Sex steroid hormones levels and testicular activity during hepatocarcinogenesis by N-2-fluorenylacetamide.

During hepatocarcinogenesis by N-2-fluorenylacetamide, the hormonal status of male Sprague-Dawley rats is modified. The testes present a reduced activity which is evidenced by a decreased spermatogenetic activity accompanied by a drastic decrease of the plasma testosterone level. In contrast, plasma progesterone and estradiol-17 beta levels were not modified despite the 80 fold augmentation of alpha-fetoprotein, an estrogen binding protein synthesized during liver carcinogenesis.

[Carcino-embryonic antigen assay in cancer of the upper respiratory and digestive tracts].

Radioimmunological assay of carcino-embryonic antigen (CEA) was performed in 238 patients with cancer of the upper respiratory and digestive tracts after a minimum follow-up period of 3 years. Secretion of CEA was observed in only 1 out of 3 cases, levels, always being low and only slightly influenced by the localization or size of the tumor, and the condition of the lymph nodes. Initial levels are of no prognostic significance. The appearance of high levels during the course of the disease corresponds to shorter survival, but as it does not precede clinical evidence of recurrence or metastases this assay cannot play a role as a tumor marker.

On the synthesis of alpha-fetoprotein during liver carcinogenesis by N-2-fluorenylacetamide and its relationships with the intracellular localization of the carcinogen.

Adult female Sprague-Dawley rats fed with 0.06% N-2-fluorenylacetamide presented an augmentation of plasma alpha-fetoprotein 2 days after the beginning of the experiment. This very early synthesis of alpha-fetoprotein was concomitant with the penetration of N-2-fluorenylacetamide detected by fluorescence microscopy, in the cytoplasm of the hepatocytes.

Modifications in adrenal activity during hepatocarcinogenesis by N-2-fluorenylacetamide.

During chemical hepatocarcinogenesis by N-2-fluorenylacetamide, the hormonal status of female Sprague Dawley rats is largely modified. Adrenals present a modified activity which is evidenced by an enlargement of glomerulosa and reticularis zones and a decrease of the fasciculata, while medulla remain unchanged. The level of aldosterone remain quite normal in N-2-fluorenylacetamide treated rats despite changes observed in the glomerulosa. The cortisol level was lowered in agreement with the decreased size of the fasciculata. The estradiol level was normal despite the reduced activity of the ovaries. The possibility of a participation of the reticularis zone in the regulation of the estradiol level is suggested. The possible protective action of the adrenals during hepatocarcinogenesis is discussed.

Study of an enzyme immunoassay kit for carcinoembryonic antigen.

We evaluated a commercial enzyme immunoassay kit for carcinoembryonic antigen (CEA) based on a non-competitive "sandwich principle" method (Abbott Laboratories). The serum sample is treated with an acid buffer and the supernate is incubated with an anti-CEA coated polystyrene bead. After washing, the bead is incubated with an anti-CEA/peroxidase conjugate. After a second washing, the activity of the enzyme bound to the solid phase is assayed after addition of a chromogenic substrate. This activity is proportional to the concentration of CEA in the serum sample. The characteristics of the assay are: (a) good sensitivity (around 0.25 microgram/L) and (b) satisfactory reproducibility (CV < 11% within assay). There is little cross reactivity between CEA and molecules such as nonspecific cross-reacting antigens that are known for their high potential of cross reactivity. No nonspecific interference was encountered with anti-globulin factors. We compared results with this enzyme immunoassay kit with those by a radioimmunoassay provided by the Commissariat à l'Energie Atomique. The correlation (r) was 0.95 (p < 0.001). The distribution of CEA values obtained for 1020 normal subjects is given. We conclude that the kit provides a simple and reliable procedure.

Changes in ovarian activity during hepatocarcinogenesis by N-2-fluorenylacetamide: role of alpha-fetoprotein.

During chemical hepatocarcinogenesis by N-2-fluorenylacetamide, the hormonal status of female Sprague Dawley rats is largely modified. Ovaries present a reduced activity which is evidenced by a decreased number of follicles and corpora lutea. The level of progesterone in N-2-fluorenylacetamide treated rats is low and agrees with the lack of corpora lutea. Conversely, estradiol level remains normal. This fact may be explained by the presence of alpha-fetoprotein, a carcino-embryonic protein which binds specifically estrogenic hormones.

Specific cleavages of pure tRNAs by plumbous ions.

After renaturation some pure tRNAs were submitted to the action of lead acetate 1 . 10(-3) M at pH 7.3 and at 37 degrees C in the presence of either 1 M or 0.5 M NaCl. These tRNAs were specifically cleaved by Pb2+. The exact cleavage points were determined by analysing the oligonucleotides obtained from three yeast tRNAs. In 1 M NaCl, tRNA(Phe) is cleaved after the hUp17 and partially cleaved after Cp73. In 0.5 M NaCl, there are cleaveages after hUp16, hUp17 as well as a partial one after pGP1. In 1 M NaCl tRNA(Asp) is not cleaved, whereas in 0.5 M NaCl 50% of the molecules are cleaved in the anticodon region after Up35, 14% after hUp19 and 6% after hUp16. In 1 M NaCl tRNA(Val) is cleaved in the hU loop: 40% after hUp16 and 60% after CP17. The action of lead on five other pure tRNAs was studied on the analytical scale only, by polyacrylamide gel electrophoresis. They could be classified into two familites, one cleaved mainly in the hU loop, the other in the anticodon loop. The minimal concentrations of Pb2+ required for cleavage were determined for several tRNAs, the most sensitive of which, yeast tRNA(Val), being still cleaved with a concentration of 5 . 10(-6) M in 0.15 M NaCl. Although the cleavage often occurs after hUp, poly (hU) is less sensitive than poly(U). This and other results indicate that cleavages depend more on the conformation than the sequence of the polynucleotide chain, bends in the tertiary structure being lead-sensitive sites. Finally, the amino acid acceptance activities of cleaved tRNA(Phe) and tRNA(Asp) were determined.

[Bioclinical study of the electro-immunodiffusion technic for the assay of serum alpha fetoprotein].

The author discusses the result that may be expected in clinical medicine from an electro-immunodiffusion method for the detection of alpha-foetoprotein. The samples studied were obtained from pregnant women, patients with liver cell carcinoma or severe digestive diseases, whether malignant or not, and a few amniotic fluids and sera obtained from the umbilical cord. This simple, rapid method is characterized by a sensitivity threshold of about 300 ng/ml and permits one to detect about 75% of cases of liver cell carcinoma.
