Substance P enhances cation permeability of neuronal cell lines.

Substance P stimulated the uptake of guanidinium in neuroblastoma X glioma hybrid cells and neuroblastoma cells but not in polyploid glioma cells. Guanidinium has previously been shown to pass the action potential Na+ channel in the two neuronal cell lines. Half-maximal stimulation was reached at 3 microM substance P and, with the hybrid cells, a saturation was seen above 10 microM. The analogue (D-Pro2,D-Trp7,9)-substance P, recently described as a substance P antagonist, caused a stimulation of guanidinium uptake comparable to that seen in the presence of substance P and did not inhibit the stimulation exerted by substance P. The pharmacological properties of the substance P-activated ion channel were investigated. Tubocurarine, phentolamine and propranolol blocked the substance P-stimulated guanidinium uptake with half-maximal inhibitory concentrations of 0.5, 5 and 50 microM. A similar characteristics has been found previously with the veratridine-activated Na+ channel in the cell lines investigated here. Peptides structurally related to substance P such as physalaemin and eledoisin, or others such as neurotensin, bradykinin, D-Ala2, Met5-enkephalinamide and ACTH(1-24) did not affect guanidinium uptake. In view of the high concentrations of substance P required for eliciting an effect in the cell lines, the involvement of specific receptors is questioned. A direct interaction of the peptide with the action potential Na+ channel is discussed.

Neuroblastoma X glioma hybrid cells synthesize enkephalin-like opioid peptides.

Partially purified extracts from neuroblastoma X glioma hybrid cells 108CC15 inhibit, like opioids, the prostaglandin E1-evoked formation of cyclic AMP in a dose-dependent manner in the same hybrid cells. The inhibition is prevented by the opioid antagonist naloxone. In addition, the same extract competes with [3H]naloxone and [3H]Leu-enkephalin for binding to opioid receptors of hybrid cell membranes and to a specific antiserum, respectively. The opioid activity in the extracts is destroyed by carboxypeptidase A and leucine aminopeptidase, but not by trypsin. Further purification of the extracts by HPLC, TLC, or high-voltage paper electrophoresis reveals in each case two active fractions which behave like Met- and Leu-enkephalin. The Met-enkephalin-like, but not the Leu-enkephalin-like, fraction is inactivated by treatment with BrCN. Dimethylaminonaphtylsulfonyl (dansyl) derivatives of Met- and Leu-enkephalin correspond to [3H]dansyl derivatives of Met-like substances from hybrid cells. Three to four times as much Met-enkephalin-like as Leu-enkephalin-like material is present in the extract. The overall concentration of opioid peptides in the hybrid cells varies between 0.03 and 1.0 pmol Leu-enkephalin equivalents per mg protein. The amount of opioids in the hybrid cells is strongly dependent on the cell density. The findings suggest that neuroblastoma X glioma hybrid cells contain opioid peptides that are very similar, if not identical, to Met- and Leu-enkephalin. Opioid activity can also be detected in other neuronal cell lines and even in glioma cells.

Pharmacological and electrophysiological characterization of lithium ion flux through the action potential sodium channel in neuroblastoma X glioma hybrid cells.

Interaction of Li+ with the voltage-dependent Na+ channel has been analyzed in neuroblastoma X glioma hybrid cells. The cells were able to generate action potentials in media containing Li+ instead of Na+. The uptake of Li+ into the hybrid cells was investigated for the pharmacological analysis of Li+ permeation through voltage-dependent Na+ channels. Veratridine and aconitine increased the uptake of Li+ to the same degree (EC50 30 microM). This increase was blocked by tetrodotoxin (IC50 20 nM). Veratridine and aconitine did not act synergistically; however, the veratridine-stimulated influx was further enhanced by the toxin of the scorpion Leiurus quinquestriatus (EC50 0.06 micrograms/ml). This stimulation was also blocked by tetrodotoxin. Thus, the voltage-dependent Na+ channel of the hybrid cells accepts both Li+ and Na+ in a similar manner.

Production and regulation of enkephalin-like peptides in neuroblastoma x glioma cells.

Partially purified extracts from neuroblastoma x glioma hybrid cells inhibit via opioid receptors the PGE1-elicited formation of cyclic AMP in the same hybrid cell system. The purification of extracts reveals two active fractions very similar to Met- and Leu-enkephalin by several criteria including treatment with cyanogen bromide. On an average, the intracellular concentration of opioids in hybrid cells is 0.1 pmol per mg protein. The concentration is strongly dependent on the cell density. Furthermore, the content in the hybrids of enkephalin-like peptides is specifically elevated by glucocorticoids.

