Antineoplastic-drug effect evaluated with a new X-ray stereophotographic measurement of the tumour volume.

A method for determination and follow up of the size of inoperable intraabdominal tumours has been devised. At laparotomy, tantalum balls are deposited into the capsule of the tumour. An X-ray stereophotogrammetric method determines their position, which define a polyhedron whose volume can be computer calculated. The value of the method was proved in four patients with inoperable ovarian carcinoma, which provides a simple way for the continuous follow up of the effect of antineoplastic therapy in advanced malignant tumours.

No increase of the fibrinolytic activity of the human endometrium by progesterone-releasing IUD (progestasertR).

An increase in menstrual blood loss is one of the most common side effects of plastic IUDs and, though less often, also of copper IUDs. This has been explained by the observation that IUD enhances the fibrinolytic activity of the endometrium. A progesterone-releasing IUD (ProgestasertR) does not increase menstrual bleeding. The fibrinolytic activity of the endometrium was studied in 17 women before and after insertion of a progesterone-releasing IUD. It did not enhance the histochemically determined fibrinolytic activity in the endometrium.

The effect of tranexamic acid on the fibrinolytic activity of vein walls.

16 patients were treated with the fibrinolytic inhibitor tranexamic acid in an initial i.v. dose of 1 g followed by an oral dose of 4 g a day for one week and 3 g a day for two weeks. Biopsy specimens of superficial veins were obtained before and on the last day of treatment and examined histochemically for their fibrinolytic activity. Tranexamic acid did not suppress the fibrinolytic activity in the vessel walls, which is the most important link in the fibrinolytic defence system against thrombosis.

Some characteristics of urokinase released in organ culture of human kidney.

Plasminogen activator produced in organ culture of human kidney, i.e. in the histotypical arrangement of the tissue, was partially purified by affinity chromatography on para-aminobenzamidine coupled to Sepharose by a 6-carbon spacer, followed by gel chromatography on Sephadex G-100. The molecular weight of 2 active peaks were 27,000 and 52,000 daltons respectively. It was inhibited by DFP and by IgG antiurokinase.

Treatment of advanced breast cancer with chemotherapeutics and inhibition of coagulation and fibrinolysis.

A case of advanced breast cancer with cerebral metastasis and pleurisy is reported in which irradiation and cytostatics had failed to retard progressive growth and spread of the tumour. Adjuvant therapy with heparin combined with the fibrinolytic inhibitor tranexamic acid was followed by regression of the cerebral metastasis as well as the pleurisy. When last seen one year later, the patient was free from symptoms.

Coagulative and fibrinolytic studies on postmenopausal women treated with a new non-steroidal oestrogen.

Treatment of postmenopausal women with a non-steroid oestrogen (P1496) in a dose of 50 mg a day for 2 ,days before operation for prolapse did not suppress the histochemically determined fibrinolytic activator content of the vessel wall. Neither was any change found in the concentration of P&P-complex (prothrombin, factor VII, factor X), factor VIII, antithrombin III, alpha1-antitrypsin, alpha2-macroglobulin or the inhibitors of urokinase induced plasminogen activation. Nothing suggested a thrombogenic effect of this non-steroidal oestrogenic compound.

Chemotherapy of metastatic carcinoma of the breast. A 4-drug regimen.

Combination chemotherapy (vincristine, adriamycin, cyclophosphamide, methotrexate with leucoverin rescue) in 32 patients with metastatic carcinoma of the breast resulted in an overall response rate of 81 per cent (9/32 complete remission, 17/32 partial remission). No serious side effects were noted. Karnofsky performance index improved in 24/26 responders.

Significance of increased fibrin/fibrinogen degradation products and presence of fibrin monomers in pregnant women.

Fibrin/fibrinogen degradation products (FDP) and fibrin monomers might occur as a sign of "athological proteolysis" in complicated pregnancies such as toxaemia, hepatosis and placental insufficiency, eut also in otherwise clinically uncomplicated pregnancies. 18 pregnant women with increased proteolysis, but without any other clinical symptoms of complications were therefore investigated to determine the outcome of their pregnancies. The investigation showed that proteolysis with moderately increased serum FDP and fibrin monomers but without any other clinical signs of complications of pregnancy need not necessarily imply any risk to the mother or the foetus.

[New method of studying the release of fibrinolysis activators in tissue cultures].

Tissue cultivation in the presence of standardized fibrin clot containing plasminogen permitted to reveal and to study quantitatively the relase of the fibrinolysis activators into the medium (by the amount of the fibrin-fibrinogen degradation products). A possibility of in vitro study of the regulation of fibrinolysis activators release by the tissues was offered by the method described.
