In order to elucidate a predisposition for so-called autoimmune thyroid disorders such as Graves' disease, chronic thyroiditis and myxedema, the incidence of thyroid antibodies was studied in relatives of patients with thyroid disorders. The relatives studied were all limited within a two degree relationship of the patients. Serum antibodies to thyroglobulin and thyroid microsome were determined by susing the method of a hemagglutination test. The following results were obtained; 1) Thirty six of 103 relatives (35%) were found to have such antibodies, and sixteen (7.0%) of 230 hospital controls were positive. This difference was significant (P less than 0.001). 2) There was no difference in the incidence among relatives of patients with Graves' disease and those chronic thyroiditis. 3) With regard to age, the incidence in the more than 20 years old group was significantly greater than that in the under 19 years old group. 4) There was no difference in the incidence between male and female. These results suggested that the relatives of patients with so-called autoimmune thyroid disorders may have a predisposition for thyroid disorders.

HGH secretion in response to TRH was studied in patients with cerebrovascular disease in order to elucidate an influence of cerebrovascular lesions on the hypothalamus-anterior pituitary function. Blood specimens were obtained before and at the time of 10, 20, 30, 40, 60, 90 and 120 minutes after the intraveneous administration of 200 microgram of TRH in 17 patients with cerebral hemorrhage and in 8 patients with cerebral infarction. With regards to the natural fluctuations of serum HGH caused by cerebrovascular disease, the blood specimens were obrained at the time of 0, 30, 60, 90 and 120 minutes without the administration of TRH in 4 patients with cerebral hemorrhage and in 4 patients with cerebral infarction. Serum HGH was measured using the RIA method and the following results were obtained: 1) An increase in serum HGH was observed in 1 patient with cerebral hemorrhage out of 8 patients with cerebrovascular disease without TRH administration. 2) In 5 out of 17 patients with cerebral hemorrhage, an increase in serum HGH was observed following TRH administration. Three out of these 5 patients were included in the group of non-coma, and 4 out of the 5 patients were female. 3) In 2 out of 8 patients with cerebral infarction, an increase in serum HGH was observed following TRH administration.

A rapid and specific method of high-pressure liquid chromatography for determining 6-(1-methyl-4-nitro-5-imidazole) mercaptopurine (azathioprine) in the blood was developed using mu Bondapak C18 as adsorbent and 30% aquous methanol as eluent. Azathioprine was completely separated from related compounds in blood. The standard curve showed a linear relationship at least between 0.6 and 30 micrograms per ml of serum. Recovery was satisfactory and the analytical time needed was less than 20 min. Disappearance rates of azathioprine from circulating blood in rats were 0.026 in probenecid-treated rats, and 0.103 in intact rats.
Three patients at various stages of remission from leukemia died following the development of massive liver necrosis within only 4-6 days. All had either hepatitis B surface antigen or antibody in their sera, and two of them experienced severe epigastric pain before the onset of liver injury. Hepatitis B surface antigen appeared in two of these patients after remission from leukemia. Serum gamma-globulin levels increased with decreasing doses of prednisolone and other antileukemic drugs, and hepatic cell necrosis occurred extensively. Localization of hepatitis B surface antigen in their livers revealed a strong positive reaction in the phagocytic cells. These observations strongly suggest that hepatitis B virus may be causally related to the fulminant hepatic failure at least in two of the reported leukemic patients.
Albumin was highly purified from a commercially available rat albumin preparation (Fraction V) using disc electrophoresis. The purified albumin had the same antigenicity as Fraction V. A monospecific anti-rat albumin rabbit serum was obtained. The antiserum was used in a double antibody quantitative method for determining rat albumin.
A reproducible animal model of fulminant hepatic failure was developed by intraperitoneal administration of D-galactosamine hydrochloride to Sprague-Dawley rats. Biochemical and morphological hepatic injury and brain edema resembled human fulminant hepatic failure. This model would facilitate further studies of the pathogenesis of brain dysfunction and evaluation of treatment in fulminant hepatic failure.
Sprague-Dawley rats given azathioprine in the diet for 3 to 4 weeks developed severe liver damage. Elevations of serum alkaline phosphatase and gamma-glutamyl transpeptidase activities were associated with increased hepatic glucose 6-phosphate dehydrogenase levels and decreased liver glucose 6-phosphatase activities, i.e., conditions which were commonly observed in various hepatotoxin-induced liver injuries. Light and electron microscopic observations revealed centrolobular necrosis with large scars and the proliferation of the mitochondria and rough endoplasmic reticulum. This model could be used to study the mechanisms of azathioprine-induced liver damage and its prevention.

The disappearance rates(K) of FT-207 from the blood in patients with primary hepatoma and advanced cirrhosis of the liver were significantly lower than those in control patients with cancer but normal liver function. Pretreatment with tocopheryl nicotinate and indomethacin increased the K values in the control subjects, but was without effect on the K values in patients with primary hepatoma.
Silicone rubber is conventionally used as a lining soft material for denture base. However it still has some properties to be improved, such as poor adhesive strenth with acrylic resin, discoloration, degradation and growth of candida. We attempted to apply chemically-stable polyfluoroethylene-copolymers for this purpose. The terpolymers consisting of difluoroethylene, tetrafluoroethylene and chlorotrifluoroethylene (or hexafluoropropylene) were prepared and tested for tensile strenth, adhesive strength to acrylic resin, water sorption, contact angle, shore hardness, and toxicity by hemolysis test, cell culture test and extraction test. One of the selected terpolymer showed less water sorption (0.1 mg/cm2), stronger tensile strength (160 kg/cm2) and adhesive strength (110 kg/cm2) than the silicone rubber lining material. The toxicity test results suggested that the terpolymer will pose a low potential acute toxicity in vivo. All the test results seem to suggest that the polyfluoroethylene elastomer described here is a promising soft liner material for denture base.