Opioids, noradrenaline and GTP analogs inhibit cholera toxin activated adenylate cyclase in neuroblastoma x glioma hybrid cells.

D-Ala2-Met5-enkephalin, morphine, and noradrenaline inhibit the adenylate cyclase in homogenates of neuroblastoma x glioma hybrid cells in a dose-dependent manner even after the enzyme has been preactivated by cholera toxin. Half-maximal inhibition and extent of inhibition are the same with native or cholera toxin-activated enzyme. The inhibition caused by opioids or noradrenaline are antagonized by naloxone or phentolamine, respectively. The effect of D-Ala2-Met5-enkephalin on cholera toxin-activated enzyme is immediate in onset and rapidly reversed by the addition of naloxone. Guanyl-5'-yl-imidodiphosphate stimulates basal activity but inhibits the enzyme activated by cholera toxin or prostaglandin E1. Stimulation occurs at a concentration of 100 microM or above, inhibition even at 0.1 microM. The inhibitory effect of the non-hydrolysable GTP analog is antagonized by GTP. Guanyl-5'-yl-methylenediphosphonate, another nonhydrolysable GTP analog, inhibits basal as well as cholera toxin-stimulated or prostaglandin E1-stimulated adenylate cyclase. Other guanine derivatives such as GDP, GMP, cyclic GMP, guanyl-5'-yl-phosphoric acid amide and guanosine have no effect under the same conditions. The results may be taken as a piece of evidence for two separate guanyl nucleotide-binding sites accompanying the adenylate cyclase in the hybrid cells and mediating, respectively, stimulation and inhibition of the enzyme by hormones.

Na+-dependent uptake and release of taurine by neuroblastoma x glioma hybrid cells.

In neuroblastoma x glioma hybrid cells, a cell line of neuronal character, a saturable uptake system for taurine is found which displays high affinity and high capacity (km = 38 micro M, V = 1.25 nmol . mg-1 . min-1). Only the closely related structural analogues hypotaurine and beta-alanine are able to inhibit the transport of radioactively labeled taurine. Imipramine or haloperidol at 100 micro M effectively blocks taurine uptake. High-affinity taurine uptake shows an absolute and highly specific requirement for Na+. The hybrid cells internalize taurine very slowly and, with 1 mM extracellular taurine, attain a plateau only after more than 20 h, at which time approximately 34 mM labeled taurine has accumulated in the cytosol. Generally there is hardly any spontaneous release of accumulated taurine. Efflux can, however, be induced by increasing the intracellular Na+ content and is then accelerated by lowering the extracellular Na+ concentration. The hypothesis is forwarded that taurine may exert its function by driving the extrusion of Na+ in emergency situations.

Cellular localization and regulation of glutamine synthetase in primary cultures of brain cells from newborn mice.

The cellular distribution of glutamine synthetase was determined by indirect immunofluorescence in cultures of dissociated brain cells from newborn mice. The enzyme could be detected in about 40% of all cells, among which cells with astrocytic morphology were clearly identified. Treatment with the glucocorticoid dexamethasone led to a strong increase in the number of positivity stained cells. Enzyme induction by dexamethasone was maximal after 36 h and at a concentration of 0.1 micrometer. Under these conditions glutamine synthetase specific activity was elevated about six fold. Steroid hormones other than corticosteroids had no effects. The basal activity in these cultures was near that found in brains of newborn mice, but far below the activity in adult brains, showing that in culture the normal development of these cells is disturbed. A comparison of glial and neuronal cell lines showed that glutamine synthetase is present in both types of cell lines at a very low specific activity. Inducibility of this enzyme by dexamethasone was found in glial but not in neuronal cell lines.

Dermorphins, opioid peptides from amphibian skin, act on opioid receptors of mouse neuroblastoma x rat glioma hybrid cells.

