal antibody against P.c. was established and cloned, which was named as 4D7 McAb. Its mean of chromosome was 82. The 4D7 McAb was shown to be IgG1 subclass. It reacted mainly with 54 kDa polypeptide of P.c. in EITB. 4D7 McAb could clearly recognize P.c. by immunohistochemical staining in the specimens from confirmed Pneumocystis carinii pneumonia (PCP) patients, and the sensitivity and specificity were both 100% for diagnosis of PCP (11/11). No cross-reaction of 4D7 McAb was found with Pneumococcus, Candida albicans and Cryptococcus neoformans, as well as all examined normal human tissues.

Effect of albendazole on the larvae and eggs of Necator americanus in golden hamster.

When golden hamsters infected with adult Necator americanus were treated ig with albendazole(Alb) 75 or 150 mg/kg, the efficacy rates were 83.8% and 100%, respectively. Three days before treatment, the mean numbers of larvae recorded from the incubation of feces from 2 treated groups were 29,716 +/- 3,299 and 28,256 +/- 118 per gram of feces, while in the control group, it was 8,828 +/- 2,659. Within 3 days after treatment, no larva was detected in the 2 treated groups, but in the control group the mean larva number was 9,066 +/- 7,416. Furthermore, in 2 treated groups none or only a few degenerated eggs could be found using saturated saline solution method. when infected golden hamsters were treated ig with Alb at a single dose of 150 or 300 mg/kg on day 3, 7 or 14 after infection with the third stage larvae, the egg positive rates in the feces were 0-9.1%, while the worm reduction rates were 96.8-100%.

Effect of albendazole on Ancylostoma caninum larvae migrating in the muscles of mice.

When mice inoculated with 1,000 third-stage larvae of Ancylostoma caninum for 1 week were treated intragastrically (ig) with albendazole (Alb) 75, 150 or 300 mg/kg.d for 3 days, the mean larva numbers collected from the muscles of each group were 2.7 +/- 1.7, 2.0 +/- 1.5 and 1.0 +/- 1.0, respectively, being much less than that 205 +/- 68 of the control group. In mice treated ig with Alb 150 mg/kg.d for 3 days, the concentrations of Alb and its effective metabolite, albendazole sulfoxide (AlbSO), were determined in plasma and the muscles at different intervals after the last medication using high performance liquid chromatography. The results showed that only low concentrations of Alb were detected in both plasma and the muscles. However, higher concentrations of AlbSO were found not only in the plasma (5.4-10.5 micrograms/ml), but also in the muscles (2.2-4.6 micrograms/g). The higher contents of AlbSO in the muscles would be helpful for killing the Ancylostoma larvae migrating in the muscles of mice.

[Chemical constituents of Sageretia theezans Brongn].

Six compounds were isolated from Sageretia theezans and identified as friedeline (1), syringic acid (2), beta-sitosterol (3), daucosterol (4), gluco-syringic acid (5) and taraxerol (6) on the basis of spectral data (1H-NMR, IR, MS) and physico-chemical constants. Among them, compounds 5 and 6 were obtained from the species for the first time.

Fusion expression vectors for recombinant gene products processed easily and purified rapidly by affinity chromatography.

A DNA fragment encoding IgG-binding domain B,C (PABC) was separated from protein A gene, cloned into phage M13 and modified by oligo-directed mutagenesis at the hydroxylamine-cleaved site from Asn-Gly to Asn-Ala in domain B and C, respectively. The modified PABCm gene fragment was used to construct one set of fusion expression vectors in different reading frames. Processing sequences such as those recognized by enterkinase, collagenase, thrombin, activated factor X and cleaved by the hydroxylamine, N-chlorosuccinimide etc. can be created in the fusion site. Using the above vectors, fusion proteins such as PABCm-IGF-I, -hGRF, -bGRF and their derivatives were highly expressed in E. coli. The yield of fusion proteins is over 100 mg per liter cultured by analysis of SDS-PAGE. The PABC fusion proteins can be rapidly purified by the affinity chromatography with a IgG-sepharose column.

Preferential overexpression of a 172Arg-->Leu mutant p53 in the mammary gland of transgenic mice results in altered lobuloalveolar development.

Regulatory sequences derived from the rat whey acidic protein gene have been used to preferentially overexpress a murine 172Arg-->Leu mutant p53 in the mammary gland of transgenic mice. Several different lines of mice expressing the 172Arg-->Leu mutant p53 displayed an impaired ability to lactate, and the mice expressing the highest levels of mutant p53 were unable to nurse their young. This failure was related to the inhibition of normal lobuloalveolar development that occurred during late pregnancy and a marked decrease in milk protein gene expression at early lactation. Interestingly, immunohistochemical analysis revealed that the mutant p53 was localized predominantly in the cytoplasm of alveolar cells. Ductal development was not overtly impaired in these mice. Expression of the 172Arg-->Leu mutant p53 resulted in radiation-induced apoptosis, and transactivation or repression of the expression of a number of genes, including mdm-2 and proliferating cell nuclear antigen, known properties of wild-type p53. The availability of lines of mice preferentially expressing specific p53 mutants in the mammary gland should facilitate evaluation of the roles of other factors, such as hormones, oncogenes and chemical carcinogens, in the etiology of breast cancer.

Effects of vitamin/mineral supplementation on the prevalence of histological dysplasia and early cancer of the esophagus and stomach: results from the General Population Trial in Linxian, China.

A randomized nutrition intervention trial was conducted among 29,584 adult residents of Linxian, China, to examine the effects of vitamin/mineral supplementation on the occurrence of esophageal/gastric cardia cancer in this high-risk population. A fractional factorial study design allowed evaluations of four different combinations of nutrients: (A) retinol and zinc; (B) riboflavin and niacin; (C) vitamin C and molybdenum; and (D) beta-carotene, vitamin E, and selenium. During the 5.25-year intervention, significant reductions in total mortality, total cancer mortality, and stomach cancer mortality occurred among those receiving beta-carotene, vitamin E, and selenium. At the end of intervention, an endoscopic survey was carried out in a sample of subjects to see if the nutritional supplements had affected the prevalence of clinically silent precancerous lesions and early invasive cancers of the esophagus or stomach. Endoscopy was performed on 391 individuals from two study villages. The prevalences of esophageal and gastric dysplasia and cancer were compared by nutrient factor. Cancer or dysplasia was diagnosed in 15% of the participants. No statistically significant reductions in the prevalence of esophageal or gastric dysplasia or cancer were seen for any of the four vitamin/mineral combinations. The greatest reduction in risk (odds ratio, 0.38; P = 0.09) was seen for the effect of retinol and zinc on the prevalence of gastric cancer. Although no significant protective effects were seen in this endoscopic survey, there was a suggestion that supplementation with retinol and zinc may protect against the development of gastric neoplasia in this high-risk population. Additional studies with larger numbers of endpoints will be needed to further evaluate this possibility.
