[The lymphocyte surface].

The main function of lymphocyte is the immunological supervision which is brought about via receptor structures of the cell surface. An antigen is recognized by surface immunoglobulin of B-lymphocytes as well as by antigen-recognizing receptors of T-lymphocytes and natural killers. The regulation of lymphocyte functions if brought about via another class of receptors (Fc, C3-receptors). In immunopathological processes certain characteristics of lymphocytes surface change. These changes are manifested at the level of cell populations and clones of antigen-specific lymphocytes.

[Detection of the cytotoxic effect of tumor cells against the targets of normal killers].

The cells of 2 mouse leukemias (L-1212 and EL-4), the cells of mouse sarcoma (MX-11) and blood lymphocytes from 5 subjects with chronic leukemia were shown to damage the membrane of K-562 and EL-4 cells maintained in vitro. The membrane damage manifested in the rise of the permeability by the pancreatic ribonuclease molecules (m. m. 12,000 d). Experiments with tumor cell suspension fractionation have shown that such permeability cannot be attributed to the admixture of lymphocytes and macrophages to the tumor effector cells. Experiments with cold inhibition have revealed that the damage of the target cell membrane can be produced only by the contact of the tumor cell with the target. It is not related to the deterioration of target metabolism in the presence of a considerable number of tumor cells, or to the accumulation of any toxic products in the culture medium.

[Immunoglobulin-positive cells in mouse bone marrow].

The content of Ig-bearing lymphocytes and their precursors in the mouse bone marrow was investigated 6 and 36 hours after the hydroxyurea treatment. Some increase of the B-cell content takes place in the trated bone marrow. Dividing and non-dividing B-cell precursors, except the stem cells, were practically absent.

[Genetic control of the content and avidity of antigen-binding B-lymphocytes in the mouse spleen].

The content (per 10 nucleated cells) and avidity curves of IgM positive B-lymphocytes forming rosettes with trinitrophenyl-sheep erythrocytes (TNP-RFC) were compared in two mouse lines (BALB/c C57BL/6), F1 hybrids (BALB /c X C57BL/6) and backcross hybrids (F1 X BALB/c, F1 X C57BL/6). Trinitrophenyl-bovine serum albumin (TNP24BSA) and dinitrophenyl-bonvine serum albumin (DNP23BSA) and sulfanil-BSA (Sulf17-BSA) in different dilutions were used as inhibitors. BALB/c spleen contained by 60% more TNP-RFC than C576/6 spleen. F1 hybrids (F1 X BALB/c) contained on the average by 35% more TNP-RFC than (F1 X C57BL/6). Inhibition curves (avidity curves) were essentially different in the two mouse lines and F1 hybrids. A conclusion was drawn that the TNP-REC content and avidity were under a strong genetic control. Together with the assumption of random expression of V-genes (idiotype genes?) in the lymphocytes the above data suggest that the simplest mechanism of the genetic control could be a definitive ratio between the corresponding groups of V-genes.

[Changes in the electroretinogram and concentration or rhodopsin in the Hunter strain of rats during development of hereditary retinal degeneration].

Electroretinographic changes as well as rhodopsin content in the isolated retina and the eye cup in Hunter rats were studied during inherited retinal degeneration. The concentration of rhodopsin in the eye cup increases for the first 45 days after birth and then decreases slowly; the visual pigment concentration in the retina reaches its maximum by the 25th day and drops abruptly after the 35th day. The amplitude of the electroretinographic b-wave evoked by near-threshold and saturating stimuli decreases evenly till 35th day. After this day the threshold drastically increases and a-wave disappears. Thus, the 35th day is crucial in the degeneration process. The possible role played by abnormal rhodopsin metabolism in the changes observed is discussed.

[Rosette formation and humoral immune response to 2,4-dinitrophenol in mice of different inbred strains].

A study was made of the content of rosette-forming cells to DNP-ovalbumin in the spleen of mice of different inbred strains. The values of the rosette-forming cells and of the titre of serum agglutinins to the DNP-group in DNP--bovine gamma-globulin immunization of mice of these strains were determined. It was shown that there were interstrain differences in respect to the normal and immune rosette-forming cells and also humoral antibodies. There was noted a direct correlation between the number of the immune rosette-forming cells and the antibody titre in the serum of mice of the same inbred strain. Immune response (both by the content of rosettes in the spleen and by the antibody level) proved to be the minimal in mice with the highest level of rosette-forming cells. There proved to be no inverse relationship.

[Treatment of amyotrophic lateral sclerosis with ribonucleotides].

In the treatment of 31 patients with lateral amyotrophical sclerosis the authors used the preparation ENKAD containing a complex of mono- and oligoribonucleotides. In 6 cases there was aggravation of the condition while in 18 of the observed cases--improvement. In 7 patients the condition was not altered. The treatment was convened under biochemical control (the assessment of the uric acid in the blood serum and urine and the excretion of oxypurine in the urine). It was possible to depict definite clinicobiochemical correlations. The conclusion is made that ENKAD can be used in the early stages of the disease and mainly in focal segmentary-nuclear lesions. The preparation is contraindicated in generalization of the process and in impetuous development of the disorder.

[Role of macrophages in the mechanism of specific entrapment of sensitized lymphocytes in lymph nodes containing antigen].

A study was made of the participation of macrophages in the phenomenon of specific trapping of lymphocytes activated with the antigen in the lymph nodes containing the same antigen. Administration of peritoneal macrophages which phagocytized in vitro sheep erythrocytes (as well as of erythrocytes alone) led to specific trapping of lymphocytes in the regional lymph nodes. Preliminary treatment of macrophages loaded with the antigen with antierythrocytic serum decreased the lymphocyte trapping. A supposition was put forward that lymphocytes with receptors on their surface were bound on the macrophage membrane into which the antigen molecules were incorporated.

Separation of rosette-forming cells with the aid of an immunosorbent based on sephadex G-25 and G-75.

A method of separating rosette-forming cells on columns with an immunosorbent based on sephadex G-75 is described and has been tested. The protein antigen (bovine serum albumin-BSA) is attatched by covalent bonds to the surface of the Sephadex granules oxidized by sodium periodate. Cells with receptors on their surface were tested by the rosette method. Up to 88% of the rosette-forming cells were retained on a column packed with BSA-Sephadex granules. Nonspecific retention of the cells was relatively small, about 4%.
