Essential role for collectrin in renal amino acid transport.

Angiotensin -converting enzyme 2 (ACE2) is a regulator of the renin angiotensin system involved in acute lung failure, cardiovascular functions and severe acute respiratory syndrome (SARS) infections in mammals. A gene encoding a homologue to ACE2, termed collectrin (Tmem27), has been identified in immediate proximity to the ace2 locus. The in vivo function of collectrin was unclear. Here we report that targeted disruption of collectrin in mice results in a severe defect in renal amino acid uptake owing to downregulation of apical amino acid transporters in the kidney. Collectrin associates with multiple apical transporters and defines a novel group of renal amino acid transporters. Expression of collectrin in Xenopus oocytes and Madin-Darby canine kidney (MDCK) cells enhances amino acid transport by the transporter B(0)AT1. These data identify collectrin as a key regulator of renal amino acid uptake.

Two tobacco proline dehydrogenases are differentially regulated and play a role in early plant development.

Proline dehydrogenase is the rate-limiting enzyme in proline degradation and serves important functions in the stress responses and development of plants. We isolated two tobacco proline dehydrogenases, NtPDH1 and NtPDH2, in the course of screening for genes upregulated in stressed tobacco (Nicotiana tabacum) microspores. Expression analysis revealed that the two genes are differentially regulated. Under unstressed conditions, their steady-state transcript levels were similar in mature pollen and apical meristems, whereas NtPDH2 was expressed predominantly in vegetative organs, styles, and ovules. The expression of NtPDH1 was maintained at a constant low level during 24 h of dehydration, whereas NtPDH2 was upregulated within 1 h after the onset of stress and subsequently downregulated to undetectable levels. Differential and sustained expression was also found for the two enzymatic isoforms of Arabidopsis thaliana AtPDH. Silencing of the NtPDH genes by RNA interference using the CaMV 35S promoter led to increased proline contents, decreased seed set, delayed seed germination and retarded seedling development pointing towards an important function of at least one of the two NtPDH genes during plant reproductive development.

Structure formation and fractionation in systems of polydisperse attractive rods.

Fractionation effects and the formation of structured domains are investigated in polydisperse systems of attractive spherocylinders with the help of Monte Carlo simulations. For sufficiently high attractive interaction strength and pressure, the large rods in the system aggregate and form a highly ordered hexatic monolayer that coexists with an isotropic fluid of smaller rods. Fractionation diminishes with decreasing interaction strength but is still observed for hard rod systems, in which the large rods form a nematic droplet rather than a monolayer. Results for polydisperse systems are accompanied by phase diagrams for monodisperse systems of attractive spherocylinders. Here, the phase behavior is shown as a function of rod length and pressure.

Polydisperse particle size characterization by ultrasonic attenuation spectroscopy in the micrometer range.

The theoretical advantages of ultrasonic attenuation spectroscopy for particle size are currently not fully utilized. Especially in the region of larger particles, there is a lack of experimental confirmation of applicable models which may be used to infer particle sizes from measured attenuation spectra. With the present work, an attempt is made to supply experimental data, obtained with a commercially available ultrasonic attenuation spectrometer, and model calculations, which are based on the resonant scattering theory. It is shown that measured attenuation results for various combinations of disperse and continuous phase for both polydisperse emulsions and suspensions are reproducible by calculation. The approach is further examined for suspensions of porous particles. Here, the resonant scattering approach is combined with the Biot model for poroelasticity to obtain attenuation results with several fractions of titania aggregates, differing in particle size and pore diameter. The results indicate that the theory of resonant scattering is a valid approach if applied to particle size characterization in the large particle limit.

Changes in carbohydrate and nutrient contents throughout a reproductive cycle indicate that phosphorus is a limiting nutrient in the epiphytic bromeliad, Werauhia sanguinolenta.

BACKGROUND AND AIMS: This study examined the physiological basis of the cost of reproduction in the epiphytic bromeliad Werauhia sanguinolenta, growing in situ in a tropical lowland forest in Panama. METHODS: Entire mature plants were sampled repeatedly over the course of 2 years, which represents the common interval between reproductive events. Due to the uncertainty concerning the appropriate currency of resource allocation to reproduction, the temporal changes of the contents of total non-structural carbohydrates (TNC) and of all major nutrient elements in different plant parts were studied (stems, green leaves, non-green leaf bases, roots and reproductive structures when present). KEY RESULTS: Although TNC varied with time in all compartments, this variation was more related to seasonal fluctuations than to reproductive status. The contents of the nutrient elements, N, P, K, Mg and S, on the other hand, showed significant differences between reproductive and non-reproductive individuals, while Ca did not change with reproductive status. Differences in nutrient contents were most pronounced in stems. Seeds were particularly enriched in P, much less so in N and the other nutrient elements. Model calculations of nutrient fluxes indicate that a plant needs about 2 years to accumulate the amount of P invested in a fruit crop, while the estimated uptake rates for N were much faster. CONCLUSIONS: Since most mature individuals of this species fruit every other year, it is hypothesized that P is the prime limiting factor for reproduction. These findings therefore add to an increasing body of evidence that P rather than N is limiting growth and reproduction in vascular epiphytes.

