Overrepresentation of horizontal and vertical orientation preferences in developing ferret area 17.

One of the fundamental principles of visual cortical organization is that neurons form a "map" in which neighboring cells have similar orientation preferences. Previous anatomical and imaging studies have shown that although the exact layouts of these orientation preference maps vary between individuals, features of iso-orientation domains such as width and spacing appear constant within a species. Using chronic optical imaging of intrinsic signals we now demonstrate that in ferret area 17 a larger proportion of cortical surface is dominated by responses to horizontal and vertical contours than to the two oblique orientations. This was true for all ferrets studied both during development and in adulthood. Interestingly, however, we found that the degree of the overrepresentation varied significantly between individual animals. In some young ferrets, responses to horizontal and vertical stimuli developed faster than responses to oblique stimuli, and a much larger percentage of the cortex responded preferentially to horizontal and vertical stimuli. In other individuals, responses to all stimuli developed at roughly the same rate, and there was relatively little overrepresentation of horizontal and vertical preferences.

A flow cytometric method for rapid selection of novel industrial yeast hybrids.

We rapidly produced and isolated novel yeast hybrids by using two-color flow cytometric cell sorting. We labeled one parent strain with a fluorescent green stain and the other parent with a fluorescent orange stain, and hybrids were selected based on their dual orange and green fluorescence. When this technique was applied to the production of hybrids by traditional mating procedures, more than 96% of the isolates were hybrids. When it was applied to rare mating, three hybrids were identified among 50 isolates enriched from a population containing 2 x 10(6) cells. This technology is not dependent on genetic markers and has applications in the development of improved industrial yeast strains.

Selective sensitization to the psychosis-inducing effects of cocaine: a possible marker for addiction relapse vulnerability?

Patients in inpatient rehabilitation for uncomplicated cocaine dependence were asked whether, compared with the time of their first regular use, they could now identify changes in the effects of similar doses of cocaine. We asked about a spectrum of cocaine effects "then" and "now" and whether the same amount of drug caused effects to occur to about the same degree, less intensely (tolerance), or more intensely (sensitization). Nearly half our sample developed predominantly paranoid psychoses in the context of cocaine use. Sensitization was consistently linked only to psychosis-related cocaine effects. It has been proposed that mesolimbic dopaminergic sensitization might contribute to addiction severity. A preliminary followup of patients who were sensitized or nonsensitized to psychosis development suggests that rehospitalization for treatment of addiction may be more frequent in the sensitized group.

The effect of tenidap on cytokines, acute-phase proteins, and virus load in human immunodeficiency virus (HIV)-infected patients: correlation between plasma HIV-1 RNA and proinflammatory cytokine levels.

Proinflammatory cytokines may be important in the pathogenesis of human immunodeficiency virus type 1 (HIV-1) disease. Tenidap decreases interleukin (IL)-6, IL-1, and tumor necrosis factor (TNF) production by peripheral blood mononuclear cells and decreases IL-6 plasma levels in rheumatoid arthritis patients. In this randomized double-blind study, 43 HIV-1-infected patients received tenidap (120 mg) or placebo daily for 6 weeks and then crossed over to the alternative therapy for an additional 6 weeks. Mean entry CD4 cell count was 140/microL. Analyses were performed on cytokines, acute-phase proteins, virus load, and CD4 cell counts. With the exception of small differences in plasma TNF levels, tenidap had no significant effect on these indices. Significant correlations of plasma IL-6 and TNF levels with HIV-1 RNA were noted. Six patients discontinued tenidap due to rash. The effects of tenidap in HIV-1 infection contrast to results in arthritis patients, in whom tenidap decreased plasma levels of IL-6 and acute-phase proteins.
