Biodesulfurization.

Microbial sulfur-specific transformations have been identified that selectively desulfurize organic sulfur compounds in fossil fuels. Recent discoveries related to biodesulfurization mechanisms may lead to commercial applications of biodesulfurization through engineering recombinant strains for over-expression of biodesulfurization genes, removal of end product repression, and/or by combining relevant industrial and environmental traits with improvements in bioprocess design.

Biocatalytic sulfur removal from fuels: applicability for producing low sulfur gasoline.

Environmental regulations are driving R&D efforts to produce low sulfur fuels, including diesel fuel and gasoline for motor vehicles. Biocatalytic sulfur removal from fuels has potential applicability for producing low sulfur gasoline. Microbial biocatalysts have been identified that can biotransform sulfur compounds found in fuels, including ones that selectively remove sulfur from dibenzothiophene heterocyclic compounds. Most attention is give to the 4S pathway of Rhodococcus, which can remove sulfur from substituted and unsubstituted dibenzothiophenes, including sulfur compounds that hinder chemical catalysis and that resist removal by mild hydrotreatment. Various bioreactor and bioprocess designs are being tested for use with biocatalysts, including recombinant biocatalysts, for use in removing sulfur from fuels and feedstocks within the petroleum refinery stream. With bioprocess improvements that enhance biocatalyst stability, achieve faster kinetics, improve mass transfer limitations, temperature and solvent tolerance, as well as broaden substrate specificity to attack a greater range of heterocyclic compounds, biocatalysis may be a cost-effective approach to achieve the production of low sulfur gasoline. The challenge will be to accomplish these improvements by the time the regulations for low sulfur gasoline and other vehicle fuels go into effect in order to be competitive with emerging nonbiological desulfurization technologies.

Application of reverse transcriptase PCR for monitoring expression of the catabolic dmpN gene in a phenol-degrading sequencing batch reactor.

A modified freeze-thaw method in combination with reverse transcriptase PCR was developed for monitoring gene expression in activated sludge. The sensitivity of the methodology was determined by inoculating non-sterile activated sludge samples with 3-chlorobenzoate-degrading Pseudomonas putida PPO301(pRO103), which contains the catabolic tfdB gene. tfdB mRNA was detected in 10 mg of activated sludge inoculated with 10(4) CFU of the target organism. This technique was subsequently utilized to analyze the in situ expression of the catabolic dmpN gene in a sequencing batch reactor (SBR) bioaugmented with phenol-degrading P. putida ATCC 11172. Greatest dmpN expression was observed 15 min after maximum phenol concentration was reached in the reactor and 15 min after the start of aeration. Decreased phenol concentrations in the reactor corresponded to reduced levels of dmpN expression, although low levels of dmpN mRNA were observed throughout the SBR cycle. These results indicate that concentration of phenol in the reactor and the onset of aeration stimulated transcriptional activity of the dmpN gene. The information obtained from this study can be used to alter SBR operational strategies so as to lead to more effective bioaugmentation practices.

Genetic engineering of bacteria from managed and natural habitats.

The genetic modification of bacteria from natural and managed habitats will impact on the management of agricultural and environmental settings. Potential applications include crop production and protection, degradation or sequestration of environmental pollutants, extraction of metals from ores, industrial fermentations, and productions of enzymes, diagnostics, and chemicals. Applications of this technology will ultimately include the release of beneficial agents in the environment. If safely deployed, genetically modified bacteria should be able to provide significant benefits in the management of environmental systems and in the development of new environmental control processes.

A discrete mathematical model of unlabelled granulocyte kinetics. A preliminary study of feedback control.

The equations used in formulating the continuous model of granulocyte kinetics developed by O'Fallon et al. (1971) were analyzed to see if they could be altered to simulate a feedback mechanism operating on the production and development of granulocytes. After extensive study and modification of the continuous model, it was found that a discrete model based on a Leslie matrix procedure was more effective for simulating the feedback system. This discrete model was used to show experimentally, from a mathematical view point, that a feedback mechanism of some kind must be operating on the production and development of granulocytes. Further, the discrete model was subjected to preliminary tests (simultaneous and cascading feedback) to demonstrate that it has the capability of responding to feedback control.
