The murine Ah locus: a new allele and mapping to chromosome 12.

The Ah locus in mice, the presumed structural gene for the Ah receptor, is polymorphic in mice: some inbred strains carrying the Ahb allele express a high affinity receptor and are sensitive to receptor agonists, while other strains carrying the Ahd allele express a lower affinity receptor and are less sensitive to agonists. Using the photoaffinity ligand for the Ah receptor, [125I]-2-azido-3-iodo-7,8-dibromodibenzo-p-dioxin, we have identified two allelic forms of the high affinity receptor (Ahb). In mouse strains of the C57 and C58 family and MA/MyJ, the photoaffinity labeled peptide in hepatic cytosol has an apparent molecular mass of 95 kDa (Ahb-1 allele), whereas in other responsive strains (e.g., C3H/HeJ, BALB/cByJ, A/J) the labeled peptide has an apparent mass of 104 kDa (Ahb-2 allele). In genetic crosses and backcrosses between C57BL/6J and C3H/HeJ mice, the expression of these peptides is consistent with codominant inheritance of two alleles at a single locus. From Scatchard plots of the specific binding of the reversible ligand, [3H]-2,3,7,8-tetrachlorodibenzo-p-dioxin, to hepatic cytosol, most strains of mice with the Ahb-1 allele were found to have an Ah receptor with a binding affinity, KD, of 0.4-0.7 nM, and a receptor concentration of 130-160 fmol/mg of protein, whereas most strains carrying the Ahb-2 allele have a slightly lower receptor affinity, KD = 0.8-1.3 nM, and a slightly lower receptor concentration, 80-110 fmol/mg of protein. From analysis of the strain distribution pattern of the Ahb-1 and Ahb-2 alleles in recombinant inbred strains, the Ah locus was linked to several unmapped loci (H-17, H-38, Ltw-2, Ly-18, D1Nyul, D1Nyu2), and this entire group mapped to chromosome 12.

Common sites of viral integration in lymphomas arising in AKXD recombinant inbred mouse strains.

Lymphomas from 21 AKXD recombinant inbred mouse strains were analyzed for retrovirally induced rearrangements in seven known or putative proto-oncogene loci. Among the rearrangements detected in the 258 lymphomas screened, most rearrangements appeared to be caused by viral integration. Rearrangements were detected in the Myc, Pvt-1, Pim-1, Mlvi-1, Mlvi-2, and Fis-1 loci, but not in the Myb locus. Seven lymphomas contained rearrangements in two loci. Nearly 90% of the rearrangements were observed in T cell lymphomas; few were detected in B cell or myeloid tumors. Rearrangements in Pvt-1, Fis-1, Mlvi-1, and Mlvi-2 were identified only in T cell lymphomas. Alterations in the Myc and Pim-1 loci, although occurring predominantly in T cell lymphomas, were occasionally detected in other types of lymphoma. These data suggest that the repertoire of cellular proto-oncogenes activated by integration of virus into lymphomas may be different for each hematopoietic cell lineage. The AKXD lymphomas represent a useful resource for identifying these sites.

Chromosomal location of the genes encoding complement components C5 and factor H in the mouse.

Complementary DNA probes corresponding to the factor H and C5 polypeptides have been used to determine the chromosomal localizations of these two complement components. Both probes revealed complex and polymorphic arrays of DNA fragments in Southern blot analysis of mouse genomic DNA. Following the distribution of these bands in panels of somatic cell hybrids carrying various combinations of mouse chromosomes on a constant rat or Chinese hamster background allowed the localization of the C5-associated fragments to proximal chromosome 2 and the localization of the factor H-associated fragments to chromosome 1 or chromosome 3. Following the inheritance of DNA restriction fragment-length polymorphisms revealed by the probes in recombinant inbred mouse strains allowed the factor H-associated fragments to be mapped to Sas-1 on chromosome 1, and the C5-associated fragments to be mapped to Hc. Analysis of three-point crosses, in turn, placed the latter locus 19 cM distal to Sd on chromosome 2. We have designated the two loci Cfh and C5, respectively. This genetic analysis raises the possibility that C5 and factor H are both encoded by complex loci composed of distinct structural and regulatory genes.

The murine gamma-chain of the T cell receptor is closely linked to a spermatocyte specific histone gene and the beige coat color locus on chromosome 13.

With the use of standard genetic techniques, we have mapped the T cell gamma-chain gene locus (Tcrg) to the proximal end of mouse chromosome 13. Close genetic linkage was demonstrated between a restriction fragment length polymorphism in the Tcrg locus and a locus (HlS) encoding a spermatocyte-specific histone electrophoretic variant (HlS) by using recombinant inbred strains. Because HlS is closely linked to the dominant visible marker extra toes (Xt), Tcrg must be within 5 cM of the beige (bg) locus. Mice homozygous for the recessive mutation beige (bg/bg) show diminished pigmentation, enlarged lysosomes in granulocytes, and a selective deficiency of natural killer cell function. The bg mutation in mice is an animal model for humans with Chediak-Higashi syndrome; we speculate that the human gamma-chain may be closely linked to the human mutation and may be a useful diagnostic tool. Seven restriction enzymes identify five haplotypes of the highly polymorphic gamma-chain in mice.

