[Quantitative determination of alpha1-antitrypsin in lung tissue from adults with emphysema and newborns with respiratory-distress-syndrome (author's transl)].

In a small number of patients, lung emphysema can be attributed to a remarkable deficiency of alpha1-antitrypsin. Concentrations reduced about 50% of the normal value represent a predisposing factor. In most cases of newborns with respiratory-distress-syndrome the concentration of alpha1-antitrypsin is decreased. Its determination can be used for the early diagnosis of the respiratory-distress-syndrome.

[Experimental model of bronchial asthma (author's transl)].

Allergic asthma-like attacks were induced in guinea pigs by intraperitoneal injections of Ovalbumin and following inhalation of Ovalbumin aerosol. The animals were narcotized, tracheotomized and a small catheter was placed in mid-oesophagus. The quantities: oesophageal pressure, respiratory flow and tidal volume were measured and recorded on magnetic tape during spontaneous breathing. A technique was applied for automated computation of the parameters of respiratory mechanics using a digital computer. The parameters compliance, resistance, work of breathing and power of breathing showed typical pathological variances and characteristic response-curves in the experimental phases during and after the asthma attacks. These parameters became normal after the injection of Alupent.

[The appearance of surface-active phospholipids of the surfactant-system of the lung during ontogenetic development of humans (author's transl)].

In order to characterize the pulmonary maturity during ontogenetic development the following parameters were determined in the lungs of 30 fetuses respectively newborns with a weight between 15 and 4450 g: Water content (WC), total phospholipid content (TPC), lecithin concentration (LC), sphingomyelin concentration (SPC) and surface activity (SA) of the extracts. The water content decreases with increasing gestation age. The total phospholipid content and the lecithin concentration increase during the ontogenetic development. The sphingomyelin concentration remains approximately constant during lung development. No significant correlation exists between phospholipid content and surface activity. A relatively high TPC and a high SA was found in a 43 g-fetus (about 14th week of gestation). This result suggests, that the surfactant-system of the lung is produced earlier than supposed till now. Lungs of infants who developed respiratory distress syndrome (RDS) have a low concentration of TPC. The lecithin/sphingomyelin-ratio rises as gestation progresses. Lungs of RDS-children have - similarly in amniotic fluid - a low lecithin/sphingomyelin-ratio.

[Rapid test for quantitative proof of emphysema].

For the purpose of an aimed diagnostics near to practice of unspecific chronic pulmonary diseases a measuring place was developed, by means of which with simple performance of the experiments and short examination-time the determination of vital capacity, forced expiratory volume, maximal breathing capacity, the measurement of the O2-tension of the arterialised ear blood and the determination of the residual volume are possible. The determination of the residual volume is performed according to the classical O2-N2-gasmixture method using the spirotest device in connection with a bag-box system and the PO2-micro-measuring chamber. On 63 patients the residual volume was determined in this measuring place and compared with the rapid He-re-breathing method. The correlation analysis between the RVbody%TK- and the RVO2% TK-values with a correlation coefficient r = 0,887 ascertains the reliability of the demonstrated method. The results of a epidemiological study on 422 test persons on account of the close correlation between RVHe % TK and RVO2% TK with r = 0.62 prove the usability for examinations of the population.

Properties of 4-ethenyl and 4-ethynyl analogs of pyridoxal phosphate and their reactions with the apo form of asparatate aminotransferase.

The binding to apoaspartate aminotransferase of analogs of pyridoxal phosphate bearing vinyl, cis- and trans-methylvinyl, and ethynyl groups in place of the fomyl group of the coenzyme has been studied. Details of synthesis of the ethynyl compound are given. The absorption spectra of all of the compounds have been analyzed and pKa values have been determined. The positions of the absorption bands can be related to those of pyridoxine but with bathochromic shifts induced by the ethenyl and ethynyl groups. However, this shift is almost completely lacking for the cis-methylvinyl compound suggesting nonplanarity of the molecule. Binding of the analogs to the apoenzyme is accompanied by a strong bathochromic shift which, from a study of solvent effects on the free analogs, appears to indicate a hydrophobic environment on the enzyme. Nevertheless, the analogs are bound as dipolar ions exclusively. Binding is accompanied by a distinct perturbation of the protein spectrum in the aromatic region. An effect on the spectrum of 1 or more tryptophan residues is indicated. Bands of the bound analogs exhibit positive circular dichroism except for that of the 4-vinyl analog. The 4-ethynyl analog reacts in a more complex way, giving at least two successive products in addition to the initial complex. The final product is reducible by sodium borohydride, is released from the enzyme by boiling, and appears to have the properties of a Schiff base. We postulate that the addition of an amino group of the enzyme to the ethynyl group is followed by tautomeric rearrangement to a Schiff base in which the ring is in a p-quinonoid structure.

Immunologic differentiation of proteins in the surfactant system of the lung.

Immunoelectrophoresis revealed the following four protein components in the lung surfactant system of guinea-pigs: albumin, transferrin, IgG, and an organ-specific protein migrating in the beta-globulin range. These proteins probably take part in the formation of the hypophase of the alveolar lining layer and thereby influence the surface properties of the alveolar air-liquid interface.
