[Follow-up checks of children born from induced pregnancy (author's transl)].

The following results were obtained by the authors from follow-up checks of 102 children, aged between 2.5 and eight years, who had been delivered from induced pregnancies. - Developmental abnormalities were recorded from seven cases, in the context of paediatric examinations. Minimal neurological deviations were exhibited by 14 children. Moderate retardation was established in psychological tests from something between 25 and 30 per cent of all cases. Positive responses to electroencephalography, however, without any clinical symptoms were observed in 20 per cent of the probands. Neurological symptoms and moderate retardation were recordable primarily from children who had originated from pathological pregnancies. The above findings are likely to support the recommendation to continue treatment of functional sterility by means of induced ovulation. Yet, treatment of that kind should be restricted to women unlikely to be afflicted with pathological pregnancy, which however, cannot be safely forecast in all cases.

[Oral treatment of severe forms of acne with 13-cis-retinoic acid. Clinical results (author's transl)].

A new, very effective method of treating severe forms of acne (papuplopustular, conglobate, fulminating and tetrad) is the oral administration of 13-cis-retinoic acid. The results described by Peck et al. (1979) in 14 patients are demonstrated in detail in 18 patients. In the initial therapy 1 to 2 mg/kg body weight were administered daily for 12 weeks without any other local or systemic treatment. In the second phase of the study half the patients continued treatment with 0.5 mg/kg body weight 13-cis-retinoic acid, the other half remaining without any further therapy. All inflammatory lesions healed after 12-16 weeks, sebum production was largely reduced. 13-cis-retinoic acid represents a remarkable turn in the treatment of most severe acne.

[13-cis-Retinoic acid. Pharmacologic and toxicologic findings in treatment of severe forms of acne (author's transl)].

This study shows that severe acne is characterized by numerous inflammatory parameters. The group of acne tetrad patients differed from those with conglobate acne in many laboratory measurements. The treatment with 13-cis-retinoic acid was well tolerated by all patients. Sebum suppression and an exfoliative cheilitis occurred in all patients, but no headache and no loss hair. Numerous previously pathological blood chemical values were normalized during the treatment. Transaminases, alkaline phosphatase, triglycerides and total cholesterol did not rise noticeably, trichological and spermatological parameters were not affected by 13-cis-retinoic acid. In an in vivo epicutaneous potassium iodide inflammation model, it was possible to demonstrate for the first time the anti-inflammatory action of 13-cis-retinoic acid.

Changes in acetylcholine in caudate nucleus tissue isolated in situ detected by gas chromatography mass spectrometry selected ion monitoring.

Acetylcholine neurons in the striatum are believed to be primarily intrinsic. In cats a cylinder of caudate tissue was isolated unilaterally from all afferent and efferent connections while preserving the blood supply through the base of the cylinder. Cats were decapitated two and four weeks after operation. Dorsal, intermediate and ventral portions of the viable isolated cylinder, and corresponding tissue from the contralateral hemisphere serving as control, were removed, weighed and frozen. Deuterated internal standards at concentrations approximating those of acetylcholine and choline in the tissue were added and acetylcholine and choline extracted, reacted with propionyl chloride and demethylated with sodium benzenethiolate. The derivatives were separated by gas chromatography, and the acetylcholine and choline concentrations in the tissue evaluated using selected ion monitoring of m/z 58, 60 and 64. The observed decrease in the acetylcholine concentration in the dorsal portion could be due either to the interruption of the cholinergic thalamic input or to the more severe ischemia in this portion. The increase in the acetylcholine concentration in the ventral portion, where blood circulation is normal, may reflect accumulation of acetylcholine in inactive intrinsic neurons after severance of their input, or indicate sprouting of intrinsic acetylcholine neurons.

Inhibitory effects of 13-cis-retinoic acid on human sebaceous glands.

The effects of 13-cis-retinoic acid, which is clinically very effective in the treatment of severe acne, on sebaceous glands in humans was studied by means of histology, planimetry, and autoradiography. Histologically and planimetrically, one finds a marked decrease of size of sebaceous glands of up to 90% of the pretreatment values after 12 weeks of treatment. Additionally, the ratio of the so-called "differentiating cell pool (DCP)" vs. the so-called "undifferentiating cell pool (UCP)" changed from 2:1 to 1:7, probably indicating a disturbance of differentiation (lipid-production) of sebocytes. The labeling index of sebocytes also regressed significantly under therapy with 13-cis-retinoic acid.

Aspartate, glutamate and GABA levels in pallidum, substantia nigra, center median and dorsal raphe nuclei after cylindric lesion of caudate nucleus in cat.