Surgery for malignant melanoma: from which limb should the graft be taken?

A study of 186 melanomas requiring skin grafts and followed for 1-13 years has shown no preferential implantation in an ipsilateral donor site, despite standard surgical teaching that skin grafts for melanoma defects should not be taken from the same limb as the primary tumour. Two cases were encountered of preferential metastasis to a distant donor site outside the lymphatic drainage of the melanoma and we present a further case where ipsilateral recurrence simulated donor site recurrence. It is clear that the major mechanism of spread to distant donor sites is by the haematogenous route to an area of acute trauma. Hence it is recommended that where possible donor skin is taken from the same limb as the tumour, so that any preferential metastasis will remain within an area amenable to regional limb perfusion.

Morbidity, mortality and local recurrence following regional node dissection for melanoma.

Eighty-six patients who underwent regional node dissection (RND) for melanoma were reviewed with respect to short and long term postoperative morbidity, mortality and local recurrence rate. Twenty-eight per cent of dissections were prophylactic and seventy-two per cent therapeutic. There were significant wound related complications which tended to delay hospital discharge, in particular following axillary and groin dissections, but symptomatic long term complications were infrequent. The postoperative mortality rate was 1.2 per cent. Local recurrence in RND scars was particularly common in the neck (33 per cent). RND for melanoma is a safe procedure, with significant short term but relatively little long term morbidity. Both recurrence and morbidity should be borne in mind when considering patients for prophylactic RND. Surgeons should be prepared to use axial or myocutaneous flaps where extensive skin resection is necessary. A controlled trial of prophylactic antibiotics would be useful to try to reduce the incidence of wound infection.

AKXD recombinant inbred strains: models for studying the molecular genetic basis of murine lymphomas.

We analyzed the lymphoma susceptibility of 13 AKXD recombinant inbred mouse strains derived from AKR/J, a highly lymphomatous strain, and DBA/2J, a weakly lymphomatous strain. Of the 13 strains used, 12 showed a high incidence of lymphoma development. However, the average age at onset of lymphoma varied considerably among the different AKXD strains, suggesting that they have segregated several loci that affect lymphoma susceptibility. A relatively unambiguous classification of lymphomas was made possible by using histopathology in addition to detailed molecular characterization of rearrangements in immunoglobulin heavy and kappa light genes and in T-cell receptor beta-chain genes. Among the 12 highly lymphomatous strains, only 2 were identified that, like the parental AKR/J strain, died primarily of T-cell lymphomas. Three strains died primarily of B-cell lymphomas, and one strain primarily of myeloid lymphomas. Six strains were susceptible to both T-cell and B-cell lymphomas. Thus, these strains have segregated genes that affect both lymphoma susceptibility and lymphoma type and should prove to be useful models for studying the molecular genetic basis of murine lymphomas.

Malignant melanoma of the hand and foot: diagnosis and management.

Forty-six patients with melanoma of the hand (5) and foot (41) studied prospectively between 1972-84 have been reviewed to determine guidelines for diagnosis and management. The clinical appearance is varied and the lesions are often misdiagnosed as warts or fungal infections. Clinical assessment of tumour thickness on the sole is difficult because of the nature of the plantar skin. Incision rather than excision biopsy is indicated because of the functional consequences of large excisions on the hand and foot. Melanoma of the toe was treated by local metatarsophalangeal amputation. Plantar melanomas were excised with a 3 cm margin. Dorsum melanomas were treated by a selective policy of 1, 2 or 3 cm margins according to clinical assessment of tumour thickness. These policies have resulted in only one case of local recurrence. An in situ Silastic foam mould facilitates immediate application of split skin grafts to irregular areas. The functional results of split skin grafts on sole and heel have been satisfactory. Toe lesions were thickest, dorsum thinnest, sole and heel intermediate. The prognosis related to these groupings of tumour thickness. Simultaneous clinical nodal involvement carried a hopeless prognosis, and most patients developing nodes within 2 years die--unless treated by prophylactic dissection. The role of prophylactic dissection is still not defined, but it is likely that it will be used more frequently in the future. Early diagnosis offers most hope of improving the outlook. Many elderly patients with nodular lesions have had moles for many years. Younger patients are now being seen with thin lesions, even on the toe.

Linkage of a 7S RNA sequence and kappa light chain genes in the mouse.

A mouse 7S RNA cDNA plasmid clone was employed to identify and map DNA restriction fragment variants using recombinant inbred (RI) and congenic mouse strains. More than a dozen such restriction variants were identified and mapped to different regions of the mouse genome. One such variant, designated Rn7s-6, showed close linkage to the Ly-2,3-Igk-V (T lymphocyte antigens 2 and 3, kappa immunoglobulin variable region) cluster of markers on chromosome 6. No recombinants were detected among three of these markers in 59 RI strains. On the basis of these data, the Rn7s-6 sequence may be placed within 1.3 centimorgans of Ly-3 and one of the Igk-V-region markers, Igk-Ef1. Two mouse stocks with previously identified crossovers within the Ly-2,3-Igk-V region were used to sublocalize Rn7s-6. The results are consistent with the gene order (Ly-2, Ly-3)-(Rn7s-6, Igk-Ef1)-Igk-Ef2. Several mouse plasmacytomas, known to have various parts of the kappa chain complex deleted, retain the Rn7s-6 sequence. The Rn7s-6 variant is a plus/minus variant; no sequence allelic to Rn7s-6 is found in inbred strains that share the Ly-3a-Igk-Ef1a haplotype.

