The effect of occlusive treatment on human skin: an electron microscopic study on epidermal morphology as affected by occlusion and dansyl chloride.

The effect of occlusion on epidermal morphology was studied at electron microscopic resolution. the morphological changes recorded were relatively mild, though conspicuous, and consisted in dilatation of the intercellular space, appearance of cytoplasmic vacuoles and an oedematous change in the dermis. Stereological analysis showed that the dilatation of the intercellular space had a biphasic time-dependent course. Dansyl chloride applied under occlusion was shown to cause the same type of changes as did simple occlusion, but the changes appeared earlier.

Reactions of epidermal keratinocytes in sensitized and non-sensitized guinea pigs after dichromate exposure: an electron microscopic study.

The reaction in epidermal keratinocytes in sensitized and non-sensitized, normal guinea pigs after dichromate exposure has been investigated by electron microscopy. The results of the investigation show that the morphological alterations in keratinocytes in irritant and contact allergic reactions to dichromate are of a nonspecific nature. This underlines the fact that morphological reactions in the keratinocyte are uniform. The reactions were dose- and time-dependent. The morphology of the contact allergic reaction to dichromate in the guinea pig is consistent with that described for DNCB.

X-ray microanalysis in dermatology.

The current trends in X-ray microanalysis as applied to dermatology are described by reviewing papers which have made a significant contribution to the field. Under separate headings, the analysis of hair, nail and epidermis is reviewed with an emphasis on quantitative results. Our recent work on green hair has revealed a higher than normal copper content derived from copper-containing tap and pool water. In nail the sulfur content was shown to be evenly distributed throughout the nail plate and the calcium content to be too low to influence on the "hardness" of the nail. Our study of nails from cystic fibrosis patients showed that the Na, K, and C1 content increased, which is diagnostic for the disease, and that the Ca content could be used to monitor the preparation of nail samples for analysis. With the aid of X-ray microanalysis it has been possible for the first time to study the electrolyte composition of the different strata of epidermis by using freeze-fixed and freeze-dried cryosections. Only the basal stratum maintains a normal Na/K quotient and all cell layers above that one appear to have dysfunctional cell membranes, i.e. inadequate pumping of Na out of the cells. A subsequent study of the effects on normal epidermis by DNCB showed that after 6 hours post-treatment, abnormal Na, K and P levels appeared which differed from those seen in post-mortem changes of normal epidermis.

The rigidity of human fingernails: a biophysical investigation on influencing physical parameters.

(A) The property of withstanding bending and buckling in a composite material such as the nail is best described by such terms as rigidity or stiffness. (B) The physical property of rigidity can be understood from the macromolecular orientation of the keratin filaments and the cellular architecture of the dorsal and ventral nail plates. (C) The rigidity is influenced by factors such as the water content of the nail material. Specimens investigated must therefore be well equilibrated prior to the measurements. (D) Detergents, organic solvents, oils etc. are factors which are likely to influence nail rigidity.

Electrolytes in nails analysed by X-ray microanalysis in electron microscopy. Considerations on a new method for the diagnosis of cystic fibrosis.

Patients with cystic fibrosis (CF) have an increased concentration of sodium in their nails. Hitherto, only neutron activation analysis has been considered for the diagnosis of cystic fibrosis by analysis of electrolytes in nails. It has been thoroughly tested methodologically and clinically. However, the intrinsic advantages of X-ray microanalysis and the results obtained in this study suggest that this method, after further testing, may be a useful diagnostic acid for cystic fibrosis. In comparison with neutron activation analysis, X-ray microanalysis has the advantage of simultaneously giving the concentrations of several elements and may be accessible at any hospital with an electron microscope fitted with the necessary equipment. Nails of CF-patients are here shown to have increased concentrations of Na, K and Cl, which will make the diagnosis of cystic fibrosis more reliable. The possibility of using sulphur as a reference element may eliminate the weighing procedure necessary in neutron activation analysis.

Assessment of chromium allergy: features of patch test reactions at electron microscopic resolution.

The possible correlation of clinically recorded test reactions and morphological changes at electron microscopic resolution in human subjects allergic to chromate (CrVI) was investigated. The effect of simple occlusion and of vehicles, e.g. distilled water, an alkaline buffer, and petrolatum was also studied 72 hours after application. The results indicate that even simple occlusion causes reactive changes in living epidermal cells. These changes become conspicuous in occlusion with distilled water, more pronounced with petrolatum, and were destructive in the case of the alkaline buffer. Chromate solutions produced varying degress of morphological change but were less pronounced in a vehicle of distilled water, compared with the changes seen when the chromate was combined with the alkaline buffer. Destructive cellular changes were observed in the electron microscope even in sections from test ares where no macroscopical reactions were recorded. When the clinical evaluation of hypersensitivity was done 72 hours after the application of the patch test the morphological changes were too advanced to provide specific pathognomonic information.

Calcium and sulfur location in human nail.

Analytical electron microscopy was performed in order to elucidate the importance of calcium and sulfur to the hardness of nails. The free surfaces adsorb a considerable quantity of calcium from the environment. It is suggested that such adsorption occurs due to ion exchange. This concept is supported by the fact that the distribution of sulfur does not vary over the nail cross-section. Analytical electron microscopy offers unique possibilities of elemental distribution analysis in tissue fragments or sections from keratinized tissues.
