[Glucose-6-phosphate dehydrogenase and succinate dehydrogenase in metastatic cells of Lewis lung carcinoma].

By the experimental model of the Lewis lung carcinoma we studied the activity of glucose-6-phosphate-dehydrogenesis (G6PDH) and succinic-dehydrogenesis (SDH) in the carcinoma itself, the lung metastases, the peripheral blood. A set of carcinoma, lung sections, peripheral blood smears were studied by histochemical techniques day by day since the 3rd day to the 15th one after the carcinoma grafting. The reported results allow us to conclude that G6PDH looks like a determinant of metastatic cell survival and growth and can be regarded as a marker of metastatic cells in the present model.

In vivo and in vitro processing of seed reserve protein in the endoplasmic reticulum: evidence for two glycosylation steps.

Cotyledons of the common bean (Phaseolus vulgaris L.) synthesize large amounts of the reserve protein phaseolin. The polypeptides are synthesized on membrane-bound polysomes, pass through the endoplasmic reticulum (ER) and accumulate in protein bodies. For a study of the biosynthesis and processing of phaseolin, developing cotyledons were labeled with radioactive amino acids, glucosamine and mannose, and isolated fractions (polysomal RNA, polysomes, and rough ER) were used for in vitro protein synthesis. Newly synthesized phaseolin present in the ER of developing cotyledons can be fractioned into four glycopolypeptides by SDS PAGE. In vitro synthesis with polysomal RNA results in the formation of two polypeptides by polysome run-off shows that glycosylation is a co-translational event. The two unglycosylated polypeptides formed by polysome run-off are slightly smaller than the two polypeptides formed by in vitro translation of isolated RNA, indicating that a signal peptide may be present on these polypeptides. Run-off synthesis with rough ER produces a pattern of four polypeptides similar to the one obtained by in vivo labeling. The two abundant glycopolypeptides formed by polysome run-off. This result indicates the existence of a second glycosylation event for the abundant polypeptides. Inhibition of glycosylation by Triton X-100 during chain-completion with rough ER was used to show that these two glycosylation steps normally occur sequentially. Both glycosylation steps are inhibited by tunicamycin. Analysis of carhohydrate to protein ratios of the different polypeptides and of trypsin digests of polypeptides labeled with [(3)H]glucosamine confirmed the conclusion that some glycosylated polypeptides contain two oligosaccharide chains, while others contain only one. An analysis of tryptic peptide maps shows that each of the unglycosylated polypeptides is the precursor for one glycosylated polypeptide with one oligosaccharide chain and one with two oligosaccharide chains.

Inhibition of superovulation and alteration of ova transportation in hamsters by prostaglandin and DL-204-IT, an anti-fertility agent.

The compound 2(3-ethoxyphenyl)-5, 6-dihydro-s-triazole- [5, 1-2] isoquinoline (DL-204-IT or L-11204) inhibited PMS-induced superovulation and reduced the number of recoverable ova in the oviducts of hamsters. PGE2 non-significantly reduced the number of ova shed but significantly decreased the number of oviductal ova. DL-204-IT was more potent than PGE2 for these effects. Administration of both compounds resulted in a complete block of ovulation. Inhibition of prostaglandin metabolism by DL-204-IT may be the cause for these effects, but no direct evidence is presently available.

Impairment of right brain-damaged patients on a linguistic cognitive task.

Forty right hemisphere-damaged patients and 35 normal controls were given five language tests, the Token test, a naming to description test, the Reporter's test, a word fluency test and a sentence anagram test. The sentence anagram test was the only measure on which right hemisphere patients performed significantly more poorly than control patients. It is surmised that this impairment may be indicative of a direct right hemisphere participation in linguistic performances.

An epidemic of type B viral hepatitis: its course and outcome.

Between June and August, 1977 there was a small epidemic of type B viral hepatitis in a market town in South-East England during which 4 people developed acute hepatitis. Their close friends and contacts were all negative for hepatitis B surface antigen (HBsAg) at this time. One year later, in July 1978, 3 of the original 4 patients were available for follow-up. All were HBsAg negative but hepatitis B surface antibody (anti-HBs) and hepatitis B core antibody (anti-HBc) positive. Two had disturbed liver function tests and chronic persistent hepatitis on liver biopsy. Also at this time 2 of the original contacts were found to show evidence, from the serum markers, of previous infection by the type B virus. Both had abnormal liver function tests and showed chronic persistent or chronic active hepatitis on liver biopsy. A further subject was identified who had had an overt hepatitis 1 year previously and who also showed evidence of continuing liver disease. The origin of the epidemic and its spread through the group were identified. The varying modes of transmission, and the variation in clinical picture and sequelae are discussed. The importance of anti-HBc as a marker of hepatitis B virus infection is emphasised.

[Still's disease. Review of the literature and considerations on a case].

Following an extensive review of all the most recent literature on primary chronic rheumatism in infants or the young, a personal case of Still's disease is reported. Special attention is paid to the good results obtained associating an antitubercular therapy, even though no clinical or laboratory findings suggest any specific tubercular condition: no definitive conclusions are reached but an aspect of the problem which is undoubtedly worth further study is stressed.

Effects of prostaglandin E2 and DL204 IT, an inhibitor of prostaglandin degradation, on ovulation and ovum transport in the hamster.

The effects of 2(3-ethoxyphenyl)-5,6-dihydro-s-triazole-[5,1-a]isoquinoline (L-11204 or DL 204 IT and PGE2 on ovulation and ova transport were studied. DI 204 IT was administered in doses of 0.2-25 mg/kg s.c. on the day of estrus. A smal reduction in ovulating follicles was observed 96 hours later, but only at the 5 mg/kg dose level. At all dose levels, however, DL 204 IT caused a dose-related reduction in the number of ova in the oviducts. PGE2 at a total dose of 2 mg/animal s.c., administered in 4 divided doses over the second and third day of the cycle did not affect ovulation or ova transport. PGE2 plus DL 204 IT (5 mg/kg), however, completely blocked ovulation in all but one animal. That animal had one ovulated follicle and a single ova was recovered from its oviduct.