Mouse serum amyloid P-component (SAP) levels controlled by a locus on chromosome 1.

Recombinant inbred strains were used to demonstrate the existence of a major locus on chromosome 1, designated Sap, which controls the endogenous concentration of the mouse acute phase reactant, serum amyloid P-component (SAP). Levels of SAP were associated with alleles at the Ly-9 locus in two sets of RI strains: BXD (C57BL/6J X DBA/2) and BXH (C57BL/6J X C3H/HeJ). Low endogenous levels of SAP were present in the C57BL/6J progenitor strain and in most of the RI strains which inherited the Ly-9b allele. High levels of SAP were present in the DBA/2J and C3H/HeJ progenitors and in most of the RI strains which inherited the Ly-9a allele. In the BXD strains 91% of the genetic variation of SAP levels was accounted for by segregation at the Ly-9 locus while an additional 9% was attributed to genetic factors unlinked to Ly-9. In the BXH strains the percentage of genetic variation accounted for by Ly-9 segregation was reduced to 46%, while 54% was accounted for by other genetic factors. Because of background genetic variation it was not possible to detect any crossovers between Sap and Ly-9. However, in the BXD strains the linkage between Sap and Ly-9 appears to be quite close. The B6.C-H-25c congenic strain, which carries a segment of BALB/c chromosome 1 including the minor histocompatibility locus H-25 on a C57BL/6By background, had the same endogenous SAP level as the BALB/c donor strain.

Development of an air displacement method for whole body volume measurement of infants.

An infant enclosed in a rigid-walled chamber displaces a volume of air equal to its own volume. The volume of air displaced can be estimated by the change in pressure produced by a standard reduction in the chamber's volume according to Boyle's law. An instrument embodying this principle has been developed in which the differential pressure between two identical chambers is measured during equal sinusoidally imposed volume changes in the two. Problems arising from variable departure of conditions of pressure cycling, from isothermal towards adiabatic, have been dealt with by empirically derived corrections. Data are presented on the use of the method for low birth-weight infants.

Cellular genes in the mouse regulate in trans the expression of endogenous mouse mammary tumor viruses.

The transcriptional activities of the eleven mouse mammary tumor virus (MMTV) proviruses endogenous to two sets of recombinant inbred (RI) mouse strains, BXD and BXH, were characterized. Comparison of the levels of virus-specific RNA quantitated in each strain showed no direct relationship between the presence of a particular endogenous provirus or with increasing numbers of proviruses. Association of specific genetic markers with the level of MMTV-specific RNA was examined by using multiple regression analysis. Several cellular loci as well as proviral loci were identified that were significantly associated with viral expression. Importantly, these cellular loci associated with MMTV expression segregated independently of viral sequences.

Chromosomal assignment of two endogenous ecotropic murine leukemia virus proviruses of the AKR/J mouse strain.

The AKR/J mouse strain is genetically fixed for three different ecotropic murine leukemia virus genomes, designated Akv-1, Akv-3, and Akv-4 (Emv-11, Emv-13, and Emv-14). With recombinant inbred strains and crosses with linkage-testing stocks, Akv-3 and Akv-4 were placed on the mouse chromosome map. Akv-3, which encodes a replication-defective provirus, maps near the agouti coat color locus, a, on chromosome 2. Akv-4, which is replication competent, maps near the neurological mutant gene locus trembler, Tr, on chromosome 11. Akv-1 and Akv-2 (Emv-12), an ecotropic provirus carried by AKR/N but not AKR/J, have previously been mapped to chromosome 7 and 16, respectively. Thus, the four Akv proviruses mapped to date are on four different chromosomes. Akv-3 is the second ecotropic murine leukemia virus provirus to be mapped near the agouti locus. The results are discussed in relation to possible nonrandomness of viral integration.

The major histocompatibility complex-restricted antigen receptor on T cells: the genetics of expression of an allotype.

The monoclonal antibody KJ16-133 binds an allelic determinant expressed on the antigen-specific, major histocompatibility complex (MHC)-restricted receptors on approximately 20% of T cells in most mouse strains. The locus controlling the presence or absence of the determinant mapped 9.8 +/- 2.2 centimorgans from the Igk/Ly-2 locus on chromosome 6 in mice, and may be the beta-chain locus. Other genetic loci were identified that controlled the frequency of cells that expressed the allele in positive mice. One of these was the MHC itself, which may control expression of the beta-chain allele by controlling T cell repertoire. The identity of the other, as yet unmapped locus is unknown. KJ16-133 was used to show that T cell receptor gene products are expressed in a manner consistent with allelic exclusion.
