[Histopathology studies on 8,673 cases of ocular adnexal hyperplastic lesions and tumors].

OBJECTIVE: To study the histopathologic classification of ocular adnexal hyperplastic lesions and tumors according to the histogenesis and the anatomic sites. METHOD: To analyze retrospectively the pathologic specimens of 8,673 cases of ocular adnexal hyperplastic lesions and tumors between 1961 - 1997. RESULTS: There were 7,265 benign cases (83.77%), and 1,408 malignant cases (16.23%). Classification by anatomic sites: eye lid 5,313 cases (61.26%), orbit 1,484 cases (17.11%) and epibulbar 1,876 cases (21.63%). Classification by histogenesis: (1) Primary tumor 6,679 cases: epitheliogenic 2,481 cases (37.15%), mesenchymal 753 cases (11.27%), lympho-hematopoietic 102 cases (1.53%), neurogenic 1,542 cases (23.09%), congenital abnormal embryogenetic 1,767 cases (26.46%), uncertain resource tumor 34 cases (0.50%). (2) Secondary tumor 13 cases; (3) Metastatic tumor 9 cases; (4) Non-tumor lesion 1,972 cases. CONCLUSION: The classification is helpful to the histopathologic diagnosis and the study about the detailed distribution of common ocular adnexal hyperplastic lesions and rare tumors, and it also increases the pathologic diagnostic data in North China.

[Clinical pathological analysis and immunohistochemical expression of primary cervical adenocarcinoma in 98 cases].

OBJECTIVE: To discuss the pathological classification, immunohistochemical expression characteristics, differential diagnosis and factors affecting prognosis in cervical adenocarcinoma. METHODS: Clinical pathological analysis and immunohistochemical study were performed on all primary cervical adenocarcinoma cases (98 patient) from 1991 to 1998. Comparison was made with 18 cases of endometrial adenocarcinoma. 54 cases were followed-up. RESULTS: These primary cervical adenocarcinoma cases fell into 5 categories, of which the endocervical type being the most common. The positive rate of CEA, HPV16 and HPV6B/11 staining were 15/18 (83.3%), 6/9 (66.7%) and 10/18 (55.6%) cases respectively, while the results of ER and vimentin staining were all negative. PCNA staining was strongly positive in all the cases. In the control group (endometrial adenocarcinoma group) the CEA and vimentin staining positive rates were 27.8% and 83.3% respectively, the difference was significant. The two year follow-up indicated that the incidence of recurrence, metastasis and mortality in the endocervical type were higher that those of other types. CONCLUSION: It is difficult to diagnose highly differentiated endocervical adenocarcinoma preoperatively, therefore adequate attention should be attached. Immunohistochemical staining can be used as an important basis for the diagnosis and differential diagnosis of cervical adenocarcinoma. The strongly positive results of PCNA staining, regardless of the degree of differentiation, proves that even highly differentiated adenocarcinoma exhibits a high degree of cell activity. Preliminary data suggests that cervical adenocarcinoma is related to HPV infection. Its relation with each HPV subtype remains to be clarified.

[Determination of the cutaneous viability of skin following incomplete avulsion and its treatment].

OBJECTIVE: In the determination of the viability of skin following incomplete avulsion, subjective criteria such as color, skin temperature, pressure reaction and the stab bleeding would often give a high rate of failure. In order to resolve this problem, a retrospective study was carried out. METHODS: In 27 patients, there were 18 males and 9 females, the age ranged from 7 to 41 years old. In operation, the blood supply of the skin was determined by above subjective criteria carefully. RESULTS: After operation, 5 cases had total survival, 7 cases had peripheral or small area necrosis and 15 cases resulted in large area of necrosis. CONCLUSION: The conclusion was that if the incompletely avulsed skin showed sign of being compressed and squeezed, or the incompletely avulsed skin had uncertain or unstable circulatory status, even though the circulatory status being good, active attitude should be given to debridement in complete removal of the avulsed skin in order to improve the successful rate. For the other 2 cases with degloving injuries of large area of the limbs, the avulsed skin was made into a subdermal vascular network skin flap and several axial incisions were made to save the blood circulation of flap. The result was satisfactory and the vital tissues were preserved and used to the greatest extent.

[Effect of bushenjiannao 1 decoction on learning and memory functions and anti-fatigue ability in mice].