Targeted metabolite profiling provides a functional link among eucalypt taxonomy, physiology and evolution.

Adaptation to aridity is considered a major factor in the evolution of the genus Eucalyptus. For the first time, targeted metabolite profiling has uncovered a quantitative yet discrete phytochemical link with eucalypt taxonomy. The distribution of cyclitols among Eucalyptus species, and a range of other Australian tree genera, support their proposed functions in plant tissues and provide putative links with the acclimation of trees to arid environments.

Short-term changes in carbon isotope composition of soluble carbohydrates and starch: from canopy leaves to the root system.

Changes in the 13C discrimination of current leaf photosynthesis might have profound impacts on root respiratory substrates. Therefore, the aim of this study was (1) to refine a method for the isolation of root and leaf starch and soluble sugars (neutral fraction) for stable carbon isotope analysis and (2) to assess the short-term temporal variability of the C isotope composition (delta13C) of starch and of the neutral fraction of beech roots and leaves at different canopy heights. An existing method for isolating starch for stable C isotope analysis based on enzymatic hydrolysis was modified to account for the low starch content of the samples. This was achieved by removing the enzyme (alpha-amylase) by ultrafiltration after the hydrolysis, resulting in very low carbon blanks. The neutral fraction was separated from organic acids and cations by ion-exchange chromatography. An anion-exchange resin in the [HCO3]--form was chosen that ensured high precision of C blanks. Beech leaves at 5, 10 and 20 m above the forest floor as well as roots were sampled six times during a day/night cycle in July 2003. Delta13C values of bulk material, starch and the neutral fraction increased from the lower to the higher canopy with mean differences between 5 and 20 m of 3.8, 3.4 and 2.7 per thousand for the delta13C values of starch, neutral fraction and bulk foliage, respectively. The delta13C value of foliar starch increased from the morning to the afternoon and decreased during the night, but diurnal differences (up to 3.1 per thousand) were only statistically significant for leaves sampled at 5 and 10 m height. In roots, no diurnal variation in the delta13C of starch was observed during the short time frame of one day and the delta13C of the neutral fraction did not differ between samples taken at 16:30 and 22:00. Calculated delta13C values of starch, which was mobilised during the night, were more positive than the total starch (all sampling times pooled) in leaves. Furthermore, the delta13C values of mobilised starch were approximately 5 per thousand more positive than that of the mobilised neutral fraction. Hence, the delta13C of potential sources for export from canopy leaves to roots varied considerably in their C isotope composition.

Increase in tropospheric nitrogen dioxide over China observed from space.

Emissions from fossil fuel combustion and biomass burning reduce local air quality and affect global tropospheric chemistry. Nitrogen oxides are emitted by all combustion processes and play a key part in the photochemically induced catalytic production of ozone, which results in summer smog and has increased levels of tropospheric ozone globally. Release of nitrogen oxide also results in nitric acid deposition, and--at least locally--increases radiative forcing effects due to the absorption of downward propagating visible light. Nitrogen oxide concentrations in many industrialized countries are expected to decrease, but rapid economic development has the potential to increase significantly the emissions of nitrogen oxides in parts of Asia. Here we present the tropospheric column amounts of nitrogen dioxide retrieved from two satellite instruments GOME and SCIAMACHY over the years 1996-2004. We find substantial reductions in nitrogen dioxide concentrations over some areas of Europe and the USA, but a highly significant increase of about 50 per cent-with an accelerating trend in annual growth rate-over the industrial areas of China, more than recent bottom-up inventories suggest.

Density of states of classical spin systems with continuous degrees of freedom.

In the last years different studies have revealed the usefulness of a microcanonical analysis of finite systems when dealing with phase transitions. In this approach the quantities of interest are exclusively expressed as derivatives of the entropy S=ln Omega where Omega is the density of states. Obviously, the density of states has to be known with very high accuracy for this kind of analysis. Important progress has been achieved recently in the computation of the density of states of classical systems, as new types of algorithms have been developed. Here we extend one of these methods, originally formulated for systems with discrete degrees of freedom, to systems with continuous degrees of freedom. As an application we compute the density of states of the three-dimensional XY model and demonstrate that critical quantities can directly be determined from the density of states of finite systems in cases where the degrees of freedom take continuous values.

