Possible molecular mechanisms of ageing.

While the error theory of ageing has attracted most interest in recent times it cannot yet be regarded as being demonstrated. Posttranslational modifications of proteins genetic theory appears loical but has little in vivo evidence to prove it. Basic mechanisms of ageing probably involve the interaction of several processes.

[Seroepidemiological investigations on the incidence of Brucella canis antibodies in man (author's transl)].

1915 serum samples of humans were examined in the serum tube agglutination test for antibodies to Brucella canis. 1400 random samples were from blood-donors, 480 samples from clinical patients and 35 samples from persons, exposed to dogs and material infected with Br. canis. Agglutinating antibodies to Br. canis antigen were detected in 6 sera (titers 1 X 1:400, 2 X 1:200, 3 X 1:100). In 3 cases positive results were confirmed by complement fixation and indirect immunofluorescence tests. All serum samples with agglutinating antibodies were from healthy blood-donors. Serum samples from patients, and from 7 veterinary surgeons, 9 laboratory assistants and 19 animal care-takers had no agglutinating antibodies to Br. canis in a serum dilution of 1:50.

[Differentiation of Brucella by acrylamide-gel electrophoresis (author's transl)].

The strains of Brucella (Tab. 1) were grown on Tryptose-blood-agar at 37 degrees C, and the strains of Bordetella, Pasteurella and Actinobacillus on blood-agar. After 24 or 48 h they were harvested and extracted in phenol-acetic acid-water solution (4:2:1) (1 ml/50 mg bacterial wet weight) at 4 degrees C over 48 h. After centrifugation (8000 X g, 1 h) 2 volumes of the supernatant were mixed with 1 volume of a 40% sucrose solution in 35% acetic acid. Different volumes (0.20, 0.15 or 0.10 ml) of this were added to 7.5% acrylamide gel (2 ml in tubes 6 X 100 mm), containing 5 M urea and 12% acetic acid. The solution in both electrode chambers was 10% acetic acid. During the first 15 min it was focused with 2 mA/tube, then it was separated for 3 or 5 h with 4 mA/tube. The protein bands were stained with amido black 10B. All species could be exactly differentiated from each other, but the protein bands of biotypes of Bruc. suis, Bruc. melitensis and Bruc. abortus were identical in each case. All investigated strains of Bruc. canis were identical too. A small relationship was noticed between Bruc. canis and Bruc. suis and between Bordetella bronchiseptica and Bruc. canis respectively Bruc. suis strains.

[Seroepidemiological investigations on the prevalence of rickettsial antibodies in man in the Federal Republic of Germany (author's transl)].

1,600 random serum samples from inhabitants of Hesse were investigated for rickettsial antibodies by complement fixation test. Antibodies were found in 19 samples (1,2%) against RMSF-group antigen with titers between 1:10 and 1:640. These results confirm recent reports on natural foci of such rickettsial infections in the Federal Republic of Germany. Antibodies against rickettsia of the Typhus-group or Coxiella burnetii could not be detected by the complement fixation reaction. None of the sera reacted with significant titer in the Weil-Felix-Test.

[Ultrastructural investigations on anionic surface sites of Brucella canis (author's transl)].

Anionic sites on the surface of Brucella canis were visualized in the electron microscope by staining with positively charged ferric oxide hydrosols in acetic acid (AI-reagent), or propanoic acid (PI-reagent), and with a polycationic ferritin derivative. With the AI-reagent, single or small aggregates of ferric oxide particles were bound to the cell surface of Br. canis, whereas, with the lipophilic PI-reagent, the microorganisms were heavily stained with focal aggregates of iron granules. The polycationic ferritin label was uniformly distributed over the entire cell surface of Br. canis. The ferritin label was not bound on the surface of the organisms after prior treatment with trichloroacetic acid or methanolic hydrochloric acid. Treatment with aqueous acetone, chloroform/methanol, diethyl ether, sodium deoxycholate, pronase, lysozyme, hyaluronidase, and sodium periodate neither influenced the morphology of the Brucella nor diminished their ionic binding sites. Our results indicate that the anionic sites on the cell surface of Br. canis may be carboxyl and phosphate groups of lipopolysaccharides.

