Flavonoids, a class of natural products of high pharmacological potency.

A review has been presented of the biochemistry and pharmacology of a class of natural products, the flavonoids. These substances which are widely distributed in the plant kingdom and present in considerable quantities in common food products, spices and beverages have in a concentrated form (Propolis) been used since ancient times by physicians and laymen to treat a great variety of human diseases but they have yet to pass the tests of modern, controlled, clinical experimentation. An attempt has been made to present the fundamental evidence from the basic biological sciences which is required to stimulate the interest of the clinicians in this new field. The few existing reports on the careful pharmacodynamic, pharmacokinetic and clinical studies which have been made have been summarized to provide a basis for a full-scale investigation of the therapeutic potential of flavonoids.

Biochemical analysis of normal human tissues and those of lymphoma patients by high-resolution protein mapping.

The technique of high-resolution, two-dimensional electrophoresis was applied to crude membrane protein fractions of various normal human tissue samples, and those, mainly spleens and lymph nodes, of patients with lymphomas of different types. The obtained protein maps show a high reproducibility, tissue specificity, and sensitivity to specific oncologic changes. The relevance of the latter aspect to tumor characterization is illustrated. Quantitatively, minor proteins were detected by a sensitive silver stain. The feasibility of objective, computerized quantification and evaluation of the protein maps is described.

Molecular weight analysis of Fc gamma-binding proteins of lymphoid leukemia, myeloid leukemia, and hairy-cell leukemia.

The molecular weights of EDTA-mercaptoethanol-soluble Fc gamma-affined proteins isolated from chronic lymphocytic leukemia of the B type, prolymphocytic leukemia of the B type, chronic myeloid leukemia and hairy-cell leukemia were compared. SDS polyacrylamide gel electrophoresis of the Fc gamma-binding material obtained from all six cases of B type leukemia revealed a single peak with an apparent molecular weight of 28,000. The Fc gamma-affined material isolated from the cells of two cases of chronic myeloid leukemia showed two peaks, one with an apparent molecular weight of 42,600 and one with an apparent molecular weight of 18,800. The Fc gamma-affined material isolated from the cells of two cases of hairy-cell leukemia electrophoresed in the form of a closely spaced double peak. One component of the double peak had an apparent molecular weight of 28,000 and thus corresponds to the Fc gamma-binding material of leukemic B cells. The second component had a slightly lower molecular weight. The latter component is not present on either leukemic B cells or myeloid cells. The results indicate that the EDTA-mercaptoethanol-soluble Fc gamma-affined proteins of different types of cells differ in molecular weight, and thus in molecular structure.

Spontaneous spatio-temporal organization in yeast cell suspension.

Under suitable conditions of substrate supply, cell suspensions of yeast Saccharomyces uvarum may display synchronized oscillations of glycolytic metabolism. Also in an unstirred suspension, spatial patterns can be induced by continuously feeding enzymically liberated glucose into the system. the absorption of NADH in the suspension was measured in an annular cuvette. The observed structures display moving waves, which travel over a distance of some centimetres in the cell suspension and seem to originate from a volume element in which the phase is ahead of those of its neighbours. The wavelength is estimated to be abot 10 cm and the velocity of spreading of the wave is about 5 cm/min. Waves are only observed in the beginning of glycolytic oscillations. Subsequently the spatial synchrony usually increases and destroys the previous organization. Similar spatio-temporal structures are observed during aggregation of the slime mould Dictyostelium discoideum and in the chemical reaction system of Belousov and Zhabotinsky.

Ethanolaminosis. A newly recognized, generalized storage disease with cardiomegaly, cerebral dysfunction and early death.

A storage disease with cardiomegaly, generalized muscular hypotonia, cerebral dysfunction, failure to thrive and early death is described in two siblings. The first one died at the age of 10 months, the second at the age of 17 months. The symptoms were mainly due to lysosomal storage of a substance which had a positive reaction to PAS and Best's stain and which was resistant to diastase. This substance was stored in nearly all the organs, especially in the heart, liver, spleen and less in the brain and skeletal muscles. An increased renal excretion of ethanolamine, a greatly increased hepatic concentration of ethanolamine and diminished hepatic ethanolamine kinase activity could be demonstrated. Ethanolamine is essential for the synthesis of phospholipids. Both parents showed increased renal excretion of taurine. In several aspects, this syndrome is similar to the glycogenosis type II described by Pompe.

Microheterogeneity of Immunoglobulin G from plasmocytomas. Identification of two types of IgG by isoelectric focusing.

Isoelectric focusing of serum immunoglobulin G (IgG) revealed that the microheterogeneity, which is expressed in the isoelectric points (IEP), partly is caused by differences in the content of N-acetyl-neuraminic acid (NANA), partly by other effects, probably including deamidation. Two different types of Plasmocytoma-IgG, which differ in IEF-pattern, i.e. population distribution, in sensitivity towards neuraminidase and in carbohydrate content, are described. The contents of carbohydrates in these 2 IgG-types are up to 30% greater and 50% less, respectively, than that of normal IgG. A precise correlation was found between the total content of NANA in moles per mole IgG-monomer and the shift in band-IEP on hydrolysis catalyzed by neuraminidase. This may be used for a rapid estimate of NANA in IgG. The results do not permit a discrimination between an anabolic and a catabolic origin of the heterogeneity in the carbohydrate moiety of IgG.
