Data are presented on a triplex type with two parallel homologous strands for which triplex formation is almost as strong as duplex formation at least for some sequences and even at pH 7 and 0.2 M NaCl. The evidence mainly rests upon comparing thermodynamic properties of similar systems. A paperclip oligonucleotide d(A12C4T12C4A12) with two linkers C4 obviously can form a triplex with parallel back-folded adenine strand regions, because the single melting transition of this complex splits in two transitions by introducing mismatches only in the third strand region. Respectively, a hairpin duplex d(A12C4T12) and a single strand d(A12) form a triplex as a 1:1 complex in which the second adenine strand is parallel oriented to the homologous one in the Watson-Crick paired duplex. In this system the melting temperature T(m) of the triplex is practically the same as that of the duplex d(A12)-d(T12), at least within a complex concentration range of 0.2-4.0 microM. The melting behaviour of complexes between triplex stabilizing ligand BePI and the system hairpin duplex plus single strand supports the triplex model. Non-denaturing gel electrophoresis suggests the existence of a triplex for a system in which five of the twelve A-T*A base triads are substituted by C-G*C base triads. The recognition between any substituted Watson-Crick base pair (X-Y) in the hairpin duplex d(A4XA7C4T7YT4) and the correspondingly replaced base (Z) in the third strand d(A4ZA7) is mutually selective. All triplexes with matching base substitutions (Z = X) have nearly the same stability (T(m) values from 29 to 33.5 degrees C), whereas triplexes with non-matching substitutions (Z not equal X) show a clearly reduced stability (T(m) values from 15 to 22 degrees C) at 2microM equimolar oligonucleotide concentration. Most nucleic acid triple helices hitherto known are limited to homopurine-homopyrimidine sequences in the target duplex. A stable triplex formation is demonstrated for inhomogeneous sequences tolerating at least 50% pyrimidine content in the homologous strands. On the basis of the surprisingly similar thermodynamic parameters for duplex and triplex, and of the fact that this triplex type seems to be more stable than many other natural DNA triplexes known, and on the basis of semiempirical and molecule mechanical calculations, we postulate bridging interactions of the third strand with the two other strands in the triplex according to the recombination motif. This triplex, denoted by us 'recombination-like form', tolerates heterogeneous base sequences.
A variety of histochemical findings have contributed to a more differentiated architectonical description of the bed nucleus of the stria terminalis (BNST) in the mammalian brain. However, in the human brain investigations of the chemoarchitecture of this nucleus have been rare. Therefore we chose this region in six human autopsy brains in order to map the distribution patterns of 13 immunohistochemical markers for neurotensin (NT), neuropeptide Y (NPY), somatostatin (SOM), enkephalins (ENK), vasoactive intestinal polypeptide (VIP), substance P (SP), neurophysins (NPH), glial fibrillary acid protein, 3-fucosyl-N-acetyl-lactosamine epitope, myelin basic protein (MBP), calbindin (CAB), synaptophysin (SYN) and chromogranin-A (CHR-A). Three chemoarchitectonically distinct areas could be defined. The lateral subdivision of the BNST contained high amounts of NPY and SP-fibre immunoreactivity and was further characterized by the occurrence of neurons labelled for NPY. The central subdivision of the BNST appeared as a histochemically clearly circumscribed compartment with massive fibre immunoreactivity for SOM, ENK, VIP, SYN, CHR-A, CAB as well as SOM, ENK, NT and CAB positive cells but lacked cytosolic or fibre-like immunolabel for NPY and SP. This structure was also ensheathed by myelinated fibres identified by means of MBP immunohistochemistry. The medial subdivision of the BNST showed moderate to high SP and NPY fibre immunoreactivity but lacked immunolabelled neurons and was only scarcely supplied with varicose or punctiform ENK immunoproduct. In the most posterior levels of our sections a cell group labelled for NPH was located lateral to the fornix columns. The lateral subdivision of the BNST (with NPY, SYN) and mainly the central BNST (with SOM, ENK, VIP, SYN and CHR-A) contributed to ventrolateral extensions of dense patchy fibre immunoreactivity throughout the basal forebrain region.
The areal development of photosynthetic efficiency and growth patterns in expanding leaves of two different dicotyledonous species - Coccoloba uvifera and Sanchezia nobilis - was investigated by imaging both processes repeatedly over 32 days. Measurements were performed using combined imaging systems for chlorophyll fluorescence and growth, with the same spatial resolution. Significant differences in potential quantum yield of photosynthesis (F (v)/F (m)), a parameter indicating the functional status of photosystem II, were found between midvein and interveinal tissue. Although base-tip gradients and spatial patchiness were observed in the distribution of relative growth rate, neither midvein nor interveinal tissue showed such patterns in F (v)/F (m). In young leaves, F (v)/F (m) of the midvein was higher than F (v)/F (m) of interveinal tissue. This difference declined gradually with time, and upon cessation of growth, F (v)/F (m) of interveinal regions exceeded those of midvein tissue. Images of chlorophyll fluorescence quenching showed that DeltaF/F (m)' in the different tissues correlated with F (v)/F (m), indicating that, in these uniformly illuminated leaves, transitions in photosynthetic electron transport activity follow those of predawn quantum efficiency. We explore the implications of these observations during leaf development, discuss effects of sucrose delivery from veins to interveinal areas on relative rates of photosynthetic development in these tissues, and propose that the initially higher photosynthetic activity in the midvein compared to the intervein tissues may supply carbohydrates and energy for leaf growth processes.
