Teratological evaluations of atrazine technical, a triazine herbicide, in rats and rabbits.

Atrazine technical was evaluated for its embryotoxic, fetotoxic, and teratogenic potential in both rats and rabbits. The compound was orally administered at doses of 0, 10, 70, or 700 mg/kg.d to groups of rats on gestational d 6-15, while rabbits were administered doses of 0, 1, 5, or 75 mg/kg.d on gestational d 7-19. Maternal toxicity was observed at doses greater than or equal to 70 mg/kg.d in rats and at doses greater than or equal to 5 mg/kg.d in rabbits. Minor fetal effects, concurrent with maternal toxicity, were observed in rats at doses greater than or equal to 70 mg/kg.d. Among rabbits, fetal effects concurrent with severe maternal toxicity were only observed at the 75 mg/kg.d dose level. There were no adverse maternal or fetal effects in either rats or rabbits at the low dose levels. These findings indicated that pregnant rabbits were more sensitive than pregnant rats to the effects produced by atrazine technical and the compound was not teratogenic at maternally toxic dose levels in either species.

Non-invasive ultrasonic study of maximal aortic blood acceleration in patients with borderline and sustained essential hypertension.

Using a pulsed Doppler method to evaluate the aortic velocity curve transcutaneously, the maximal aortic blood acceleration was evaluated in normal subjects and in age-matched patients with borderline and sustained essential hypertension. Maximal aortic blood acceleration was significantly elevated in patients with borderline hypertension and remained within the normal range in patients with sustained hypertension. Since maximal aortic blood acceleration is relatively insensitive to preload and afterload, these findings might give indirect information on the contractile state of the myocardium in patients with hypertension.

Comparison of quality of life on nitrendipine and propranolol.

A multicenter, randomized, double-blind, comparative study was conducted in 274 patients with mild to moderate hypertension to assess the impact of nitrendipine and propranolol on quality of life. After placebo baseline, 136 patients were given nitrendipine (5-20 mg b.i.d.) and 138 were given propranolol (40-120 mg b.i.d.). Quality of life was evaluated at baseline, weeks 6-10, and weeks 14-18 of the maintenance period. At weeks 6-10, the nitrendipine group became significantly more vigorous (p less than 0.01) and less fatigued (p less than 0.05) than the propranolol group. Propranolol subjects noted decreased problems of trembling hands (p less than 0.01) and alcohol use (p less than 0.05) than the nitrendipine subjects. No other significant differences between groups in mood states, troublesome conditions (insomnia, headaches, and loss of appetite), or sexual satisfaction were noted at this visit, and patient willingness to continue study medication was marginally significantly higher (p less than 0.1) in the nitrendipine group than in the propranolol group. At weeks 14-18, the propranolol subjects perceived significantly decreased problems with the "felt worried, tense, and drank alcohol to cope" factor (p less than 0.05); however, there were no differences between groups at this visit for Profile of Mood States (POMS) scores, sex life variables, or medication preference. Based on within-group analysis, the propranolol group perceived a reduction in partner sexual satisfaction (p less than 0.05). Overall, nitrendipine seemed to be better tolerated than propranolol.

[Cleft lip and palate in Campeche Mayas].

It has been suggested that among American Indians, as in some genetically-related Asiatic ethnic groups, incidence of cleft lip and/or cleft palate is higher than among people of Caucasian extraction. Such hypothesis, plus growing demand for services observed at a center for the surgery of cleft lip and cleft palate in Campeche state, led the authors to undertake research among the Maya residents of that region. However, neither careful review of case histories nor field research performed in several Indian communities could confirm the hypothesis of a higher incidence among this ethnic community.

Effect of vasodilatators on the structure of the aorta in normotensive ageing rats.