OBJECTIVE: To observe the anti-dementia effect of Bushenjiannao 1 decoction on mice. METHOD: The learning and memory tests were realized with a water maze on the learning and memory impact of mice induced by scopolamine and cycloheximide. The effect of Bushenjiannao 1 decoction on the independent activity and anti-fatigue ability of mice were also investigated. RESULT: Bushenjiannao 1 decoction used at a medium dose of 15 g crude drug/kg and a higher dose of 45 g crude drug/kg could significantly improve the learning and memory abilities of mice injected with scopolamine. Higher doses could improve these abilities of mice injected with cycloheximide, and also improve the anti-fatigue ability of mice. No significant differences were observed in the independent activity of mice between the groups. CONCLUSION: Bushenjiannao 1 decoction helps improve the learning and memory abilities and enhance the anti-fatigue ability, but has no effect on excitability of the central nervous system.

[Changes and its measurement of intracellular Ca2+ during exposure to hyperbaric oxygen].

Objective. To investigate the rule of change of intracellular Ca2+ and its measurement method during exposure to hyperbaric oxygen (HBO). Method. Microfluorescence method was used to measure the changes of intracellular Ca2+ concentration in co-cultured endothelial cells (EC) and smooth muscle cells (SMC) in a specially designed miniature oxygen chamber. Result. (1) [Ca2+]i changes during HBO exposure can be clearly observed directly in the chamber. (2) Within 5 min during 0.2 MPa and 0.3 MPa HBO exposure, EC[Ca2+]i and SMC[Ca2+]i showed a tendency, to increase at first and then decrease with time throughout the exposures, especially the SMC[Ca2+]i. The EC ruptured under exposure to 0.4 MPa HBO. Conclusion. During HBO exposure, EC[Ca2+]i and SMC[Ca2+]i showed biphasic changes, which increased at first and then decreased. The technique and method used were reliable and suitable for investigating of intracellular [Ca2+]i changes under HBO.

[Changes of simple epithelial cytokeratin during oral carcinogenesis].

OBJECTIVE: To investigate whether the simple epithelial cytokeratin CK18 and CK19 can be used as a marker of oral precancerous lesions. METHODS: Formalin-fixed, paraffin-embedded tissue sections of normal oral mucosa, epithelial hyperplasia, mild epithelial dysplasia, moderate epithelial dysplasia, severe epithelial dysplasia and oral squamous cell carcinomas were stained with a CK18-specific antibody and CK19-specific antibody respectively by LSAB immunohistochemical method. The stained sections were observed under light microscopy. The results were described and analyzed with Rank Sum Test. RESULTS: CK18 was not detected in normal and abnormal oral tissue sections. But in normal nonkeratinized mucosa, CK19 was detected in the basal cell layer dispersively. In epithelial dysplasia, CK19 was detected in the suprabasal cell layer and the number of CK19-positive cell layers was correlated with the dysplasia degree of epithelia. Furthermore, CK19 was detected in oral squamous cell carcinoma, especially in the poor-differentiated cancer cells. CONCLUSION: CK19 expression in suprabasal cell layer of oral mucosa can be used as a candidate marker for diagnosis of oral precancerous lesions and determination of the differentiation level of oral squamous cell carcinoma.

[Expression of CDK4 protein in oral carcinogenesis].

OBJECTIVE: To study the expression of cyclin dependent kinase 4(CDK4) protein in oral carcinogenesis. METHODS: An LSAB immunohistochemistry method was applied to 80 cases of paraffine-fixed tissues to study the expression and distribution of CDK4 in normal oral mucosa, oral premalignant lesion (OPL) and oral squamous cell carcinoma. RESULTS: The overexpression of CDK4 protein was observed in the progression of OPL. Meanwhile, a correlation analysis was applied to investigate the relationship between P16 and CDK4 expression, and the result showed that the coefficient of correlation was 0.813, which was statistically significant. CONCLUSION: The overexpression of CDK4 is involved in the progression of OPL, and there is a positive feedback loop between CDK4 and P16.

[Psychological aspects of patients with burning mouth syndrome: an investigation of personality, psychopathological profiles and social life background stress].