A prospective follow-up-study of 97 patients operated because of a carpal tunnel syndrome was carried out to investigate postoperative use of physiotherapy. Postoperative 32 patients were treated at least 12 times and 65 patients were not treated physiotherapeuticly. Physiotherapists postoperatively treated by activating joints of fingers and hand. We examined the patients one day preoperative and on an average of 9 months postoperative. Complaints of the patients, local findings, vigorimetrically measured strength and electromyography were registered. The vigorimetrically measured strength of the operated hand improved significantly less in patients treated physiotherapeutically compared to not treated patients. All other complaints and findings improved equally. We interpreted this difference as a result of spare caused by awareness of a sick hand.
After more than 8 years of "in-vivo"-experience with ceramic bearing surfaces in hip endoprostheses clinical and tribological aspects of damage-analysis in 29 cases of retrieved Al2O3 prostheses are performed. The balls and sockets are investigated in view of wear-quantification by relative metric determination and wear-quantification by SCAN-microscopy. In 2/3 of all cases the microstructure average grain standard was superelevated. As hypothesis of the destruction mechanism an avalanche-like acceleration of grain-excavations is discussed.
A case of retrograde perineural tumor that spread from malignant melanoma of the face is reported. Contrast-enhanced CT and plain and contrast-enhanced MR studies, including fat suppression technique, showed tumor extension along the maxillary division of the trigeminal nerve. Histopathological examination of the surgical specimen revealed neurotropic malignant melanoma and confirmed the extent of tumor spread in the perineurium.
One of the factors determining the wear of UHMWPE used as acetabular cup material is the lubricating properties of the head materials. In order to determine the lubricating properties, the wettability of Al2O3, ZrO2, CoCrMo and N-coated TiAl6V4 was established. Wettability is determined by the surface energy of the head material (solid-state material), which was measured via the contact angle of drops of fluid. As head material, appropriate rings from the Ring-on-Disc Test as per International Standard 6474 were used. The poorest wettability was associated with CoCrMo. The wettability of Al2O3 and ZrO2 was comparable. Worthy of note is the tendentially good wettability of N-coated TiAl6V4.
BACKGROUND AND OBJECTIVE: To evaluate the potential of optical coherence tomography (OCT) for imaging and quantifying structural changes in the retinal architecture following venous occlusive disease. PATIENTS AND METHODS: We studied 20 patients with retinal venous occlusive disease: 5 patients suffered from branch retinal vein occlusion (BRVO), and 15 from central retinal vein occlusion (CRVO). Patients with CRVO were studied within 72 hours after the first onset of symptoms. Patients were examined by OCT after a complete ophthalmologic examination including fluorescein angiography. A standard set of linear scans through the center of fixation and individual scans over areas of special interest were performed. Scans were qualitatively evaluated and quantitative measurements were performed on single A-scans at 5 locations of linear scans. Measurements were taken in the center of fixation, at the foveal rim, and at the edge of the scan. We measured total retinal thickness, thickness of cystoid spaces, and thickness of the highly reflective outer band. RESULTS: OCT produced detailed images of retinal thickening, intra- and subretinal hemorrhage, intra- and subretinal fluid accumulation, and formation of intraretinal cystoid spaces. Quantitative evaluation revealed a mean central retinal thickness of 274 +/- 181 microm (72-760 microm) in patients with central retinal vein occlusion. Visual acuity did not correlate with central or extrafoveal retinal thickness. CONCLUSION: Optical coherence tomography is a useful imaging technique for studying changes of retinal architecture following venous occlusive disease. Even minor changes like localized subretinal fluid accumulation or beginning epiretinal membrane formation can be visualized in detail. In contrast to other diseases like diabetic maculopathy, quantitative assessment of macular thickness does not correlate to visual acuity. Nevertheless, because of detailed imaging, OCT might become a useful tool in the follow-up and treatment of individual patients.

Microfilariae of Wuchereria bancrofti were observed in cytologic material in 35 cases. The material included cervicovaginal smears (17 cases), effusions (14), urine (2), bronchial washings (1) and ovarian cyst fluid (1). The initial diagnosis was made from the cytologic smear in all cases; none had clinical filariasis. Symptomatic vaginal bleeding in 9 of the 17 cases with microfilaria-positive cervicovaginal smears was reflected in the large numbers of red blood cells found in the smear. Blood eosinophilia was present in 11 of 19 cases investigated. Eosinophils were seen in the smears in 20 cases. In the majority of the cases of effusions with microfilariae the effusions were malignant. Significant adherence of inflammatory cells and macrophages to microfilariae was present in 7 of the 35 cases. The significance of these findings is discussed.
