An alternate binding site for the P1-P3 group of a class of potent HIV-1 protease inhibitors as a result of concerted structural change in the 80s loop of the protease.

Structures of the complexes of HIV protease inhibitor L--756,423 with the HIV-1 wild-type protease and of the inhibitors Indinavir, L-739,622 and Saquinavir with the mutant protease (9X) containing nine point mutations (Leu10Val, Lys20Met, Leu24Ile, Ser37Asp, Met46Ile, Ile54Val, Leu63Pro, Ala71Val, Val82Thr) have been determined. Comparative analysis of these structures reveals an alternate binding pocket for the P1-P3 group of Indinavir and L--756, 423. The alternate binding pocket is a result of concerted structural change in the 80s loop (residues 79-82) of the protease. The 80s loop is pulled away from the active site in order to accommodate the P1-P3 group, which is sandwiched between the flap and the 80s loop. This structural change is observed for the complexes of the wild type as well as the 9X mutant protease. The study reveals that the 80s loop is an intrinsically flexible loop in the wild-type HIV-1 protease and that mutations in this loop are not necessary to result in conformational changes. Conformation of this loop in the complex depends primarily upon the nature of the bound inhibitor and may be influenced by mutations in the protease. The results underscore the need to understand the intrinsic structural plasticity of the protease for the design of effective inhibitors against the wild-type and drug-resistant enzyme forms. In addition, the alternate binding pocket for the P1-P3 group of Indinavir and L--756,423 may be exploited for the design of potent inhibitors.

Regulation of interleukin 2 and interleukin 2 receptor gene expression in human T cells: I. Effect of Ca2+-ionophore on phorbol myristate acetate co-stimulated cells.

Human peripheral blood T lymphocytes were treated with phorbol myristate acetate (PMA), an activator of protein kinase C (PKC) activity, and with the calcium ionophore A23187. The resulting accumulation of specific mRNA for interleukin 2 (IL-2) and interleukin 2 receptor (IL-2R), as well as IL-2 secretion and membrane IL-2R expression were examined. At low concentrations (0.1 microM), A23187 synergized maximally with PMA to induce proliferation, to increase IL-2R mRNA levels and the expression of membrane IL-2R, and to produce a low but sufficient accumulation of IL-2 mRNA and IL-2 secretion. A high concentration of A23187 (1.0 microM) did not show any synergism for the accumulation of IL-2R mRNA, membrane IL-2R expression and inhibited the proliferation of PMA co-stimulated T cells. It did, however, induce maximum accumulation of IL-2 mRNA and IL-2 secretion.

The relationship between women's basketball performance and will to win.

An analysis of the determinants of athletic performance indicated that "desire" or "Will to Win" was an important factor while an interactionist approach to personality suggested that differences in "Will to Win" might be expected both for different sports and at different levels of performance within a sport. The purpose of this investigation was to determine the relationship between "Will to Win" and performance in an eight-team women's basketball tournament. Players completed a 14-item "Will to Win" questionnaire. Team and individual scores were compared with various indices of performance. Results revealed that between two competing teams, the game was more likely to be won by the team with the higher average "Will to Win". The correlation between the mean "Will to Win" score for a team and its position of finish in a tournament was not significant. An ANOVA and Newman-Keuls comparisons revealed that the last place team had to "Will to Win" score that was significantly lower than five of the seven other competing teams. Data obtained on individual players indicated a significant but low correlation between "Will to Win" and the mean number of points scored by a player.

Age-related changes in the immune system of mice of eight medium and long-lived strains and hybrids. II. Short- and long-term effects of natural infection with parainfluenza type 1 virus (Sendai).

Studies on the effects of natural infection with parainfluenza type 1 virus (Sendai) on the immune system of 8 strains and hybrids of aging mice revealed that (a) 55 of 63 indices tested were abnormal as late as 8 months after the disappearance of clinical systoms, (b) thymus weight and cell indices and lymph node T cell activity still exhibited a statistically significant negative correlation with age following infection, and (c) immunologically immature young and immunodeficient old mice suffered more severe sequelae following viral infection than did adult or middle-aged mice as determined by accelerated decline of thymus indices, mortality, IgM Coombs' titer, and in the case of old mice, anemia. These findings suggest that chronic viral infection can accelerate immunologic aging and that viral infection in utero can suppress or retard the development of the T cell component of the immune system. In support of our previous study, they also suggest that the thymus regulates immunologic aging and that genetic factors regulating immunologic aging are not easily influenced by environmental factors.

Evaluating ambulatory care: three approaches.

The article describes three approaches to establishing an ambulatory audit program for nursing. These approaches are variations of a single methodology--that of the Joint Commission on Accreditation of Hospitals--used by the University of Minnesota Hospitals and Clinics. The authors discuss three audit topics from ambulatory care; postkidney transplant; desensitization; and well child.
