Isolation and chemical properties of a haptenic substance from buckwheat dialysate.

A haptenic substance was isolated from the dialysate of the aqueous extract of buckwheat. This substance, BWD II 22-3, which was composed of Asp(1), Thr(1), Ser(1), Gly(1), Gly(4), Ala(1), Val(1), Leu(1), Orn(2), Lys(1), Arg(1), Cys(1) and glucose was demonstrated to be homogeneous by gel filtration and paper electrophoretic analyses. The molecular weight was estimated to be 1600 by gel filtration on Sephadex G-50. The haptenic substance apparently has-SH group determinant caused about 50% inhibition at a concentration of 100 micrograms/disc in the RAST procedure using human serum sensitive to buckwheat. An active component, BWD II 22-3, might prove effective in the hyposensitization therapy of buckwheat-sensitive patients.

Immunopharmacological studies of the aqueous extract of Cinnamomum cassia (CCAq). II. Effect of CCAq on experimental glomerulonephritis.

Effect of the aqueous extract of Cinnamomum Cassia (CCAq) on experimental glomerulonephritis was studied and compared with that of cobra venom factor (CoVF). In rat nephrotoxic serum (NTS) nephritis, CCAq clearly inhibited the excretion of protein into the urine and the increase of peripheral leucocyte counts. The histological score in CCAq administered animals was significantly lower than that in control animals. However, CCAq did not inhibit or lower the serum complement level. Contrary to CCAq, hypocomplementation was observed by the administration of CoVF, and the excretion of protein into urine was inhibited in a high dose group. In immune complex (IC) and autologous IC nephritis in rats, CCAq clearly inhibited the excretion of protein into urine and the elevation of blood urea nitrogen (BUN). The administration of CoVF caused hypocomplementation, but little inhibition of the excretion of urinary protein was observed in both types of immune complex nephritis. The histological score was slightly inhibited by a low dose of CCAq and a high dose of CoVF. In the experiment employing NZB/NZW F1 mice, the proteinurea, the elevation of BUN level, and the production of antibodies were clearly inhibited by the administration of CCAq. Similar inhibition was observed by CoVF at a high dose. However, the histological changes of the kidney in NZB/NZW F1 mice were not prevented by the administration of CCAq or CoVF.

Information-processing load and variability of visual evoked potentials.

5 subjects were assigned two tasks during which they were simultaneously presented brightness-changes of the spot light on the CRT and the speech in a foreign language. They were asked to count the number of the changes in the visual task and listen to the speech to discern the content in the auditory task. A pair of averaged visual evoked potentials (VEP) to the brightness-changes were obtained for each task to calculate a subset correlation as an index of the variance of VEP. The mean correlation coefficient of VEP across the subjects was significantly higher during the visual task than that in the auditory task. This means that VEP was more stable during the visual task. The result suggests that the subset correlation measure might be available to evaluate visual information-processing load.

Diagnostic evaluation of CT and ERCP based on a retrospective analysis of hepato-biliary and pancreatic diseases.

A retrospective analysis was performed in order to evaluate diagnostic accuracies by computed tomography (CT) and by endoscopic retrograde cholangiopancreatography (ERCP). Materials studied were 67 lesions out of 56 cases with hepato-biliary and pancreatic diseases confirmed mainly at surgery. Diagnostic accuracy of the CT & ERCP for the hepato-biliary lesions was 59.5% & 86.5% and that of CT & ERCP for the pancreatic lesions was 60% & 80% respectively. CT scan is useful for diagnosing abscess, cyst or tumors. On the contrary, lesions which invade the hepato-biliary tract or pancreatic duct can be readily diagnosed by ERCP examination. ERCP is less useful for the diagnosis of parenchymatous lesions or infiltration of lesions into the surrounding organs. Therefore, the combined use of both CT and ERCP is important for the diagnosis of hepato-biliary or pancreatic disease.

Visual signal detection and lambda responses.

On the basis of the result in earlier studies that the lambda response was associated with offset of the saccade, e.g., onset of the fixation pause, the relationship between the lambda response and the visual information processing load was examined. Eleven subjects were presented simultaneously random patterns including visual signals on Goldman perimeter and tone pips including auditory signals at random. In the visual task, the subject was instructed to search on the random pattern and detect the visual signals. In the auditory task, he was instructed to listen to tone pips without ceasing eye movements and detect the tone signals. EEGs from Oz and Cz, and the vertical and the horizontal EOGs were recorded. The vector EOG was calculated from both EOGs for detecting the absolute value of the saccade size. The occipital EEGs time-locked to offset of saccades were averaged 100 times to obtain lambda responses. The vertex EEGs time-locked to tone signals were averaged 100 times to obtain vertex potentials to auditory stimuli (AEP). Amplitudes of the lambda response (NOP100 and P100-N180) in the visual task were significantly higher than in the auditory task. Amplitudes of AEP (N1 and N1-P2) in the auditory task were enhanced significantly more than in the visual task. The results showed that the lambda response varied with information-processing load. The lambda response can be applicable as an index of visual information processing in eye movement situations.

Study on cases of nonvisualized cholecystogram by the use of ERCP.

We used Endoscopic Retrograde Cholangiopancreatography (ERCP) in 77 patient in whom oral cholecystography failed to visualize the gallbladder. ERCP did not visualize the gallbladder in 42%, and poorly visualized it in 58%, however, there was no significant difference in the disease distribution between non- and poorly visualized cases. 63 of the 77 patients underwent surgery. In cases postoperatively diagnosed as gallbladder carcinoma, the prognosis was better in poorly than in non-visualized gallbladder cases. Non- or poor visualization by ERCP in operated cases was due to gallstones in 71%, to common duct stones with intrahepatic calculi in 22%, and to malignant tumors in 17.5%. Our findings suggest that non-visualization of the gallbladder by ERCP should alert the diagnostician to the possibility of malignancy, especially in patients over 50 years of age.

Lambda waves associated with offset of saccades: a subject with large lambda waves.

The temporal relationship between the lambda wave and the saccade was investigated in a subject with large amplitude lambda waves. The peak latency of the lambda wave from onset of the saccade was increased in proportion to increase of the saccade duration while the latency from offset was constant. The result showed that the lambda wave was associated with offset of the saccade rather than onset of it. It is suggested that the lambda wave might be the evoked response elicited by the afferent inflow beginning a fixation.

Studies on aculeacin. I. Isolation and characterization of aculeacin A.

Aculeacin A, a new antifungal antibiotic was isolated from the mycelial cake of Aspergillus aculeatus M-4214. The antibiotic is a white amorphous powder soluble in lower alcohols and hardly soluble in other organic solvents or water. Aculeacin A gave palmitic acid and five ninhydrin-positive products including theonine, hydroxyproline upon acid hydrolysis. The antibiotic showed a potent activity against molds and yeasts, but exhibited no antibacterial activity. Aculeacin A has relatively low toxicity in mice.

Studies on aculeacin. II. Isolation and characterization of aculeacins B, C, D, E, F and G.

Six new antibiotics were isolated as the minor components related to aculeacin A from the culture broth of Aspergillus aculeatus M-4214 and named as aculeacins B, C, D, E, F and G. Their physico-chemical properties were analogous to those of aculeacin A and they showed significant activity against fungi. All of the minor components liberated palmitic acid on alkaline hydrolysis. Amino acid analysis showed that threonine and hydroxyproline are common constituents of aculeacins.
