Novel syntheses of chiral beta- and gamma-amino acid derivatives utilizing N-protected (aminoacyl)benzotriazoles from aspartic and glutamic acids.

Friedel-Crafts reactions of N-protected (alpha-aminoacyl)benzotriazoles with hetero- and benzenoid- aromatics give alpha-amino ketones that can be reduced by either triethyl silane or sodium borohydride to form the corresponding beta- and gamma-amino acid derivatives. The preservation of chirality throughout this process is confirmed by chiral HPLC results.

Structural elucidation of nitro-substituted five-membered aromatic heterocycles utilizing GIAO DFT calculations.

The GIAO (Gauge Including Atomic Orbitals) DFT (Density Functional Theory) method is applied at the B3LYP/6-31+G(d,p)//B3LYP/6-31+G(d), B3LYP/6-311++G(d,p)//B3LYP/6-31+G(d), B3LYP/6-311+G (2d,p)//B3LYP/6-31+G(d) and B3LYP/6-311++G(d,p)//B3LYP/6-311++G(d,p) levels of theory for the calculation of proton and carbon chemicals shifts and coupling constants for 25 nitro-substituted five-membered heterocycles. Difference (1D NOE) spectra in combination with long-range gHMBC experiments were used as tools for the structural elucidation of nitro-substituted five-membered heterocycles. The assigned NMR data (chemical shifts and coupling constants) for all compounds were found to be in good agreement with theoretical calculations using the GIAO DFT method. The magnitudes of one-bond (1JCH) and long-range (nJCH, n>1) coupling constants were utilized for unambiguous differentiation between regioisomers of nitro-substituted five-membered heterocycles.

QSAR modeling of anti-invasive activity of organic compounds using structural descriptors.

The anti-invasive activity of 139 compounds was correlated by an artificial neural network approach with descriptors calculated solely from the molecular structures using CODESSA Pro. The best multilinear regression method implemented in CODESSA Pro was used for a pre-selection of descriptors. The resulting nonlinear (artificial neural network) QSAR model predicted the exact class for 66 (71%) of the training set of 93 compounds and 32 (70%) of validation set of 46 compounds. The standard deviation ratios for the both training and validation sets are less than unity, indicating a satisfactory predictive capability for classification of the nature of the anti-invasive activity data. The proposed model can be used for the prediction of the anti-invasive activity of novel classes of compounds enabling a virtual screening of large databases of anticancer drugs.

Synthesis of mono- and symmetrical di-N-hydroxy- and N-aminoguanidines.

Novel mono- and symmetrical di-N-hydroxy- and N-aminoguanidines were readily prepared from the reaction of diverse hydroxylamines or hydrazines with reagent classes di(benzotriazol-1-yl)methanimine 6, (bis-benzotriazol-1-yl-methylene)amines 8a,b, benzotriazole-1-carboxamidines 10a-i, benzotriazole-1-carboximidamides 11a,b, and N'-hydroxy-1H-1,2,3-benzotriazole-1-carboximidamide 18. The preparation is described for a variety of N-hydroxy- and N-aminoguanidines with different substitution patterns in good yields.

QSAR study of antiplatelet agents.

A QSAR methodology that involves multilinear (Hansch-type) and nonlinear (ANN backpropagation) approaches was developed to correlate the antiplatelet activity of 60 benzoxazinone derivatives against factor Xa. The statistical characteristics provided by multilinear model (R2 = 0.821) indicated satisfactory stability and predictive ability, while the ANN predictive ability is somewhat superior (R2 = 0.909). The multilinear model provided insight into the main factors that modulate the inhibitory activity of the investigated compounds.

Efficient microwave assisted access to chiral O-(alpha-protected-aminoacyl)steroids.

Chiral O-(alpha-protected-aminoacyl)steroids 4a-f, 6a-b, 8 and 4a+4d and O-(alpha-protected-dipeptidoyl)steroids 12a,b are conveniently prepared under microwave irradiation in isolated yields of 65-96%, with complete chirality retention. The reaction utilized readily available N-(Z-alpha-aminoacyl)benzotriazoles 2a-f and Z-dipeptidoylbenzotriazole 11, with naturally occurring steroidal alcohols 3,5,7,9.

In search of ionic liquids incorporating azolate anions.

Twenty-eight novel salts with tetramethyl-, tetraethyl-, and tetrabutylammonium and 1-butyl-3-methylimidazolium cations paired with 3,5-dinitro-1,2,4-triazolate, 4-nitro-1,2,3-triazolate, 2,4-dinitroimidazolate, 4,5-dinitroimidazolate, 4,5-dicyanoimidazolate, 4-nitroimidazolate, and tetrazolate anions have been prepared and characterized by using differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), and single-crystal X-ray crystallography. The effects of cation and anion type and structure on the physicochemical properties of the resulting salts, including several ionic liquids, have been examined and discussed. Ionic liquids (defined as having m.p.<100 degrees C) were obtained with all combinations of the 1-butyl-3-methylimidazolium cation ([C(4)mim](+)) and the heterocyclic azolate anions studied, and with several combinations of tetraethyl or tetrabutylammonium cations and the azolate anions. The [C(4)mim](+) azolates were liquid at room temperature exhibiting large liquid ranges and forming glasses on cooling with glass-transition temperatures in the range of -53 to -82 degrees C (except for the 3,5-dinitro-1,2,4-triazolate salt with m.p. 33 degrees C). Six crystal structures of the corresponding tetraalkylammonium salts were determined and the effects of changes to the cations and anions on the packing of the structure have been investigated.

