[The health of adolescents: what do parents think about it?].

Parental care of adolescent children was studied in 501 families who attended the Centre of Preventive Medicine of Vandoeuvre-lès-Nancy. The health of their adolescent children was the parents' main concern and they considered morale, hygiene and alimentation as being the most important health determinants. Twenty-eight per cent of the parents indicated that they were anxious about their adolescent children. When they contacted the Centre of Preventive Medicine, parents were mostly seeking a regular check-up and information for their offspring. A correspondence analysis showed a breakdown of the parents into several groups, according to their responses and degree of concern for the health of their adolescents.

Vascular postsynaptic effects of some 5-HT1-like receptor agonists in the pithed rat.

5-HT induced an increase in blood pressure in the pithed rat which was antagonized by LY 53857 a selective 5-HT2 receptor antagonist. It was not antagonized by spiroxatrine, MDL 72222, idazoxan or AR-C 239, respectively 5-HT1-like and 5-HT3 receptor antagonists, alpha 2- and alpha 1-adrenoceptor antagonists. 5-MeODMT also induced an increase in blood pressure which was antagonized by LY 53857 but not by the other 5-HT receptor antagonists and alpha-adrenoceptor antagonists used, suggesting a 5-HT2 component in the pressor effect of 5-MeODMT. The maximal effect of 5-MeODMT was less marked than that of 5-HT. 8-OH-DPAT, RU 24969 and TFMPP were far less effective than 5-HT and 5-MeODMT to increase blood pressure. In contrast, 5-CT induced a vasodepressor effect. It is therefore suggested that the vasoconstriction induced by 5-HT and by 5-MeODMT in pithed rats could be due mainly to the selective stimulation of postjunctional 5-HT2 receptors because selective alpha 1- and alpha 2-adrenoceptor antagonists were ineffective against the vasoconstrictor effects of 5-HT and 5-MeODMT. The relative lack of effect of 8-OH-DPAT, RU 24969 and TFMPP to increase blood pressure suggested that postjunctional 5-HT1-like receptors play only a minor role - if any - in 5-HT induced vasoconstriction in the pithed rat.

Comparison of effects of some 5-HT1 agonists on blood pressure and heart rate of normotensive anaesthetized rats.

The present experiments served to compare the effects of the 3 5-HT1 agonists, 8-OH-DPAT, 5-MeODMT and TFMPP on the blood pressure and heart rate of normotensive anaesthetized rats. All the agonists induced, after i.v. injection, a decrease in blood pressure and heart rate. The hypotensive effects of 5-MeODMT and TFMPP were preceded by an increase, suppressed by both ketanserin and methysergide. The decrease in blood pressure induced by 5-MeODMT and 8-OH-DPAT was not antagonized by ketanserin, cocaine (and methysergide for 8-OH-DPAT) but was antagonized by methysergide (for 5-MeODMT) and spiroxatrine (for both). Bradycardia was not susceptible to ketanserin and cocaine (for 5-MeODMT) or to ketanserin and methysergide (for 8-OH-DPAT) but to methysergide and spiroxatrine (for 5-MeODMT) and cocaine and spiroxatrine (for 8-OH-DPAT). These results suggested that the hypotension and bradycardia induced by 5-MeODMT and 8-OH-DPAT are due to the stimulation of '5-HT1-like' receptors and probably to the 5-HT1A subtype; the 5-MeODMT-induced hypertension being ascribed to the stimulation of 5-HT2 receptors.

Radioimmunoassay for osteonectin. Concentrations in bone, nonmineralized tissues, and blood.

We describe a double antibody radioimmunoassay for the bone protein osteonectin using a polyclonal antibody raised against bovine fetal bone osteonectin. This assay, which cross-reacts with human osteonectin, was used to explore the tissue distribution of the protein. Osteonectin was detected in extracts of nonmineralized tissues from bovine fetuses, with concentrations ranging from 10 ng/100 micrograms of proteins in the lens to about 80 ng/100 micrograms in collagen-rich tissues (e.g., tendon), which represents respectively from 0.01% to 0.5% of the concentrations found in bone extracts. Osteonectin is thus a widespread component of extracellular matrix, although its concentration is minute in tissues other than bone and dentin. Adult human serum contains 200 ng/ml of osteonectin and values obtained in plasma prepared without platelet activation are about one-third of those in matched sera. This platelet-independent fraction might be used as a marker for bone metabolism.

