The mechanism of superoxide anion generation by the interaction of phenylhydrazine with hemoglobin.

The mechanism by which superoxide anion is generated by the interaction of phenylhydrazine with either oxy- or methemoglobin was investigated. Rather than superoxide anion generation resulting from an accelerated autooxidation of oxyhemoglobin, it was found that both oxy- and methemoglobin function as peroxidases toward phenylhydrazine with the resultant oxidation of this compound to phenyldiazine. Generation of phenyldiazine from the oxidation of phenylhydrazine by hemoglobin or by the hydrolysis and subsequent decarboxylation of methyl phenylazoformate (C6H5N=NCOOCH3) resulted in the production of superoxide anion. It is suggested that under certain conditions hemoglobin may function as a drug-metabolizing peroxidase.

Left ventricular function in chronic renal failure.

Left ventricular function was studied in 14 patients with end-stage chronic renal failure using non-invasive methods (echocardiography and systolic time intervals). Patients were divided into 3 groups. Group 1 consisted of 5 patients who were normotensive at the time of study and group 2 of 7 patients who were hypertensive when studied. Group 3 consisted of 2 patients: one was receiving propranolol and the other, studied 302 days after renal transplantation, was receiving digitalis for recurrent episodes of cardiac failure. All except the patient receiving propranolol had normal left ventricular function in systole with normal measurements of fractional fibre shortening (% delta S, EF) and normal measurements relating to the velocity of ventricular contraction (mean Vcf, mean velocity of posterior wall motion). Stroke volume and cardiac output were normal in some patients but were increased in patients with fluid overload. Early diastolic compliance of the left ventricle seemed to be normal except in the patient with recurrent cardiac failure. The study provided no evidence for the existence of a specific uraemic cardiomyopathy.

Radioimmunoassay of procollagen in serum of patients with Paget's disease of bone.

A new radioimmunoassay for human procollagen showed that the sera of 46 of 50 untreated patients with Paget's disease of bone contained increased concentrations of procollagen protein as compared to normal adults. After therapy with disodium etidronate, all the elevated serum procollagen concentrations decreased significantly, falling to normal levels in 33 of 40 patients. The procollagen levels before and after treatment were coordinate with the values for urinary total hydroxyproline and serum alkaline phosphatase activity. The data show that the radioimmunoassay for procollagen is a dependable and useful adjunct to the study of Paget's disease of bone.

Procollagen peptidase: its mode of action on the native substrate.

Procollagen peptidase was recovered from the medium of human and mouse fibroblast cultures by precipitation with ammonium sulfate. The test substrate for the in vitro enzymatic reaction was radioactively-labeled, disulfide-linked procollagen prepared from the medium of human fibroblast cultures. The enzymatic digests were analyzed by electrophoresis in polyacrylamide gets containing sodium dodecyl sulfate and urea. The human and mouse enzymes reacted with the substrate to generate the same intermediates and final products. Procollagen peptidase acts as an endopeptidase which cleaves each of the three procollagen chains in turn. The final products of the reaction are collagen and a three-chain, disulfide-linked fragment derived from the nonhelical aminoterminal residues of procollagen.

Significance of exercise-induced second degree atrioventricular block.

A case of second degree, type II, atrioventricular block precipitated by exercise is presented. It is suggested that exercise and other vagolytic manoeuvres may be used as a means of differentiating type I from type II atrioventricular block: the conduction disturbance of type I improves with exercise, whereas that of type II deteriorates with exercise.

Radioimmunoassay for human procollagen.

Rabbit antiserums were produced against the procollagen molecule secreted into the medium of cultured human skin fibroblasts. The isolated antigenic, amino terminal portion of the procollagen molecule was purified, labeled with iodine-125, and used in a radioimmunoassay which detected nanogram quantities of the same antigen. With the assay, immunologically identical molecules were detected in the culture mediumn of different strains of human fibroblasts and in normal human serums. Serumns from human cord blood contained a 12-fold higher concentration of the antigen than serums from adults, while serums other vertebrates gave reactions to incomplete cross-reactivity or non-reactivity.

Susceptibility of an insect Leptomonas and Crithidia fasciculata to several established antitrypanosomatid agents.

Growth inhibition of the lower trypanosomatids Crithidia fasciculata and a Leptomonas from a hemipteron by several established trypanocides and leishmanicides were compared in four complex and one defined media. The Leptomonas was more susceptible than C. fasciculata in all media, especially to phenanthridines (ethidium, prothidium, isometamidium) and diamidines (pentamidine, diminazene diaceturate [Berenil], hydroxystilbamidine, stilbamidine); concentrations of these drugs required for 50% inhibition of the Leptomonas were <5 mug/ml. In contrast, C. fasciculata was uninhibited by <20 mug of diamidines per ml and was three- to sixfold less susceptible than the Leptomonas to isometamidium and prothidium. Both trypanosomatids were susceptible to nucleoside antibiotics, e.g., nucleocidin. Neither was inhibited by suramin, melarsen, melarsen oxide, or tryparsamide. The Leptomonas was more susceptible to standard trypanocides than five other insect trypanosomatids in a complex medium; it was the only one inhibited by <20 mug of stilbamidine and hydroxystilbamidine per ml.