Temperature-dependent shift from labile to recalcitrant carbon sources of arctic heterotrophs.

Soils of high latitudes store approximately one-third of the global soil carbon pool. Decomposition of soil organic matter (SOM) is expected to increase in response to global warming, which is most pronounced in northern latitudes. It is, however, unclear if microorganisms are able to utilize more stable, recalcitrant C pools, when labile soil carbon pools will be depleted due to increasing temperatures. Here we report on an incubation experiment with intact soil cores of a frost-boil tundra ecosystem at three different temperatures (2 degrees C, 12 degrees C and 24 degrees C). In order to assess which fractions of the SOM are available for decomposition at various temperatures, we analyzed the isotopic signature of respired CO2 and of different SOM fractions. The delta13C values of CO2 respired were negatively correlated with temperature, indicating the utilization of SOM fractions that were depleted in 13C at higher temperatures. Chemical fractionation of SOM showed that the water-soluble fraction (presumably the most easily available substrates for microbial respiration) was most enriched in 13C, while the acid-insoluble pool (recalcitrant substrates) was most depleted in 13C. Our results therefore suggest that, at higher temperatures, recalcitrant compounds are preferentially respired by arctic microbes. When the isotopic signatures of respired CO2 of soils which had been incubated at 24 degrees C were measured at 12 degrees C, the delta13C values shifted to values found in soils incubated at 12 degrees C, indicating the reversible use of more easily available substrates. Analysis of phospholipid fatty acid profiles showed significant differences in microbial community structure at various incubation temperatures indicating that microorganisms with preference for more recalcitrant compounds establish as temperatures increase. In summary our results demonstrate that a large portion of tundra SOM is potentially mineralizable.

[The reliability of stereotaxy in diagnosis of intracranial space occupying lesions].

A total of 79 patients with a suspect space occupying intracranial lesion was operated stereotactically. After CT and MR image acquisition, entry and target coordinates were defined and biopsy trajectories were simulated preoperatively using a special planning software. Biopsy specimens allowed a satisfactory neuropathological examination and diagnostic result in 74 of 79 (93.7%) cases. In the remaining 5 of 79 (6.3%) patients, a glioses was diagnosed in three cases; in only two patients could no pathological process be proved. 10 of 79 (12.7%) patients showed an intraoperative bleeding out of the biopsy-cannula during serial stereotaxy, which was associated with a CT-detectable hematoma up to 7 mm in diameter in only three cases (3.8%). In no patient was the detection of intracerebral bleeding the reason for any neurological deficit. In summary, the high percentage of satisfactory neuropathological diagnoses, the low rate of stereotaxy-associated morbidity and the comfortable and safe use of computer and stereotactic devices justify this kind of minimally invasive diagnostic measure as a routine method.

A study of the trace gas columns of O3, NO2 and HCHO over Africa in September 1997.

Retrievals of trace gas columns from the measurements of backscattered radiation by GOME (Global Ozone Monitoring Experiment) show that enhanced tropospheric columns of ozone (O3), nitrogen dioxide (NO2) and formaldehyde (HCHO), over the African continent occur frequently. This study focuses on the behaviour of trace gases over Africa in September 1997, a period impacted by the strongest known El Niño phase of the ENSO. It investigates our qualitative and quantitative understanding of the retrieved tropospheric trace gas column densities. The emissions of NOx and volatile organic compounds (VOC) from biomass burning, biogenic sources and lightning and their photochemical transformation have been investigated. By performing a trajectory analysis, the transport of air masses from the different emission regions was analysed and the potential atmospheric spatial distribution determined. BRemen's Atmospheric PHOtochemical model (BRAPHO) was applied to compute the chemistry along a large number of trajectories. From these results, tropospheric column amounts of O3, NO2 and HCHO were derived. Tropospheric trace gas columns retrieved from GOME measurements and those calculated are in reasonable agreement. Their general spatial extent was similar in the lower troposphere but the modeled trace gas columns in the upper troposphere were located south of the retrieved columns. We attribute this behaviour to uncertainties in the ERA-40 meteorological data in the upper troposphere. The significance of biomass burning and of biogenic emissions with respect to HCHO columns over Africa was investigated. The analysis reveals that the total amounts of HCHO generated over Africa during September 1997 as a result of biomass burning and biogenic emissions are similar. However the HCHO from biogenic sources has the highest specific columns and these are located close to their source. In comparison the HCHO from biomass burning is predicted to be produced and transported over a much wider area. Overall all the emission processes mix together to produce the plume of O3.