OBJECTIVE: To know the psychosocial background of patients with burning mouth syndrome (BMS) in China. METHODS: In this study, 30 patients with BMS and 30 normal subjects of control group were evaluated for their personality, psychopathological profile and existence of recent life events. All subjects were required to complete the EPQ questionnaire, the SCL-90 questionnaire and a Life Event Scale, and chi-square test analysis of variance and rank sum test were used in this study. RESULTS: The personality of introversion and instability was common in BMS group. The scores of 9 emotional factors and negative life events were higher than that of the control group significantly. CONCLUSION: BMS may be a series of psychological disorder and somatic discomfort caused by stresses of social life events on the people with special personality.

[Cloning and sequence characterization of heavy chain variable region gene from hybridoma cells secreting MAB against VEGF].

OBJECTIVE: To clone the variable region of heavy chain (VH) gene from hybridoma secreting monoclonal antibody against human vasoendothelial growth factor (VEGF) for construction of recombinant antibody (scFv). METHODS: One step method was used to extract total RNA and degeneration primer was used to amplify the cDNAs by retropolymerase chain reaction (RT-PCR), then the products were inserted to T-vector and its sequence was analysed. RESULTS: The VH gene contained 369 base pairs encoding 123 amine acid resides. Analysis of the nucleotide sequence of the light chain variable region revealed that the VH sequence was 87% identical to the sequence present in mouse Ig VH region. CONCLUSION: The VH gene was 369 bp. According to Kabat classification, the VH gene belonging to mouse kappa light chains subgroup I (A), resulting from VH-D-JH4 rearrangment.

[The effect of partial middle turbinectomy upon the frontal sinus].

OBJECTIVE: To investigate the effect of partial middle turbinectomy under endoscopy upon the frontal sinus. METHOD: A retrospective analysis of 87 patients undergone endoscopic sinus surgery was made. RESULT: Among 55 sinusitis patients (no frontal sinus) suffered Fess, 29 cases had partial middle turbinectomy and 7 cases were found postoperative frontal sinusitis. In other 26 cases without partial middle turbinectomy, 5 cases were found frontal sinusitis. No significant difference between two groups was found (P > 0.05). In 32 nonsinutis patients no postoperative frontal sisusitis was found. CONCLUSION: The partial middle turbinectomy has no effect on the incidence of frontal sinusitis.

[Prevention of keloids of the earlobes].

OBJECTIVE: To investigate the causing of keloids of the earlobes. METHOD: We have treated 41 keloides of earlobes in 28 cases, and analyzed the factors. RESULT: It has been found that infections, inappropriate supporter, allergy to nickel and gold, local delayed allergy are the chief factors to form keloides of the earlobes. CONCLUSION: Some measures to prevent those cousing mentioned above have been proposed.

[Clinical study on antibodies against EBV in sera of patients with rheumatoid arthritis].

OBJECTIVE: Study on the relationship between the infection of Epstein-Barr virus (EBV) and the pathogenesis of rheumatoid arthritis (RA). METHODS: The IgA and IgG antibodies against EBV capsid antigen (IgA/VCA and IgG/VCA respectively), and anti-Z protein IgG antibodies (IgG/Z) in the sera from the patients with RA, systemic lupus erythematosus (SLE), and normal controls were detected by using indirect immunofluoresence and immunobloting techniques. RESULTS: The positive rate of IgA/VCA antibody in the serum of RA patients which was significantly higher than that in SLE patients and normal subjects. The anti IgG/Z antibodies were only found in RA patients with IgA/VCA antibody. CONCLUSIONS: The results suggested that activated EB virus may play a role in the pathogenesis of RA. It is a useful laboratory method for the clinical diagnosis of RA.

[Detection of hepatitis C core antigen C33].