[Bronchoplastic methods in the resection treatment of malignant bronchial tumors].

1548 patients who were hospitalized 1964--1975 for diagnosis and treatment of bronchogenic carcinoma, 779 underwent resection. 17 patients could be operated by lobectomy or bilobectomy and bronchial resection (sleeve resection). Postoperative complications were frequent (n = 8): 4 times bronchopleural fistula, 2 times empyema, once fatal pneumonia and once bronchial stenosis. The overalll mortality and the survival rates are comparable to those of patients with radical resections. Sleeve resection is therefore a suitable alternative to pneumonectomy in elderly patients with reduced pulmonary function, rarely indicated also by a favourable tumor size. Sleeve resection increases the resectability of malignant bronchogenic tumors by 2%. Methods to prevent or cure the postoperative complications consisted in the use of absorbable suture material and long lasting intrathoracic suction.

[Operative mortality, mortality and late results in bronchial carcinoma].

457 patients with a bronchial carcinoma of the years 1947--1962 are compared with 126 patients of the years 1969--1970 and set up in comparison to the whole clinical material of the years 1960--1975. In the period between 1969 and 1970 radical resections were performed in 20% more patients than in the first period (1947--1962). Older patients were in the last period (1970--1974) more frequent. 8% of all patients in which lobectomy was performed and 5.5% of the patients in which pneumonectomy was performed were older than 70 years of age. Overall mortality has remained unchanged for exploratory thoracotomy (19%), palliative resection (17.5%) and "radical" resection (14%). For simple pneumonectomy the overall mortality went down from 19.2% (1960--1964) to 9.6% (1970-1974) and for simple lobectomy from 14.4% to 3.8%. The five-year survival rate after radical resections was 8% (first period 0%) and after simple resections 26% (26.2%). Patients with stage I epidermoid carcinoma had a five-year survival rate of 46%. Those with stage II (spread to ipsilateral hilar nodes) a five-year survival rate of 14.3% Patients with palliative resections and exploratory thoracotomy died within 16 months.

[Osteotomy of the pelvis (Chiari) (author's transl)].

Judging by 203 cases the pelvic displacement osteotomie by Chiari has proven to be a very effective method to correct the deficient femoral head cover of the dysplastic acetabulum, in many cases however it is difficult to normalize the roof angle. We examined 51-joints in which the pelvic osteotomie by Chiari had been performed just before and particularily after growth had come to an end, in view of the prophylaxis of arthrosis of this operation. Hereby we observed that development or increase of anthrotic signs on an average of 10 1/2 years was less frequent than could be expected without the osteotomy, the clinical condition remaining approximately the same as before the operation, but nevertheless did not deteriorate.

[Investigations of amino acid metabolism by thin layer chromatography for differentiation of Brucella (author's transl)].

The amino acid metabolism of 23 different Brucella strains was investigated for differentiation purposes. The results were evaluated by thin layer chromatography, after enzymatic incubation. The organisms (Tab. 1) were grown on Tryptose blood agar at 37 degree C for 24 or 48h. Two mg wet weight of bacteria in 0.2 ml PBS, 0.01 M, were incubated with 12.5 microng (0.025 ml) amino acid in small tubes for 16h at 37 degree C, and centrifuged for 15 min at 7500 X g. For controls, bacterial suspensions were heated for 15 min at 100 degree C to destroy enzymatic activity, and also contrifuged for 15 min at 7500 X g. Usually 4 micronl of the supernatant fluids (6 micronl for L-asparagine, and 10 micronl for L-proline) were pipetted on the thin layer plate. The tests were run in n-butanol acetic acid water, 20:5:5, with a distance of 8 cm. Amino acids were stained with ninhydrine. The tests were repeated 3-5 times with identical results. Amino acid metabolism was indicated by different staining intesities (+ to ++) in comparison to control preparations. All species could be exactly differentiated from each other, with the exception of B. suis, biotype 2, and B canis, which could not be differentiated by their amino acid metabolism. Biotyps of the same species were mostly identical. The results of these investigations could be reproduced qualitatively as well as quantitatively. The method described is recommended for routine investigations.