The effects of hypertension and its treatment on the function and structure of arteries have been extensively studied, but no data are yet available on the effect of hypotensive drugs on the large arteries in normotensive rats. Two groups of 11-month-old normotensive breeder male rats were treated for 5 months, one with an angiotensin converting enzyme (ACE) inhibitor (MK-421, 2 mg/kg per day; n = 10) and the other with dihydralazine (15 mg/kg per day; n = 8). A group of 14 rats served as controls. Blood pressure was recorded every 14 days by the tail-cuff method. At the end of the treatment period (5 months), the rats were killed under anaesthesia and the descending thoracic aorta was removed and fixed. The different components of the aorta were assessed by automated morphometric image analysis after specific coloration of elastin (orceine), collagen (sirius red) and nuclei (haematoxylin after periodic acid oxidation. Both the ACE inhibitor and dihydralazine caused similar decreases in systolic blood pressure (SBP) compared with controls. This hypotensive effect was associated with a reduction in medial thickness, from 120 +/- 15 microns in controls to 104 +/- 9 microns in ACE inhibitor-treated and 103 +/- 10 microns in dihydralazine-treated normotensive rats. Elastin density significantly increased in the two treatment groups but the greatest increase was in the dihydralazine-treated group (P less than 0.001). Elastin fibre thickness increased significantly in the dihydralazine-treated group only. Collagen density was not significantly modified by either treatment.(ABSTRACT TRUNCATED AT 250 WORDS)

Ontogeny of the bovine neurohypophysial hormone precursors. III. Identification of neurohormones, neurophysins and copeptin in the early bovine fetus.

Neurohypophysial hormone precursors are small proteins processed into several fragments during axonal transport from hypothalamus to neurohypophysis. From 3-month-old fetal bovine pituitaries the three fragments of vasopressin precursor, arginine vasopressin, MSEL-neurophysin and copeptin, and the two fragments of oxytocin precursor, oxytocin and VLDV-neurophysin, have been isolated and characterized. These polypeptides are identical to those previously identified in the late fetus (7-9 months old) and in the adult. It is concluded that the same genes are expressed during fetal and adult lives, the vasopressin gene appearing roughly four times more active than the oxytocin gene in the early fetus. Vasotocin, mesotocin and additional neurophysin have not been detected in the early fetus.

A comprehensive study of chronic schizophrenic patients. I. Quantitative computed tomography: cerebral density, ventricle and sulcal measures.

Based on recent computed tomography (CT) studies that have suggested the presence of distinct structural abnormalities in the brains of chronic schizophrenics, the authors evaluated 23 chronic schizophrenics and 23 controls by CT, using linear and area lateral ventricular measures, sulcal and Sylvian fissure widths and cerebral density measures. Technical problems inherent in CT studies and their impact on the use of data generated by such studies are discussed. Although there were few differences between groups, there emerged a distinct subgroup of schizophrenic patients with mild lateral ventricle enlargement associated with increased cortical density. The possible etiopathology for this atypical combination is discussed, including the striking parallels with a recent neuropathological study's findings.

A comprehensive study of chronic schizophrenic patients. II: Biological, neuropsychological, and clinical correlates of CT abnormality.

There are numerous reports of lateral cerebral ventricle enlargement on computed tomography (CT) in schizophrenics, but the significance and its relationship to traditional notions of organicity remain unclear. Therefore we studied a subgroup of chronic schizophrenics who had lateral ventriculomegaly (and also cortical hyperdensity) on a battery of relevant biological, neuro-psychological, and clinical parameters such as electroencephalogram (EEG), platelet monoamine oxidase (MAO) and serum dopamine-beta-hydroxylase (DBH) activity, the Halstead Reitan Neuropsychological Battery (HRB), premorbid personality adjustment, drug response, positive and negative symptoms, employment history, and family history. Our findings support the notion that there is an "organic" subgroup of schizophrenia that has 1) CT structural abnormalities such as lateral ventricle enlargement and cortical hyperdensity; and cerebral dysfunction or deficits as evidenced by 2) an increased incidence of abnormal EEGs and also 3) greater impairment on neuropsychological tests. The biochemical measures, platelet MAO and serum DBH activity, nor any of the clinical measures could differentiate between the subgroups. The implications of these findings for the subtyping of schizophrenia are discussed.

Upper extremity sensory feedback therapy in chronic cerebrovascular accident patients with impaired expressive aphasia and auditory comprehension.

