Macrophage function in senescence.

Although the parenteral injection of dextran sulfate 500 (DS 500; 50 mg/kg body weight) into 22-month-old NMRI mice resulted in a complete loss of resistance when administered 1 day before the sublethal (4.5 X 10(3)) infection with Listeria monocytogenes, the consequences in aged animals were less dramatic than in young adult (2- to 3-month-old) controls. This was documented by prolonged survival times as well as reduced numbers of Listeriae in spleen. This indicates that aged mice possess an increased paghocytic capacity, as compared to young adult animals. In aged mice the DS 500-induced loss of resistance could be completely abolished by pretreatment with 3 X 10(9) heat-killed Bordetella pertussis organisms 4 days before the DS 500 injection, i.e., 5 days before listeric infection. This indicates that aged NMRI mice possess remarkable reserves in phagocytic activity.

[Testicular biopsy for diagnosis of fertility disorders (author's transl)].

The wide range of clinical possibilities of diagnosis of male fertility disorders (spermiogram, hormone determination, etc) raises the question whether there is any indication at all for testicular biopsy. For this reason, in the present survey, after presentation of the technical requirements and histological criteria of assessment, the most important pathomorphologically definable findings are described. By comparison of the results of studies of testicular biopsy and spermiogram the additional information obtained histologically is pointed out. it comprises diagnostic statements not only, but also suggestions on prognosis and therapy.

[Anterior resection and abdomino-perineal extirpation in patients with rectal cancer: a retrospective analysis (author's transl)].

Eighty-seven patients with an abdomino-perineal extirpation of the rectum and 75 patients with an anterior resection of the rectum could be analysed retrospectively. The mean observation period was 36 months for anterior resection and 38 months for abdomino-perineal extirpation. The rate of recurrences, mortality, and postoperative complications of each surgical method was correlated to tumor-localization, -staging, and -grading. The poor prognosis of patients with abdomino-perineal extirpation was mainly due to relatively more cases in advanced tumor stage. In rectal cancer above 7 cm anterior resection should be performed.

Biological activity of Bordetella pertussis in lipopolysaccharide-resistant mice.

Effects of Bordetella pertussis organisms, such as adjuvanticity, induction of hypersplenia, and leukocytosis as well as modification of nonspecific resistance to infection and typical morphological response of lymphatic organs, were studied in the lipopolysaccharide-resistant C3H/HeJ mouse strain. It was shown that B. pertussis exerted all of these effects in C3H/HeJ mice, although the morphological response, hypersplenia, and modification of resistance to infection with Listeria monocytogenes in such animals were less pronounced than those in lipopolysaccharide-sensitive mouse strains. This indicated that the biological activity of B. pertussis as determined in the present studies, is due partly to structural components other than lipopolysaccharide.

[Skin reactions to isolated bacterial cell wall components, bacterial peptidoglycan in particular].

To elucidate the pathogenetic mechanism of bacterial-allergical diseases and of reactions provoked by bacterial test substances (e.g. the intracutaneous test), it is necessary to analyse carefully various factors involved in these reactions. To try to separate toxic from immunological reactions minute amounts of peptidoglycan in four different preparations were applied to humans by intracutaneous injections. Peptidoglycan, an immunologically well defined wall components of almost all bacteria and which is therefore ubiquitous, was isolated from staphylococci and streptococci. The skin reactions were evaluated by macroscopical, microscopical and immunohistological observations, as well as by electromicroscopy and statistical means. Several findings indicate the involvement of immunological factors in the observed skin reactions. In some cases no measurable reaction was seen, whereas in others typical skin reactions occurred. Similar reactions were produced by the antigenically related peptidoglycans isolated from Staphylococcus aureus and Staphylococcus epidermis. These reactions differed, however, from those elicited by peptidoglycan preparations from Streptococcus pyogenes type A. Patients with chronic bacterial infections and who were injected by four bacterial preparations demonstrated a significant increase in delayed hypersensitivity reaction compared with two other groups of patients (atopic patients without chronic infection). Because of the very low test dose, necrotic skin reactions, which occur in animal experiments, were not observed.

[The role of the complement system in experimental pancreatitis (author's translation)].

Two different experimental procedures were used to induce pancreatitis in rats: distal ligature of the common bile duct and the Pfeffer method in modification of Nevalainen. It could be shown that consumption of complement can be demonstrated even in pancreatitis induced by other than immunological models. Further examination in decomplemented rats showed the possibility of inducing acute pancreatitis by the above-mentioned models, although of less intensity. Therefore, the complement system will be an important factor for the progress and intensity of the inflammation process.

[Intraluminal hypertension as a pathogenetic factor of acute pancreatitis in the rat (author's transl)].

By the use of various experimental procedures with rats, the significance of intraluminal hypertension, caused by ductal obstruction, reflux of bile and duodenal reflux were considered with regard to their capacity for producing pancreatitis. The extent of the histological changes in the pancreas after 48 hours as well as the period of survival in the individual experimental groups served as the parameters for the comparison. The results show that the rise in intraluminal pressure is alone enough to produce acute pancreatitis. It follows from this that rise of pressure in the duct system caused by ductal obstruction may be the triggering factor in the induction of pancreatitis. Reflux of bile, bacterial colonisation of the pancreas by duodenal reflux as well as activation of proteolytic enzymes were not necessary a prior factors but ones which have an aggravating effect in the development of the disease.

[Significance of the complement system in acute pancreatitis in rats].

The influence of complement system in acute pancreatitis in the rat was examined. Two different experimental procedures were used to induce pancreatitis: distal choledochal ligature and the Pfeffer model as modified by NEVALAINEN. In both procedures an acute pancreatitis developed and complement depression was noted. These results showed that consumption of complement will occur even in pancreatitis induced by other than immunologic models. In a second series the same experimental procedures were used in rats that had been decomplemented by application of cobra venom factor. It was possible to produce an acute pancreatitis of moderate severity.
