The kinetics of IgG and IgM antibody-forming cells in ACI and F344 rats immunized with poly(glu52lys33tyr15).

The cellular kinetics of antibody production in high and low responder rats immunized with poly(Glu52Lys33Tyr15) or with poly(Glu52Lys33Tyr15)/MeBSA were characterized: serum antibody and IgG and IgM antibody-forming cells in the spleen and in selected lymph nodes were assayed in male and female rats following immunization by several routes. Aggregation of the antigen with MeBSA enabled the poorly responding F344 rats to produce antibody, which was almost exclusively IgG. High responder ACI rats, under the same conditions, produced antibody of both IgG AND IgM classes. These data suggest that in low responders one defect, possibly at the T-cell level, can be overcome by aggregation but that a second defect, involving the regulation of IgM production, still exists.

The avidity of IgM antibody in high and low responder rats.

This study examined IgM antibody produced by highly responding ACI and poorly responding F344 rats follwing immunization with poly(Glu52Lys33Tyr15) or poly(Glu52Lys33Tyr15) aggregated with methylated bovine serum albunim (MeBSA). The ACI rats produced both IgM and IgG plaque-forming cells (PFC) following immunization with either form of antigen. The F344 rats did not respond to unaggregated poly(Glu52Lys33Tyr15), but they produced significant amounts of IgG PFC and extremely small amounts of IgM PFC after immunization with poly(Glu52Lys33Tyr15)/MeBSA. Both high and low responder rats had similar kinetic profiles of IgM antibody production, and this antibody had nearly identical avidity in both strains with no evidence for any maturation in avidity. thus, one of the genetic defects in the antibody response to poly(Glu52Lys33Tyr15) is an inability of the F344 strain to produce large amounts of IgM in response to this antigen.

Effect of maternal immunization on the antibody response of low responder rats.

The offspring of low responder F344 female rats that were immunized with poly(Glu52Lys33Tyr15) aggregated with MeBSA prior to mating showed a higher antibody response to the polypeptide antigen than did the offspring of unimmunized females. Immunization of the mothers with unaggregated polypeptide, or with DNP-BGG, did not affect the antibody response of the offspring even when high doses of antigen were used. When the polypeptide used to immunize the mothers was given as an aggregate, some crossed the placenta to the fetus. The antigen was first detected in the placenta, blood and liver of the fetus at 15 days of gestational age. After birth, it was in the liver and spleen up to 6 weeks af age, and thereafter it was present only in the bone marrow.

Genetic studies in inbred rats. VI. Linkage relationships of mixed lymphocyte reactivity, serologically defined antigens (Ag-B, Ag-C) and the immune response to poly(Glu52Lys33Tyr15).

The Ag-B allotype, mixed lymphocyte reactivity (MLR) and the immune response to poly(Glu52Lys33Tyr15) were assayed in male rats from the F2 hybrid and two backcross generations of the F344 and DA strains in order to investigate the structure of the rat major histocompatibility complex. No disparity between Ag-B type and mixed lymphocyte reactivity was found in 263 animals. The immune response to poly(Glu52Lys33Tyr15) was closely linked to the Ag-B locus, and both antibody production and the delayed hypersensitivity response were under polygenic control. These results suggest that the genetic loci which determine these responses in the rat are closely linked and that recombinational events between the Ag-B and MLR loci are infrequent.

Influence of serum albumin on the fertilizing ability in vitro of rat spermatozoa.

Under defined conditions, in the presence of 10 mg/ml of bovine serum albumin, cauda epididymal rat spermatozoa displayed vigorous motility, and a high proportion (81%) of eggs were fertilized. In contrast, no fertilization was observed after omission of albumin, or replacement of the protein by 10 mg/ml of cytochrome c, beta-globulin, gamma-globulin, hemoglobin, lysozyme, and polyvinylpyrrolidone, and 5 mg/ml of ribonuclease. However, high motility occurred in suspensions containing 3 x 10(6) spermatozoa/0.1 ml of medium with cytochrome c, beta-globulin, or gamma-globulin. In medium with 1 mg/ml of ovalbumin, 7% (2/29) eggs were fertilized. Use of defatted albumin resulted in a higher rate of fertilization than unmodified albumin (87 vs 70%), and this difference approached statistical significance. No fertilization was obtained in the presence of albumin presaturated with cholesterol. These results suggest that: (a) rat sperm cells failed to capacitate in the absence of albumin; (b) the protein exerted more than a nonspecific macromolecular effect; and (c) lipids associated with albumin may modify its ability to promote sperm capacitation.

Inhibitory effect of synthetic phospholipid vesicles containing cholesterol on the fertilizing ability of rabbit spermatozoa.

Suspensions of dimyristoyl and dipalmitoyl phosphatidylcholine vesicles bearing 10 to 40 (w/w)% cholesterol inhibited the fertilizing ability of uterine-capacitated rabbit spermatozoa at concentrations of 1 to 10 mg of lipid/ml. Recovery of fertilizing ability by treated sperm cells was observed following insemination into the uterus 5 to 6 hr before ovulation. Vesicles lacking the sterol were not inhibitory under the conditions employed. Suspensions of cholesterol (0.4 to 4 mg of sterol/ml) with out phospholipid, in contrast, inhibited fertilization. Implication of cholesterol in sperm decapacitation by seminal plasma membrane vesicles is discussed in terms of these results.

