Multistep origin of tumor-forming ability in Chinese hamster embryo fibroblast cells.

Twenty-one anchorage-independent subclones and ten subclones with reduced serum requirements were isolated as single-step mutants spontaneously or after ethylmethanesulfonate or N-methyl-N'-nitro-N-nitrosoguanidine mutagenesis of CHEF/18 diploid Chinese hamster embryo fibroblasts. Anchorage-independent mutants retain the high serum requirement and nontransformed morphology typical of CHEF/18. Only four of 21 anchorage mutants have spontaneously produced tumors when injected at 10(7)/site in nude mice, and these were only at a fraction of sites. Low-serum (LS) mutants acquire transformed morphology and increased anchorage-independent growth simultaneously with the loss of high-serum requirement. Only two of ten LS mutants have spontaneously produced tumors. However, when some anchorage and LS mutants were remutagenized and when mutagenized populations were injected into nude mice, tumors appeared at many of the injected sites. In contrast, untreated CHEF/18 cells have never given tumors(0 of 34 sites), and mutagenized DHEF/18 cells have given tumors at only three of 29 sites. These results demonstrate that malignant transformation is a multistep process in the Chinese hamster embryo fibroblast system. Most one-step mutants selected for anchorage independence or reduced serum requirements do not have tumor-forming potentials higher than that of the parent CHEF/18. Thus anchorage-independent and LS phenotypes per se do not account for the increase in tumor-forming potential. It is proposed that the genomic rearrangement process as well as specific mutations may contribute to tumorigenicity.

The role of polytomography in the diagnosis and treatment of cervical spine injuries.

Ninety-one sets of polytomograms were analyzed for diagnosis and treatment of 123 patients with cervical spine injuries. Pluridirectional polytomographis is valuable in evaluation of acute cervical spine injuries when the findings on plain roentgenograms do not correlate well with the clinical symptoms and signs. The polytomographic study had a dramatic impact on the diagnosis and a lesser but nevertheless significant impact on the treatment of cervical spine injuries. Polytomograms were especially valuable for the diagnosis and treatment of atlantoaxial injuries.

A comparative study of pre-meaningful vocalizations produced by normally developing and Down's syndrome infants.

Pre-meaningful vocalizations produced by nine normally developing and 10 Down's syndrome infants were recorded as part of a longitudinal study of language development. The recordings were phonetically transcribed using a modified version of the International Phonetic Alphabet. Data were analyzed in terms of (1) age at onset of reduplicated babbling (2) developmental trends for place of consonant articulation, and (3) developmental aspects of vocalic productions. In general, substantial similarities between the two groups of infants were observed with regard to the selected parameters. Both groups began to produce canonical, reduplicated babbling at 8-8 1/2 months of age, and trends regarding consonantal and vocalic development for the two groups were very similar during approximately the first 15 months of life.

The effects of prenatal stress on rat offsprings' learning ability.

The present study (N = 30 female rats, 11 male rats) was conducted to determine the effects of prenatal stress on rat offsprings' learning ability. Pregnant mothers were stressed by exposure to an SD (light) which had been paired with shock. A second group of pregnant rats was handled an equivalent amount of time, and a third group received no treatment. The offspring of the three groups were tested on several learning tasks and the handled and stressed groups were found to be inferior to the control group on four of six learning measures.

The haemodynamic, histopathological and hormonal features of alcoholic cardiac beriberi.

Five cases of cardiac beriberi occurring in chronic alcoholics are described. The clinical diagnosis was based on the presence of biventricular failure, low dietary intake of thiamine and the therapeutic response to oral thiamine. Complicating cardiac disease was excluded by haemodynamic studies, left ventriculography; coronary angiography and endomyocardial biopsy. Haemodynamic measurements including quantitative left ventriculography are reported. They indicate that left ventricular function is depressed despite elevated cardiac output. Biopsy material was studied by light and electron microscopy. No lesion specific to beriberi was detected by either technique although the biopsies were quantitatively abnormal. The histological changes resemble those in early reports based on necropsy material, and consist of vacuolation and intercellular oedema in the early stages with myofibre hypertrophy, fibrosis and cellular infiltration in the chronic cases. The transketolase test and response to intravenous thiamine during catheter studies are valuable diagnostic tests. Plasma renin, angiotensin II and aldosterone levels were lower than in patients with low output heart failure. The incidence of cardiac beriberi appears to be greater than is generally realized.

Simultaneous combination radiotherapy and multidrug chemotherapy for stage III and stage IV squamous cell carcinoma of the head and neck.

Combined simultaneous radiotherapy and multidrug chemotherapy have been utilized in an attempt to eradicate or shrink tumors of the head and neck area in advanced stages to allow subsequent surgical extirpation. Thirty-six patients (1 stage II; 11 stage III; 24 stage IV; 1 unknown primary) were treated with simultaneous radiotherapy and fractionalized doses of bleomycin, adriamycin, and 5-fluorouracil. Thirty of 36 patients completed the treatment schedule. Twenty-nine of 30 (97%) had a 75% or greater response to treatment; 20/30 (66%) had 100% response; 9/30 (30%) had 75% response; and 1/30 had 50% response. Of the 30 patients completing the regimen, 19/30 (63%) are alive three to 27 months later, and 12 (40%) of these have no evidence of disease. Six patients died within four months of initiation of therapy, mostly from complications of their disease. This treatment regimen appears effective in the control of locoregional disease or to produce enough shrinkage of tumor to allow subsequent surgical extirpation.

Loss of epidermal growth factor requirement and malignant transformation.

Serum provides growth factors that regulate and limit the growth of normal cells in tissue culture. Animal cells that are malignantly transformed usually exhibit diminished serum requirements for growth in culture. We have used a defined, serum-free medium to determine which of these growth factors becomes dispensable for the growth of transformed Syrian and Chinese hamster fibroblast cells. The medium's four growth factors-epidermal growth factor (EGF), insulin, fibroblast growth factor, and transferrin-were added or omitted as desired. A decreased requirement for EGF was most closely related to tumorigenicity of chemically (ethyl methanesulfonate) transformed cells in nude mice. All lines examined retained their requirement for transferrin, which is needed throughout the growth cycle, in contrast to the other factors, which are needed primarily in G(1) phase. Lines that had lost their EGF requirement but had retained their insulin requirement were arrested in G(1) by insulin deficiency, indicating that their growth control system remained. Mutagenesis with ethyl methanesulfonate can also create requirements of the transformed cells for unknown factors in serum. We conclude that an initial step that reduces the serum requirement in culture, and in tumorigenesis, is relaxation of the growth-regulatory function of EGF.
