Herpes simplex virus-encoded ribonucleotide reductase: evidence for the dissociation/reassociation of the holoenzyme.

35S-labeled cells infected with herpes simplex virus type 1 (HSV-1), temperature-sensitive (ts) mutant ts 1222 were used as a source of the large subunit of the viral ribonucleotide reductase (RR) to investigate the binding of the large (RR1) and small (RR2) subunits in the active enzyme. Mixing 35S-labeled RR1 from ts 1222 with unlabeled RR1/RR2 complex from wild type (wt) infected cells resulted in the formation of a complex between 35S-labeled RR1 and unlabeled RR2, indicating that the complex between the RR1 and RR2 subunits is dynamic and subunit dissociation/reassociation occurs during enzyme function. Similar results were obtained when unlabeled HSV-2 RR was substituted for HSV-1 RR, demonstrating that the holoenzyme can be formed the large subunit of HSV-1 RR and the small subunit of HSV-2.

Production of IGF-binding proteins by vascular endothelial cells.

Conditioned serum-free media from cultured human, bovine and rodent endothelial cells contained binding proteins with high affinity for the insulin-like growth factors (IGFs). After partial purifications on heparin or Multiplication Stimulating Activity (MSA)-affinity columns, 2 species of binding protein were identified, a major protein having Mr approximately 35,000 and a minor 22-28,000 protein. The binding proteins had greater affinity for IGF-I than IGF-II with no affinity for insulin or proinsulin. Substantial amounts of the binding proteins remained cell-associated, loosely bound to the outer cell surface of the endothelial cell. Binding protein(s) from human endothelial cells cross-reacted with antibodies to the 53,000 dalton acid-stable human serum binding protein. Production of endothelial binding proteins was not stimulated by growth hormone or insulin. We conclude that endothelial cells in culture produce large quantities of specific IGF binding proteins. Such binding proteins should be relevant in understanding the complex metabolism and function of the IGFs in the intact host.

Specific and varied practice of motor skill.

To assess the potential effect of specific and varied practice on the development of motor schema, 64 children in two age groups (8 yr. and 12 yr.) were tested on a simple throwing task at the beginning and end of a 12-wk. physical education program. For the throwing test the children were assigned to either a specificity or a schema group (varied practice). The task was performed without visual feedback, but knowledge of results was given after each trial. Prior to both tests at the criterion target, the specific group was given practice throws using the criterion target distance, whereas the schema group practiced on two other targets. No differences were found between the two groups at the beginning of the program but on the posttest those children receiving a variety of practice on the throwing task performed significantly better than the specific-practice group. Thus it was suggested that a varied practice schedule may facilitate the initial formation of motor schema, and this process may be enhanced by participation in a physical education program.