Skin permeation rate as a function of chemical structure.

Multilinear and nonlinear QSAR models were built for the skin permeation rate (Log K(p)) of a set of 143 diverse compounds. Satisfactory models were obtained by three approaches applied: (i) CODESSA PRO, (ii) Neural Network modeling using large pools of theoretical molecular descriptors, and (iii) ISIDA modeling based on fragment descriptors. The predictive abilities of the models were assessed by internal and external validations. The descriptors involved in the equations are discussed from the physicochemical point of view to illuminate the factors that influence skin permeation.

Combustible ionic liquids by design: is laboratory safety another ionic liquid myth?

The non-flammability of ionic liquids (ILs) is often highlighted as a safety advantage of ILs over volatile organic compounds (VOCs), but the fact that many ILs are not flammable themselves does not mean that they are safe to use near fire and/or heat sources; a large group of ILs (including commercially available ILs) are combustible due to the nature of their positive heats of formation, oxygen content, and decomposition products.

Correlation of blood-brain penetration using structural descriptors.

Experimental blood-brain partition coefficients (logBB) for a diverse set of 113 drug molecules are correlated with computed structural descriptors using CODESSA-PRO and ISIDA programs to give statistically significant QSAR models based respectively, on molecular and on fragment descriptors. The linear correlation CODESSA-PRO five-descriptor model has correlation coefficient R2=0.781 and standard deviation s2=0.123. The 'consensus model' of ISIDA gave R2=0.872 and s2=0.047. The developed models were successfully validated using the central nervous system activity data of an external test set of 40 drug molecules.

QSAR modeling of the inhibition of glycogen synthase kinase-3.

Quantitative structure-activity relationship (QSAR) models of the biological activity (pIC50) of 277 inhibitors of Glycogen Synthase Kinase-3 (GSK-3) are developed using geometrical, topological, quantum mechanical, and electronic descriptors calculated by CODESSA PRO. The linear (multilinear regression) and nonlinear (artificial neural network) models obtained link the structures to their reported activity pIC50. The results are discussed in the light of the main factors that influence the inhibitory activity of the GSK-3 enzyme.

Direct synthesis of esters and amides from unprotected hydroxyaromatic and -aliphatic carboxylic acids.

A facile method for the activation of hydroxy-substituted carboxylic acids using benzotriazole chemistry without prior protection of the hydroxy substituents is presented. The N-acylbenzotriazole intermediates 2a-g, 6a-d, and 9a-c have been used for high-yielding synthesis of both aliphatic (3a-l) and aromatic (7a-h, 10a-f) hydroxy carboxamides. High yields of aromatic hydroxy esters 12a-h and 13a-i were obtained using either neat alcohols in neutral microwave conditions or nucleophilic alkoxides and the intermediate N-(arylacyl)benzotriazoles. Moderate yields were obtained in the case of aliphatic hydroxy esters 11a,b and thiolesters 11e-g from the intermediates 2a-c.

Efficient microwave access to polysubstituted amidines from imidoylbenzotriazoles.

Microwave reactions of primary and secondary amines with imidoylbenzotriazoles 6a-w gave diversely substituted amidines 7a-Aa in 76-94% yields. Convenient preparations of a variety of amides 5a-Ab (87-96%) and imidoylbenzotriazoles 6a-w (56-95%) have also been developed using microwave irradiation under mild conditions and short reaction times. These results demonstrate further the advantages of microwave synthesis and introduce a new application of imidoylbenzotriazoles in the preparation of polysubstituted amidines.

QSAR study of mosquito repellents using Codessa Pro.

Protection times provided by 31 synthetic repellents against Aedes aegypti mosquitoes were correlated with the chemical structures of these repellents using Codessa Pro software. Two statistically significant quantitative models with R2 values of ca. 0.80 are presented and discussed.

Antimalarial activity: a QSAR modeling using CODESSA PRO software.

A quantitative structure-activity relationship (QSAR) modeling of the antimalarial activity of two diverse sets of compounds for each of two strains D6 and NF54 of Plasmodium falciparum is presented. The molecular structural features of compounds are presented by molecular descriptors (geometrical, topological, quantum mechanical, and electronic) calculated using the CODESSA PRO software. Satisfactory multilinear regression models were obtained for data sets of the D6 and NF54 strains, with R2 = 0.84 and 0.89, respectively. The models were also satisfactorily validated internally. The descriptors involved in these equations were related to the mechanism of antimalarial protection.

Selective peptide chain extension at the N-terminus of aspartic and glutamic acids utilizing 1-(N-protected-alpha-aminoacyl)benzotriazoles.

Diverse N-protected di-(3a-d, 3a + a', 5a-d, 5d + d', and 7a-g) and tripeptides (10a-h) were synthesized under mild reaction conditions in good yields (65-97%) by acylation with 1-(N-protected-alpha-aminoacyl)benzotriazoles of the amino groups of free aspartic and glutamic acids. Complete retention of chirality was demonstrated by parallel experiments involving D-Ala, L-Ala, and DL-Ala for the preparation of dipeptides and tripeptides, and supported by NMR and HPLC analyses.