Aspartate, glutamate and GABA levels were determined in afferent and efferent projection nuclei of the striatum after unilateral cylindric lesion in the head of the caudate nucleus in cats. Two and four weeks after operation, GABA content was significantly reduced in substantia nigra and pallidum ipsilateral to the lesion. Glutamate (GLU) level was decreased in substantia nigra and pallidum only 4 weeks after lesion, whereas aspartate content in substantia nigra decreased significantly already after 2 weeks. No changes in the contents of these amino acids were detected in the dorsal raphe nucleus, which receives a projection from the caudate, as well as in the center median nucleus, which projects to the striatum. These experiments using longer survival times substantiate the role of GABA in caudato-pallidal and caudatonigral projections. The possibility is discussed that aspartate (ASP) could function as transmitter of cortico-nigral fibers.

[The influence of 2,4-dichlorphenoxyacetate (2,4-D) on the membrane potential, membrane resistance and electromyogram of the frog skeletal muscle before and after stimulation (author's transl)].

The influence of 2,5-Dichlorphenoxyacetate (2,4-D) on the membrane potential, membrane resistance and electromyogram of the isolated frog skeletal muscle was studied. The membrane potential and membrane resistance were measured in the rest and after an impulse series of 10 impulses which was applicated with 1 impulse/s. The membrane resistance was not significantly variable. The membrane potential decreased about 30 mV when the muscle was 4 hours in 2,4-D (Fig. 1). At rest the membrane potential as well as the muscle in a solution with and without 2,4-D had constant values when the muscle was only a short time in 2,4-D. But when the muscle was stimulated several times with 1 impulse/s the membrane potential decreased about 20 mV. After a time of 30 s the normal value of the rest-potential was reached again. The effects induced by 2,4-D were reversible in Ringer-solution (Fig. 2). These findings are discussed and beside changes in conductances the role of the Ca-ions is also mentioned as a possible cause. An anomalous temporary binding of ions on the membranes of the muscle is assumed.

Myotonic effects of 2,4-Dichlorphenoxyacetate (2,4-D) on the isolated frog skeletal muscle (author's transl).

2,4-Dichlorphenoxyacetate (2,4-D) produces myotonic reactions not only in mammalian skeletal muscles but also in muscles of cold blooded animals. There was induced a myotonic reaction by 2,4-D on isolated frog skeletal muscle and the changes of mechanogramm and electromyogram were studied. The relaxation of contraction was prolonged about 40% when the muscle was 5 min in a solution with 3 mMol 2,4-D and the muscle was stimulated with an impulse of 1 Hz (Fig. 1 and 2). There was only a slight increase in the amplitude of the mechanogram. A higher stimulation frequency clearly showed the warm-up phenomenon. The amplitude of electromyogram increased during tetanic stimulation (Fig. 3). Finally we found also myotonic discharges when the muscle came into contact with 2,4-D (Fig. 4). All the actions induced by 2.4-D were reversible. These changes of mechanogram and electromyogram induced by 2,4-D were interpreted as a variation of the Ca++-kinetic in sarcoplasmatic reticulum and as an alteration of the sarcolemma. We interpreted the difference to the findings on mammalian skeletal muscle as a variation in metabolism between warm- and cold-blooded animals.

[Electromyography and electroneurography in peripheral nerve lesions. Part I].

First, the possible forms of the lesion of peripheral nerves are defined in the form of a survey: pressure damage (neurapraxia), axon interruption with maintained envelope structures (axonotmesis) and complete separation of the nerve (neurotmesis). From these, the various prognostic characteristics of a nerve lesion can be derived. An neurological examination is not sufficient for a thorough study of these phenomena. A particularly valuable aid is offered by the neuroelectrodiagnostic methods of electromyography and electroneurography. The technical fundamentals of these are briefly dealt with by the authors and indications are derived from them for the use of the two methods. In case of the open nerve lesion, the methods are used for a demonstration of intact nerve fibres, and, in the later phase, for the proof of returning spontaneous activity as well as the degree of the progress of a reinnervation. As far as chronic pressure damage is concerned, for example in bottle-necks, information regarding the seat and the degree of the nerve damage can be derived. And finally, EMG and ENG permit to make statements about the segmental height of radicular damage; this is necessary in case of plexus damage, but also for the demarcation of distal lesions. In the second part, the combination with the clinical practice and therapy will be dealt with in greater detail.

[Myopathia distalis tarda hereditaria. Results of a genealogical, clinical, clinical-chemical and electromyographic family study].

A family is described which displays a typical picture of autosomal-dominant inherited myopathia distalis tarda hereditaria, which is very rare outside Sweden. Three subjects still living who are carriers show, as probable heterozygotes, a very homotypical clinical picture but with distinct participation of the muscles of the proximal extremities and the limb girdles, in some cases also of the mimic muscles, a generalised myogenic disturbance in the EMG and clearly increased CPK activities in the serum. An early diagnosis might be possible from the age of 30, in which the EMG is of prime importance, ahead of clinical microsymptoms and the increase in the CPK activity.

Catecholamines in adult iron deficiency patients.

Patients with iron deficiency whether uncomplicated or associated with other types of anemias, had plasma catecholamine levels which were significantly increased above normal controls. Patients with a variety of other anemias had no significant increase in catecholamine levels. Plasma catecholamine levels in uncomplicated iron deficient patients approached normal values as early as 3 h following oral FeSO4.