Antidiuretic factor from the nervous corpora cardiaca of the migratory locust: improvement of an existing in vitro bioassay.

Nervous lobes of the migratory locust corpora cardiaca contain an antidiuretic (AD) factor which increases the fluid reabsorption of isolated recta in a dose-dependent manner. Several extraction procedures were developed and compared. The very satisfactory thermostability of the AD factor was demonstrated. By using a succession of different incubating media on the same preparation, we improved our bioassay. The developed variant is faster and more sensitive than the existing test and saves time and biological material.

Variations of phylloquinone concentration in human milk at various stages of lactation and in cow's milk at various seasons.

An improved high-performance liquid chromatographic method was used to measure phylloquinone concentrations in human and cow's milk. Concentrations in human milk obtained serially from 10 mothers 3, 8, and 21 days after delivery were found to increase during the course of lactation. No correlation was found between phylloquinone concentration and lipids but a significant negative correlation was observed between phylloquinone and cholesterol concentrations. Determinations in cow's milk showed increased levels during the summer months. Also, levels in cow's milk were significantly higher than in human milk. Notwithstanding marked individual variations in human milk, phylloquinone concentrations in cow's milk always proved higher than in human milk. Results prompt the necessity of protecting breast-fed infants by applying relevant prophylactics measures.

Vitamin K1 binding protein in milk.

Cow's milk has been shown to contain a protein complex which is able to bind vitamin K1 in a reversible manner. This binding property has been investigated by the celite method which consists in creating a dynamic equilibrium between the adsorbent, the celite and the protein complex for the ligand (vitamin K1). Based on competition experiment, the binding is specific and the vitamin K1 binding protein complex has a molecular weight equal to or higher than 7.5 X 10(2) KD.

Pharmacological properties of the enantiomers of idazoxan: possible separation between their alpha-adrenoceptor blocking effects.

The alpha-adrenoceptor blocking properties of the two enantiomers of idazoxan have been investigated in rats, dogs and chicks, as well as their agonistic effects in pithed rats. At peripheral sites, (+) idazoxan was equipotent for blocking both postsynaptic alpha-1 and alpha-2 adrenoceptors of the rat and revealed to be a potent antagonist at presynaptic sites of rats and dogs. (-) Idazoxan revealed to be selective for postsynaptic alpha-2 adrenoceptors with an apparent selectivity ratio of about 10. This selectivity of (-) idazoxan was greater in vitro. (-) Idazoxan also antagonized presynaptic alpha-2 adrenoceptors of rats and dogs. At central sites, (+) and (-) idazoxan antagonized the hypotension, bradycardia, inhibition of sympathetic nerve activity induced by clonidine in rats and dogs and sedation induced by clonidine and azepexole in chicks. Although (+) idazoxan was more potent than (-) idazoxan, binding studies revealed (-) idazoxan to be more selective than (+) idazoxan at central sites. It is concluded that (+) idazoxan antagonizes both alpha-1 and alpha-2 adrenoceptors and (-) idazoxan is selective for alpha-2 adrenoceptors. In the pithed rat, only (-) idazoxan possesses both alpha-1 and alpha-2 agonistic effects. These results show little differences between the two enantiomers of idazoxan as for those of imidazoline derivatives.

[Action of idazoxan on tachycardia following electric stimulation of the thoracic sympathetic chain in the rat].

Selective stimulation of sympathetic nerves of the pithed rat (instead of stimulation of the whole sympathetic outflow) induced a frequency-dependent increase in heart rate. This electrical-induced tachycardia is decreased by clonidine and increased by idazoxan. The effect of idazoxan was not observed in experiments using continuous stimulation of the sympathetic outflow. It is then concluded that alpha 2 antagonists are able to increase the electrical-induced tachycardia in the pithed rat as they do in the dog or the cat.

Role of mediastinoscopy in pretreatment staging of patients with primary lung cancer.