Genetic control of the immune response to poly(Glu52Lys33Tyr15) in neonatally thymectomized high and low responder rats.

The immune response to poly (Glu52Lys33Tyr15) is under polygenic control and linked to the major histocompatibility complex of the rat. Aggregation of this antigen with methylated bovine serum albumin (MeBSA) eliminates the expression of genetic control by increasing the response of low responders and decreasing that of high responders. Humoral and cellular aspects of the immune response to both unaggregated and aggregated poly (Glu52Lys33Tyr15) were investigated in neonatally thymectomized high-responder ACI and low-responder F344 rats. T cells are necessary for responses to unaggregated poly (Glu52Lys33Tyr15) since thymectomy significantly decreased numbers of antibody-forming cells and serum antibody levels, and delayed hypersensitivity responses and antigen-induced in vitro proliferation. However, thymectomy had no significant effect on these parameters of immune responsiveness in either ACI or F344 rats immunized with poly (Glu52Lys33Tyr15)/MeBSA. Aggregation also increased IgG production and delayed hypersensitivity and antibody affinity in low responders.

Membranoproliferative glomerulonephritis. Localization of early components of complement in glomerular deposits.

A body of evidence suggests that in membranoproliferative glomerulonephritis (MPGN), complement is activated by the alternate pathway. Therefore, deposition of early components of complement should not be expected in glomeruli. The renal tissues of 16 patients--13 with classic MPGN and 3 with dense deposit disease, a variant of MPGN--were studied by light and electron microscopy and by means of elution and immunofluorescence for the localization of complement (C1q, C4, and C3), immunoglobulins (1gG, IgM, and 1gA), and other serum proteins. Variable amounts of C3, C4 and/or C1q, and IgM were detected in the glomeruli of all patients, whereas IgG and IgA were present, respectively, in 15 of 16 and 6 of 16 patients. Deposits were localized in mesangium and in peripheral capillary loops in a typical lobular distribution. The specificity of each antiserum was verified by immunodiffusion, immunoelectrophoresis, and blocking experiments utilizing unlabeled antibody. Glomerular-bound IgG was eluted with acid citrate buffer, suggesting that IgG might be complexed with antigen(s) in glomerular deposits. By light microscopy, lesions ranged from focal proliferation and lobulation to more severe involvement with typical splitting of glomerular basement membranes, sclerosis, and less frequently, crescent formation. Ultrastructurally, all patients with classic MPGN exhibited mesangial and subendothelial deposits, and in 5 of these patients, subepithelial deposits were demonstrated. With the exception of ultrastructural lesions, patients with the dense deposit variant lacked distinguishable features when compared with those with classic MPGN. The significance of these findings is discussed in relation to a) activation of complement and the possible role of an immune complex mechanism and b) the variability of the morphologic expression.

Reversible anti-fertility effect by non-occluding oviductal copper coils in the rabbit.

Placement of two 1 cm copper (Cu) coils around the mid-region of each oviduct in a group of five female rabbits with proven fertility, 34 days before mating, virtually prevented fetal implantation in these animals. The oviducts remained patent during a 47 day implantation period and following removal of the Cu coils, does that were remated became pregnant. In two of these does an implantation rate (living fetuses/corpora lutea) of 33% (7/21) was observed and one post-Cu implanted doe had a litter of two. Among four does bearing a single Cu coil on their left oviduct for 28 days prior to mating there was 19% (4/21) implantation of the left side and 56.3% (9/16) on the untreated right side, suggesting a local action by the metal. The results indicate that Cu is an effective anti-fertility agent when implanted around the oviducts and, as a corollary, it is apparent that the metal is not restricted to placement in the uterus in order to prevent pregnancy in rabbits. Non-occluding Cu containing devices may offer a new approach to reversible fertility control in the female that merits further investigation.

Diffusion of polymer gel implants.

Crosslinked polyacrylamide and polyvinylpyrrolidone gels have been used to subcutaneously implant (125)I-labeled immunoglobulin, (125)I-labeled luteinizing hormone, (125)I-labeled bovine serum albumin, (125)I-labeled insulin, [(3)H]prostaglandin F(2alpha), and Na(125)I into hamsters. From the rates of absorption of the solutes, their diffusion coefficients were determined. The diffusion coefficients showed a logarithmic dependence on implant polymer concentration and solute molecular weight. Release of the solutes from gel preparations incubated 10 mM phosphate buffer (pH 7.2) at 37 degrees revealed a similar relationship between solute diffusion coefficient, molecular weight, and the concentration of polymer. A general equation was derived that gives the expected diffusion coefficient of a substance in a polymer gel from its molecular weight, diffusion coefficient in solvent, and polymer concentration of the gel.