Electromyographic (EMG) sensory feedback therapy (SFT) was used in the neuromuscular retraining of the nonfunctional upper extremity in five chronic left cerebrovascular accident (CVA) patients with impaired expressive and auditory comprehension. Speech diagnoses included global, moderate-to-severe Broca, and Wernicke aphasias. These patients had experienced increased despondency associated with previous therapy failures and often had indicated that they wished to have their involved extremity amputated, so that it would no longer be in the way. In this study, specific behavioral training strategies to increase patient involvement were used, including: general relaxation, modified SFT instruction, and home exercises, which were supported by family and/or friends. After an average of 50 therapy sessions, all patients were successfully retrained to use their right upper extremity at the gross-assist level. This resulted in feelings of increased self-esteem to the extent that amputation was no longer requested. It is concluded that EMG SFT can be beneficial in the neuromuscular reeducation of paretic upper extremity muscles of CVA patients with expressive aphasia and (impaired) auditory comprehension.

Ontogeny of bovine neurohypophysial hormone precursors. II. Foetal copeptin, the third domain of the vasopressin precursor.

Vasopressin, MSEL-neurophysin and a glycopeptide, here referred to as copeptin, are three fragments of a common protein precursor processed during axonal transport from hypothalamus to neurohypophysis. Neurohormones and neurophysins purified from 7-9-month-old bovine foetuses have previously been shown to be identical with those found in the adult. Copeptin has now been isolated from 7-9-month and 3-month-old bovine foetuses and chemically characterized. It can be concluded from the nature of the three precursors that the same vasopressin gene is expressed in the adult and the 7-9-month-old foetus.

Quantitative Doppler blood flow measurement method and in vivo calibration.

Femoral blood flow (FBF) was measured in seven dogs, simultaneously with both an electromagnetic perivascular probe and a transcutaneous range gated Doppler velocimeter. Measurements were made in basal conditions and during intraarterial infusions of noradrenaline (10 to 400 ng . kg-1 . min-1) and isoprenaline (10 to 400 ng . kg-1 . min-1) thus allowing comparisons of ultrasonic (DBF) and electromagnetic (EMBF) blood flow at 91 different blood flow rates ranging from 5 to 300 cm3 . min-1. The linear regression line through the data of ultrasonic and electromagnetic simultaneous measurement was: DBF = 0.8 + 1.016 EMBF +/- 19.0 cm3 . min-1 with a highly significant correlation (r = 0.96, p less than 0.001) but there was a wide scattering about the mean. Errors in DBF were mainly due to positioning of the probe and determination of arterial diameter.

Quantitative ascending aortic Doppler blood velocity in normal human subjects.

We have developed digital, Apple II microcomputer-based methods for the numerical analysis of pulsed, range-gated, ultrasonic Doppler blood velocity signals. These methods were then used to analyse Doppler data recorded every 5 ms from the ascending aorta via the suprasternal notch in normal subjects ranging in age from 3 to 62 years. Normal values for peak velocity, the integral of velocity over the time of systole, and the rate of change of velocity in early ejection are reported. It was found that, after an initial step increase, the velocity of blood flow in early ejection increased in a linear manner in more than two thirds of the individual beats analysed. The time for which the linear acceleration in the aorta was constant (circa 50 ms) was unrelated to the age or size of the subject.

Simultaneous Doppler blood velocity measurements from aorta and radial artery in normal human subjects.

We used two independent, pulsed, range-gated, ultrasonic, Doppler blood velocity meters to record blood velocities in the aorta and a peripheral artery in 32 normal subjects aged 8 to 62 years. Aortic signals were obtained from an unfocussed transducer in the suprasternal notch using a 2.25 MHz instrument. Simultaneous tracings were obtained from the radial or posterior tibial artery using an 8 MHz instrument. The audio Doppler signals were subjected to spectral analysis and mean velocity was calculated at 5 ms intervals during 11 successive heart beats at each site. The increase in mean velocity at the start of systole in the aorta followed a linear pattern for the first 45 ms of ejection in two thirds of the beats, irrespective of the age or size of the subject. A similar linear velocity increase in early systole was seen in the peripheral arterial signals after a delay due to the time taken for the flow wave to pass to the periphery. Thus the constant acceleration seen in aortic blood velocity tracings is transmitted to peripheral arteries in an attenuated and delayed but undistorted form.