Since the introduction of mediastinoscopy, there has been a great deal of discussion regarding indications for this technique and the significance of positive findings. We undertook this study to determine the role of clinical staging and the value of routine mediastinoscopy in the treatment selection of patients with primary lung cancer. From 1975 to 1983, 1,259 consecutive patients with proven and operable lung cancer underwent preresection mediastinoscopy. Nodes were sampled at three levels, and findings were recorded by location, invasiveness, and histology. There were no operative deaths, but 3 patients had a major complication. Mediastinoscopy was positive in 339 (27%) patients and negative in 920 (73%). In the group with positive findings, 303 patients had no operation because a curative resection was not possible (extranodal metastases, 180; location, 76; histology, 47). No patient survived 5 years, and only 4% survived 2 years. Of the 36 patients considered to have operable disease, 28 underwent resection with a projected 5-year survival of 18%. In the group with negative findings, 89% had a curative resection with a hospital mortality of 3.2% and 5-year survival of 53%. When results of mediastinoscopy were correlated with findings at thoracotomy, the sensitivity of the test was 93% on nodes in the superior mediastinum and the specificity, 100%. This study shows that mediastinoscopy is safe and is an accurate indicator of the presence or absence of tumor in superior mediastinal nodes. If positive nodes are found, a curative resection is generally not possible, thoracotomy is avoided, and the overall survival is low.

Pre- and postsynaptic a-adrenoceptor blockade by (imidazolinyl-2)-2-benzodioxane 1-4 (170 150): antagonistic action on the central effects of clonidine.

While the specificities of alpha 1-adrenoceptor blocking agents are considered as satisfactory, those of alpha 2-adrenoceptor blocking agents are only weak. Therefore, a more selective alpha 2-adrenoceptor blocking agent is needed. In anaesthetized dogs and rats, (imidazolinyl-2)-2-benzodioxane 1-4 (170 150), a benzodioxane derivative, antagonized the pressor effects induced by adrenaline, noradrenaline and phenylephrine, but did not modify the effects of acetylcholine, histamine and isoprenaline. Therefore, its selectivity is satisfactory. In the anaesthetized dog, 170 150 (0.1 mg.kg-1) increased the tachycardia induced by electrical stimulation of the cardiac sympathetic nerve and antagonized the inhibitory effects of clonidine. In addition, 170 150 blocked and reversed some centrally mediated effects of clonidine such as the fall in blood pressure, bradycardia and reduction of splanchnic discharges in the dog, and the loss of the righting reflex induced by clonidine in the chicken. The centrally mediated effects of clonidine are attributed to stimulation of alpha 2-adrenoceptors; therefore, 170 150 appears to antagonize these alpha-adrenoceptors. Moreover, 170 150 appears to be more potent than piperoxan.

[Uterine and sigmoid perforations following voluntary interruption of pregnancy by aspiration. A case report (author's transl)].

A recent case of mixed uterine and colic perforations associated with generalized peritonitis is reported, the lesions developing within the context of the digestive complications of the method known as aspiration for voluntary interruption of pregnancy. The frequency, severity, and treatment of these lesions are described, urgent laparotomy being necessary to correct both abdominal digestive tract and genital lesions. Prevention of these complication technique. Coelioscopy is essential if a perforation is suspected, and must be followed by a laparotomy if its presence is confirmed.

Effect of carrageenan-induced inflammation on the binucleate keratinocytes of guinea pig palatal mucosa.

A 1% carrageenan solution was injected into the palatal mucosa of male guinea pigs between the two first molars. Biopsy specimens were taken 1, 3, 6, 12 and 24 hours after the injection. Control specimens with healthy mucosa and tyrode injected mucosa were used in order to evaluate the carrageenan-induced inflammation. An intense inflammatory reaction occurred within hours after the carrageenan solution injection. The palatal epithelium exhibited a considerable increase in the number of binucleate cells (P less than 0.001). As proposed by several authors, the inflammation of the underlying connective tissue might explain this phenomenon. The presence of binucleate cells would be an indication that the migration of epidermal cells from the basal to the horny layer proceeds in a hasty and immature fashion.

Biochemical values of immigrant groups in north-east France.

Laboratory-test results for a population of immigrants living in north-east France revealed differences between the concentration of the blood constituents of immigrants and native French people. By immigrants we mean persons now living in France but coming from other geographical locations such as Italy, the Iberian Peninsula, northern or central Europe, northern Africa, or the Near or Middle East. Multiple regression analysis confirmed the need to establish reference limits in immigrants for many blood constituents because of a significant shift of the curve relative to that for the French population. We have determined appropriate reference limits, for some ten blood constituents in each group of immigrants.
