Bovine adenovirus type 3 internalization is independent of primary receptors of human adenovirus type 5 and porcine adenovirus type 3.

Usefulness of adenoviral vectors derived from human adenovirus (HAd) type 5 (HAd5) is mainly limited by wide prevalence of preexisting anti-HAd5 immunity as well as non-specific tissue tropism of these vectors. As an alternative, non-human adenoviral vectors including bovine adenovirus type 3 (BAd3) are currently being investigated. Non-prevalence of BAd3 in humans and its ability to evade preexisting HAd immunity are some of the features that make BAd3 a promising vector for human gene delivery. BAd3 appears to have a tissue tropism distinct from that of HAd5 and also the repertoire of cells efficiently transduced by BAd3 is different. We performed antibody-mediated receptor blocking experiments to show that BAd3 internalization was independent of coxsackievirus-adenovirus receptor, the primary determinant of HAd5 tropism, or integrin alpha(v)beta3, a secondary molecule involved in HAd5 entry. Using homologous and heterologous knob-mediated competition assays with recombinant knobs of HAd5, porcine adenovirus type 3 (PAd3), or BAd3, we observed that BAd3 internalization was independent of the primary receptors of HAd5 and PAd3. These results provide support for further exploration of BAd3 vectors for designing targeted vectors for human gene therapy.

Perfectly matched layer in numerical wave propagation: factors that affect its performance.

The perfectly matched layer (PML) boundary condition is generally employed to prevent spurious reflections from numerical boundaries in wave propagation methods. However, PML requires additional computational resources. We have examined the performance of the PML by changing the distribution of sampling points and the PML's absorption profile with a view to optimizing the PML's efficiency. We used the collocation method in our study. We found that equally spaced field sampling points give better absorption of beams under both optimal and nonoptimal conditions for low PML widths. At high PML widths, unequally spaced basis points may be equally efficient. The efficiency of various PML absorption profiles, including new ones, has been studied, and we conclude that for better numerical efficiency it is important to choose an appropriate profile.

New method for nonparaxial beam propagation.

A new method for solving the wave equation is presented that is nonparaxial and can be applied to wide-angle beam propagation. It shows very good stability characteristics in the sense that relatively larger step sizes can be taken. An implementation by use of the collocation method is presented in which only simple matrix multiplications are involved and no numerical matrix diagonalization or inversion is needed. The method is hence faster and is also highly accurate.

Delayed embolization of Amplatzer septal occluder device: an unknown entity--a case report.

A case of delayed embolization of Amplatzer septal occluder, occurring at 2 weeks postimplantation in a 10-year-old girl with an oval-shaped secundum atrial septal defect is reported. The structurally intact device dislodged into the left atrium owing to reversal of transatrial pressure gradients and embolized to the left ventricular outflow tract from where it was retrieved surgically.

Microbial activity at gigapascal pressures.

We observed physiological and metabolic activity of Shewanella oneidensis strain MR1 and Escherichia coli strain MG1655 at pressures of 68 to 1680 megapascals (MPa) in diamond anvil cells. We measured biological formate oxidation at high pressures (68 to 1060 MPa). At pressures of 1200 to 1600 MPa, living bacteria resided in fluid inclusions in ice-VI crystals and continued to be viable upon subsequent release to ambient pressures (0.1 MPa). Evidence of microbial viability and activity at these extreme pressures expands by an order of magnitude the range of conditions representing the habitable zone in the solar system.
