The hydrolysis of (14)C-labeled ribulose-1,5-bisphosphate carboxylase (RuBPCase) by two partially purified endoproteinases from senescing barley (Hordeum vulgare v. Numar) leaves is described. The major thiol proteinase, EP(1), exhibits biphasic kinetics which appear to be caused by a region of the large subunit of RuBPCase that is highly sensitive to attack by EP(1). This proteinase further hydrolyzes both the large and small subunit to smaller peptides. A second proteinase, EP(2), appears to convert the small subunit of RuBPCase rapidly to a 13.7-kilodalton fragment during initial stages of hydrolysis and then to degrade both this fragment and the large subunit. The presence of a third endoproteinase, EP(3), was discovered when [(14)C]RuBPCase, which appeared to be homogeneous by sodium dodecyl sulfate polyacrylamide electrophoresis, seemed to undergo very low but significant rates of "autolysis." The large molecular weight fragments produced by EP(3) were different from those of EP(1) and EP(2).

Two major endoproteinases were purified from senescing primary barley leaves. The major enzyme (EP(1)) appeared to be a thiol proteinase and accounted for about 85% of the total proteolytic activity measured in vitro. This proteinase was purified 5,800-fold and had a molecular weight of 28,300. It was highly unstable in the absence of dithiothreitol or at a pH greater than 7.5. Leupeptin, at a concentration of 10 micromolar, inhibited this enzyme 100%. A second proteinase (EP(2)) was purified approximately 50-fold and had a molecular weight of 67,000. It was inhibited 20% by 1 millimolar dithiothreitol and 50% by 1 millimolar phenylmethyl sulfonylfluoride. EP(2) contributed about 15% of the total proteolytic activity measured in vitro. Both proteinases hydrolyzed a variety of artificial and protein substrates, and both had pH optima of 5.5 to 5.7 when either azocasein or [(14)C]ribulose-1,5-bisphosphate carboxylase ([(14)C]RuBPCase) was the substrate. The thiol endoproteinase hydrolyzed azocasein linearly but hydrolyzed [(14)C]RuBPCase biphasically. A third endoproteinase (EP(3)), not detected by standard proteolytic assays, was observed when [(14)C]RuBPCase was the substrate.

The efficacy of chrysotherapy appears to increase with prolonged administration. In patients who were followed for 3 years after gold therapy was started, remissions occurred in 73% of those who continued chrysotherapy. Only 6% of those who stopped gold injections during the first 18 months subsequently developed "spontaneous" remissions, although initially both groups had comparable moderately severe rheumatoid arthritis of 6 to 8 years' duration. Therefore, in the absence of definite toxicity, maintenance gold injections should be continued indefinitely.

A case of ventricular parasystole is presented which appeared in an otherwise healthy newborn infant. The dysrhythmia persisted until 10 months of age. In contrast to its occurrence in adults, this rare dysrhythmia appears not to be associated with primary cardiac disease in children.

Circadian variations in whole brain tryptophan level and food intake were investigated in both normally grouped and isolated male Albino-Swiss mice, maintained on a 12:12 light-dark regimen (lights on 08.00--20.00). A significant diurnal rhythm in brain tryptophan level was evidenced only by grouped animals, in contrast to the relatively sustained and significantly lower tryptophan level found to exist in isolated mice. Significant, daily rhythms in food intake, for both isolated and grouped mice were found to exist, that were highly correlated in phase. Isolated mice, however, consistently demonstrated an increased food consumption throughout the 24-hour period. These results suggest that a possible alteration in tryptophan metabolism exists in isolated mice, as tryptophan, an essential amino acid, is obtained by mammals only through their diet.
Cyanine 863 inhibited the efflux of tetraethylammonium ion from slices of rat or rabbit renal cortex while the efflux of p-aminohippuric acid was unaffected. Dinitrophenol increased the efflux rate of both the cation and anion. Cyanine also decreased the dinitrophenol-enhanced efflux of tetraethylammonium (TEA).

Sigmoid volvulus was diagnosed in two patients with progressive system sclerosis. In one case, this was initially confused with a high fecal impaction. Both cases responded to early surgical intervention. Sigmoid volvulus should be considered in the differential diagnosis of intestinal obstruction associated with progressive systemic sclerosis.
A study was done to relate the in vivo reduction of nitrate to nitrate uptake, nitrate accumulation, and induction of nitrate reductase activity in intact barley seedlings (Hordeum vulgare L. var. ;Numar'). The characteristics of nitrate uptake in response to both time and ambient concentration of nitrate regulated reduction and accumulation. Uptake, accumulation, and in vivo reduction achieved steady state rates in 3 to 4 hours, whereas extractable (in vitro) nitrate reductase activity was still increasing at 12 hours. In vivo reduction of nitrate was better correlated exponentially than linearly over time with in vitro activity of nitrate reductase. A similar relationship occurred over increasing concentration of nitrate in the ambient solution. The results suggest that the rate of in vivo reduction of nitrate in barley seedlings may be regulated by the rate of uptake at the ambient concentrations of nitrate employed in the study.
Electrophysiological studies of verapamil on isolated cardiac tissue show its effects to occur during Phase 2 and Phase 4 of the cardiac action potential and to be a result of blocking the slow inward Ca2+ current. It is important to know if verapamil effects the transmembrane movement of ions other than Ca2+. By studying uptake and efflux of 42K by canine cardiac Purkinje fibres in the presence and absence of verapamil (1.1 x 10-6 M-4.4 X 10-6 M), it was found that the drug inhibits both 42K uptake (15%) and efflux (30%).