Dermorphin and its Hyp6 analogue are opiate-like heptapeptides originally discovered in frog skin and characterized by the presence of a D-Ala2 residue in their sequence. They were assayed for their capacity to compete with [3H]Leu-enkephalin for binding to opioid receptors in membranes of neuroblastoma x glioma hybrid cells. In the presence of 7 nM-[3H]Leu-enkephalin, the concentrations at which they caused 50% inhibition of [3H]enkephalin binding (IC50 values) are 0.1 micro M and 0.3 micro M, respectively. In contrast, the synthetic L-Ala2-dermorphin shows very low affinity for the opioid receptors. In addition, like other opioid peptides, dermorphin and hyp6-dermorphin inhibit the elevation by prostaglandin E1 (PGE1) of the level of adenosine 3':5'-cyclic monophosphate (cyclic AMP) (IC50 values 0.2 micro M and 0.4 micro M, respectively). The inhibition is prevented by the opiate antagonist naloxone, L-Ala2-dermorphin is at least three orders of magnitude less potent in inhibiting the PGE1-evoked increase in the level of cyclic AMP. The results show that peptides with an amino acid sequence quite different from that of the enkephalins can bind to opioid receptors of the hybrid cells.

Regulation by secretin, vasoactive intestinal peptide, and somatostatin of cyclic AMP accumulation in cultured brain cells.

Secretin stimulates the accumulation of cyclic AMP (half maximally stimulating concentration: 10-20 nM) in cultured mouse brain cells mainly consisting of glioblasts. Vasoactive intestinal peptide (VIP) is much less potent in raising the level of cyclic AMP in these cultures. The effect of secretin but not that of VIP is inhibited by secretin-(5-27), a synthetic antagonist of secretin. Stimulation of the adrenergic alpha-receptors and the adenosine A1-receptors present on the cells attenuates the increase in cyclic AMP evoked by secretin and VIP. Somatostatin at low concentrations inhibits the accumulation of cyclic AMP (half-maximally inhibitory concentration: 3 nM), in the absence or presence of secretin, VIP, or isoproterenol. The results suggest that secretin might regulate the concentration of cyclic AMP in brain and provoke the question of a possible involvement of glial cells in the action of peptide hormones in the brain.

Uptake and energy-dependent extrusion of calcium in neural cells in culture.

The metabolism of Ca2+ was studied in a neuronal model system, the clonal mouse neuroblastoma x rat glioma hybrid cell line 108CC5. 1. Homogenates of the hybrid cells exhibit a specific activity of Ca2+-ATPase considerably higher than that of homogenates of the parental cells. 2. Uptake and release of 45Ca2+ by the hybrid cells display two and three distinct phases, respectively, and indicate that 40--50% of the cell-associated Ca2+ is located at the cell surface. 3. The influx of 45Ca2+ is insignificantly affected by Mg2+ or Na+, slightly diminished by Ba2+ or Sr2+, strongly inhibited by La3+, Co2+ or prenylamine, and considerably enhanced by high (i.e., depolarizing) concentrations of K+. The efflux of 45Ca2+ is reduced by La3+. 4. The hybrid cells tend to maintain Ca2+ homeostasis with an overall cellular Ca2+ concentration of 0.5--0.7 mM. At 1.8 mM Ca2+ in the medium this implies the necessity of an extrusion pump in the plasma membrane. 5. A reduction in the hybrid cells of the level of ATP is paralleled by a decline in the content of Ca2+. This can only be explained by the existence of energy-dependent intracellular Ca2+ stores that effectively compete for Ca2+ with a Ca2+ pump located in the plasma membrane. The internal stores are not identical with the mitochondria because mitochondrial inhibitors hardly change Ca2+ metabolism. 6. Micromolar concentrations of the ionophore A23187 can switch off the internal Ca2+ stores without affecting considerably the influx of Ca2+ through the plasma membrane. 7. With switched-off Ca2+ stores it is possible to increase the cellular Ca2+ content distinctly and to bring it back again to the control values in an ATP-dependent manner, i.e. to demonstrate the action of a Ca2+-extrusion pump in the plasma membrane. 8. Under some conditions active extrusion of Ca2+ depends not only on ATP but also on the presence of extracellular Na+. 9. Similar results as with hybrid cells are also obtained with rat glioma cells. The methodology of combining energy deprivation with the application of the ionophore A23187 is possibly generally applicable to obtain insight into the Ca2+ metabolism of various cell types.

Clonal hybrid cell lines expressing cholinergic and adrenergic properties.

Different cholinergic cell lines were fused with an adrenergic neuroblastoma cell line (N115-BU-8). Its fusion with a cholinergic neuroblastoma-glioma hybrid produced a "hybrid-hybrid" line containing cholinergic and adrenergic enzyme activities. Both activities were also present in subclones of this line. The presence of catecholamines in single cells was confirmed by microspectrofluorimetry. These results are discussed with respect to the possibility of a simultaneous synthesis of noradrenaline and acetylcholine in single cells. The cholinergic and adrenergic enzyme activities are influenced by cell density, by dexamethasone, and by conditioned medium.