OBJECTIVE: To investigate the distribution of C33 antigen of HCV in serum and liver tissues from normal subjects and patients with different diseases. METHODS: Balb/C mice were immunized by HCV core antigen peptide C33(33 amino acids) which was conjugated with mice serum albzmin by a heterotypic difunctionalconnector(SPDP). Two strains of monoclonal antibody to C33 were produced. By using one of them as coated and the other as HRP(horseradish peroxidase)-labelled antibodies, an ELISA was developed to detect C33 antigen of HCV in 1236 serum samples from patients. RESULTS: The detectable rate of peptide C33 was 14.0%(6/43) in acute hepatitis C group, 8.8%(35/396) in anti-HCV-IgG positive group, 6.5%(4/62) in chronic hepatitis C group, 4.3%(1/23) in leukemia group, 3.3%(19/576) in hemodialysis group, 1.5%(2/136) in other diseases group, and 1. 4%(6/438) in blood donors. Results of immunohistochemistry and immunofluorescence for liver tissues obtained from biopsy showed that C33 peptide was found in nucli, cytoplasm and membranes of hepatic cells, scattered or laterally concentrated in the cytoplasm. CONCLUSION: The detection of C33 antigen of HCV may consider as one of the markers of HCV infection.

[Preliminary study on NS5 region gene sequencing and variants of hepatitis G virus in China].

OBJECTIVE: To study the genomic specificity and variation of Hepatitis G virus(HGV) in China. METHODS: The 141nt cDNA fragments locating at nonstructural region 5 (NS5) of HGV were obtained from the sera of 16 HGV infected patients by RT-nested-PCR using NS5 primers, then purified and directly sequenced by dideoxynucleotide chain termination method. RESULTS: The sequence diversity near the N-terminus of the region between nt 7,820 and 7,960 was more marked than that near the C-terminus. Compared with the HGV-US isolate, the nt alterations of NS5 in Chinese isolates usually took place at nt 7,824, 7,892, 7,914, 7,917, 7,935 and in region nt 7,845-7,860, the rate of variability at one nt position was 0-89.5%. The nucleotide homology was 83.6%-95.7% (89.6% in average) and the homology of deduced amino acid sequence was 83.0-100%(91.2% in average). CONCLUSION: The nucleotide and deduced amino acid sequences of NS5 of Chinese HGV are relatively conserved but are obviously different from the HGV and GBV-C reported in USA in this study.

[GFAAS determination of trace lead in toothpaste using L'vov platform].

Sensitivity in determining low melting point metals is highly improved by means of platform technique in graphite furnace AAS. In this paper we describe a method of determining trace lead in toothpaste by this technique. The method of making the platform and the selection of the platform coating were discussed and the results with and without the platform were compared. The optimum ashing temperature, atomization temperature and experimental conditions were investigated. The interference of large amount Al and Ca in the matrix on determination of trace lead can be minimized by using platform technique. The precision of about 0.9% and detection limit of about 15 ng obtained by this improved method are much better than those by methods without platform.

[Spectroscopic study on the structure and orientation of 1-hydroxyl-5-dodecyloxy-naphthalene LB films].

Ultraviolet-visible (UV-Vis), infrared transmission reflection absorption (RA) and ATR spectra were used to study the mono-and multi-layer Langmuir-Blodgett (LB) films of C12H25ONaph (1,5)OH at various temperatures to investigate dependencies of the aggregation and phase transitions on the number of monolayers. The UV-vis spectra of the LB films revealed that C12H25ONaph (1,5)OH forms aggregates in the LB film. From the temperature dependence of UV-vis absorption spectra, it is clear that the aggregate was gradually converted to monomer with the temperature increased. A comparison of the infrared transmission and ATR spectra reveals that the hydrocarbon chains were tilted considerably in the LB film.

Increased hydroxyl radical production and apoptosis in PC12 neuron cells expressing the gain-of-function mutant G93A SOD1 gene.

Mutations of the SOD1 gene (formerly known as Cu,Zn-SOD) are frequently associated with the familial form of amyotrophic lateral sclerosis (ALS). The G93A mutation of SOD1 with substitution of Gly to Ala at residue 93 results in gain of a peroxidative function. Here we report that transfection of PC12 neuron precursor cells with the G93A mutation of SOD1 results in increased production of hydroxyl radicals (*OH) and an enhanced rate of cell death by apoptosis. Notably, PC12 cells transfected with the H63C/G93A mutant of SOD1 with a mutation in the catalytic site that converts histidine at position 63 to cysteine showed a dramatically reduced production of *OH and rate of death by apoptosis. Thus the gain of function of the mutant G93A SOD1 can be reduced by an active site mutation. These results provide additional genetic evidence for the hypothesis that the increased *OH production and induced cytotoxicity in neuron cells expressing the mutant G93A SOD1 results from the gain of peroxidative function by the enzyme's catalytic site.