Microbial and immunological responses relative to high-altitude exposure in mountaineers.

PURPOSE: High-altitude exposure is often associated with gastrointestinal disorders, inflammation, and an increased risk of infection. We suspected microbial and immunological responses to high-altitude exposure in mountaineers resulting from changes in the balance of the intestinal microflora. METHODS: We investigated fecal samples and serum of seven mountaineers who took part in a 47-d German expedition to the Nepalese Himalayas in 2002, for microbial response by changes in different fecal bacterial population groups (fluorescent in situ hybridization), immune response by serum levels of IgG-, IgM-, and IgA anti-LPS (E. coli J5), and inflammatory response by serum levels of C-reactive protein (CRP) (ELISA). In addition, measurements of body temperature, pulse rate, arterial oxygen saturation, and questionnaire (Lake Louise score for altitude illness) were performed. RESULTS: The data indicate a distinct alteration in the composition of the fecal microflora relative to high-altitude exposure above 5000 m. Bifidobacteria and species belonging to the Atopobium, Coriobacterium, and Eggerthella lenta group decreased, whereas potential pathogenic bacteria of the gamma subdivision of Proteobacteria and specific Enterobacteriaceae such as Escherichia coli increased. Possible endotoxemia resulting from the increase of the latter Gram-negative bacteria was indirectly indicated by the reduction in serum levels of IgM- and/or IgA anti-LPS. CRP was elevated relative to high-altitude exposure. The Lake Louise score correlated with the changes in CRP, IgA-, and IgM anti-LPS but did not correlate with the bacterial alterations. CONCLUSION: Changes in the composition of intestinal microbiota may be associated with indicators of an immunological challenge and may result in an increased health risk of mountaineers during exposure to very high altitude.

Retrieval of profile information from airborne multiaxis UV-visible skylight absorption measurements.

A recent development in ground-based remote sensing of atmospheric constituents by UV-visible absorption measurements of scattered light is the simultaneous use of several horizon viewing directions in addition to the traditional zenith-sky pointing. The different light paths through the atmosphere enable the vertical distribution of some atmospheric absorbers, such as NO2, BrO, or O3, to be retrieved. This approach has recently been implemented on an airborne platform. This novel instrument, the airborne multiaxis differential optical absorption spectrometer (AMAXDOAS), has been flown for the first time. In this study, the amount of profile information that can be retrieved from such measurements is investigated for the trace gas NO2. Sensitivity studies on synthetic data are performed for a variety of representative measurement conditions including two wavelengths, one in the UV and one in the visible, two different surface spectral reflectances, various lines of sight (LOSs), and for two different flight altitudes. The results demonstrate that the AMAXDOAS measurements contain useful profile information, mainly at flight altitude and below the aircraft. Depending on wavelength and LOS used, the vertical resolution of the retrieved profiles is as good as 2 km near flight altitude. Above 14 km the profile information content of AMAXDOAS measurements is sparse. Airborne multiaxis measurements are thus a promising tool for atmospheric studies in the troposphere and the upper troposphere and lower stratosphere region.

myo-Inositol and sucrose concentrations affect the accumulation of raffinose family oligosaccharides in seeds.

Raffinose family oligosaccharides (RFOs) fulfil multiple functions in plants. In seeds, they possibly protect cellular structures during desiccation and constitute carbon reserves for early germination. Their biosynthesis proceeds by the transfer of galactose units from galactinol to sucrose. Galactinol synthase (GolS), which mediates the synthesis of galactinol from myo-inositol and UDP-galactose, has been proposed to be the key enzyme of the pathway. However, no significant relationship was detected between the extractable GolS activity and the amount of RFOs in seeds from seven pea (Pisum sativum L.) genotypes selected for high variation in RFO content. Instead, a highly significant correlation was found between the levels of myo-inositol and RFOs. Moderately strong relationships were also found between sucrose and RFO content as well as between myo-inositol and galactinol. Further evidence for a key role of myo-inositol for the synthesis of galactinol was obtained by feeding exogenous myo-inositol to intact pea seeds and by the analysis of four barley (Hordeum vulgare L.) low phytic acid mutants. In seeds of three of these mutants, the reduced demand for myo-inositol for the synthesis of phytic acid (myo-inositol 1,2,3,4,5,6-hexakisphosphate) was associated with an increased level in myo-inositol. The mutants seeds also contained more galactinol than wild-type seeds. The results suggest that the extent of RFO accumulation is controlled by the levels of the initial substrates, myo-inositol and sucrose, rather than by GolS activity alone.
