Defective natural killer activity within human ovarian tumors: low numbers of morphologically defined effectors present in situ.

Tumor-associated lymphocytes (TAL) were isolated from 17 ascites and 7 solid ovarian carcinomas. TAL had defective natural killer (NK) activity against K562. Large granular lymphocytes, the morphologically identified effectors of NK activity, were poorly represented in TAL from ovarian carcinomas as compared to peripheral blood lymphocytes from the same patients or from normal donors. Similar results were obtained when effectors of NK activity were identified with an anti-NK (HNK-1) monoclonal antibody. When four TAL preparations were separated on discontinuous Percoll gradients, they were able to be enriched for NK activity and large granular lymphocyte morphology in the lower density fractions as observed with blood. These observations suggested that a low concentration of the relevant effector cells was the major factor determining the defective NK cytotoxicity of lymphoid cells associated with these human neoplasms.

Discrimination of hepatobiliary diseases by the evaluation of bromsulfophthalein blood kinetics.

Blood BSP kinetics was studied by compartmental analysis in 10 normal subjects, 11 cirrhotic patients, and nine patients suffering from obstructive jaundice. Plasma levels of both free and conjugated BSP were determined at different times after intravenous injection of a standard dose (5 mg/kg body weight) of the dye. A six-compartment model was found suitable for the correct evaluation of hepatobiliary function, but simpler models cannot be used in this respect. Some of the parameters of the model can be adequately evaluated in the studied measurement situation and are used to carry out a diagnostic discrimination; they are lambda21 (transfer constant from blood to liver cells), alpha1 (expression of both conjugating and excretory activity), and beta (related to the blood level of conjugated BSP). It is shown that the values of these parameters allow a fairly good separation among the three groups of subjects under study.

Absorption and excretion of 3H-raubasine in human subjects and dogs.

The pharmacokinetic behavior of (3,5,6-3H)-raubasine (RAU) was investigated in human subjects after oral administration and in dogs after both intravenous and oral administration. By the oral route RAU peak plasma levels appeared in human subjects after 1 h and in dogs after 2 h. Three-day cumulative urinary excretion was 22% by i.v. route and 13% by oral route in dogs, being 29% in human subjects after oral administration. Three-day cumulative faecal excretion in dogs was 51% by i.v. route and 57% by oral route whilst it was 24% in humans. From a comparison between the urinary excretion values observed after i.v. and oral administration, a RAU intestinal absorption of 59% may be obtained.
