Oscillations of cytosolic Ca2+ in pituitary cells due to action potentials.

Electrical activity in non-neuronal cells can be induced by altering the membrane potential and eliciting action potentials. For example, hormones, nutrients and neurotransmitters act on excitable endocrine cells. In an attempt to correlate such electrical activity with regulation of cell activation, we report here direct measurements of cytosolic free Ca2+ changes coincident with action potentials. This was achieved by the powerful and novel combination of two complex techniques, the patch clamp and microfluorimetry using fura 2 methodology. Changes in intracellular calcium concentration were monitored in single cells of the pituitary line GH3B6. We show that a single action potential leads to a marked transient increase in cytosolic free calcium. The size of these short-lived maxima is sufficient to evoke secretory activity. The striking kinetic features of these transients enabled us to identify oscillations in intracellular calcium concentration in unperturbed cells resulting from spontaneous action potentials, and hence provide an explanation for basal secretory activity. Somatostatin, an inhibitor of pituitary function, abolishes the spontaneous spiking of free cytosolic Ca2+ which may explain its inhibitory effect on basal prolactin secretion. Our data therefore demonstrate that electrical activity can stimulate Ca2+-dependent functions in excitable non-neuronal cells.

[Electrophysiology of anterior pituitary cells (author's transl)].

Anterior pituitary cells in culture are excitable. As neurons or neuroendocrine cells they display action potentials either spontaneously or when depolarized. However, the ionic requirements for action potentials in endocrine cells appear to be different to those for neurons and even between the various types of endocrine cells. In anterior pituitary cells action potentials are largely calcium (Ca2+ -dependent. The involvement of Ca2+ ions in the spiking activity of anterior pituitary cells may be a determinant factor in the mechanism of secretion.

[Histofluorometric study of catecholamines of the external layer of median eminence in the rabbit (author's transl)].

In order to study the role of the Biogenic Amines of the Median Eminence (M. E.) in the ovulatory neuroendocrinological reflex in the female rabbit, we analysed by semi-quantitative histofluorometry (Zeiss) the post-coïtum variations in formaldehyde induced fluorescence (Falck and Hillarp's technique) in the dopaminergic layer of the M. E. 116 female rabbits (62 control, 54 post-coïtum) were examined. In all the animals the dopaminergic layer of the M.E. displayed variations in fluorescence, both in thickness and intensity, along the antero-posterior axis and lateraly. In this respect, these results partly recovered those reported in the Rat (Lofstrom, 1976). Moreover, in half of the post-coïtum animals, from the 5 th min. up to the 30 th min, the intensity of the fluorescence was increased in the whole external layer. These results provide some evidence for a correlation between the variations in the Dopamine content of the M.E. and the involvement of the nervous mechanisms implied in the ovulatory neuroendocrinological reflex.
