[Revelation and identification of laminin in the structure of plasma membrane of ascitic Zajdela hepatoma cells].

Cell dysdifferentiation during neoplastic transformation is a crucial problem of cell biology and oncology. Antigenic diversion of cancer cells is a typical characteristic of dysdifferentiation. It involves the appearance of antigens which are unusual for normal tissue of this type. Components organospecific for membrane proteins of normal kidney were previously found among plasma membrane proteins of hepatocellular rat tumors, rat hepatocytes after carcinogen treatment, and regenerating liver, respectively. In the present work we showed that a protein with mol. weight about 200 kDa reacting with laminin-1 immunoserum is the basic component of plasma membranes of the rat Zajdela hepatoma cells, which is responsive for organospecific anti-kidney immunoserum in Western blot. A mass-spectrometer analysis of trypsin proteolysis fragments was carried out in SDS-PAGE slices containing the investigated component. The analysis showed the presence of beta1, beta2 and alpha4 laminin chains peptides. The component with mol. weight about 180 kDa, found in the Western blot with laminin-1 immunoserum, was also subjected to the mass spectrometer analysis. As a result, a gamma1 laminin chain was found. An increased amount of laminin was revealed in the ascitic liquid and sera of rat with developed Zajdela hepatoma, in comparison with sera of normal rats. In addition, we found the appearance of laminin on the hepatocyte surface on the 4th day after hepatocarcinogen injection (N-diethylnitrosamine, DENA). Thus, for the first time tumor associated antigens were revealed and identified in the structure of plasma membranes of Zajdela hepatoma cells, being specific to rat kidneys. Our results allow to conclude that in the process of carcinogenesis in rat liver laminin synthesis occurs, which is also characteristic of the rat hepatoma Zajdela cells.

[Antibiotic prophylaxis, antibiotic therapy and microbiological situation in surgical unit].

The clinical and economic efficacies of antibiotic prophylaxis in the surgical unit of the Hospital were confirmed by the results of the analysis of 1313 case records of the patients operated during a year for acute appendicitis and acute cholecystitis. At the same time it was shown advisable to use antibiotic therapy in the patients with various pathological processes. The dynamics of the microbial dissemination in the surgical unit and some other units of the Hospital, as well as the dynamics of antibiotic resistance of the microflora, its interrelation with the volume of the antibacterials used and their rotation are described.

Laser-induced fluorescence line narrowing of native porphyrins in animal tissue.

Using monochromatic laser excitation, narrowing of the porphyrins 0-0 fluorescence line in rat muscle tissue to a width of 50-70 cm-1 at 4.2 K is observed. Fluorescence spectra at different excitation wavelengths and excitation spectra are measured. Fluorescence line peak follows the shift of the excitation frequency. Several of the most prominent vibrational frequencies of porphyrins in the excited electronic state are determined.

[Genetic approaches to Itsenko-Cushing disease].

107 patients with Itsenko-Cushing disease were examined for heredity: family history was analyzed in 75 cases, dermatoglyphics was assessed in 44 cases, I- and II-class HLA antigens were studied in 68 cases. The patients were found to have hereditary loading both by Itsenko-Cushing and other diseases (hypertension, atherosclerosis, autoimmune disorders). Clinico-genealogical evaluation made it possible to identify forms of the disease which are inherited autosome-recessively and autosome-dominantly. However, in the majority of patients the disease onset had multifactorial nature, as there were HLA-antigen associations by DR4, DR5, DR7, DRw53, DQw3. Pilot experience with genetic study of the disease showed its genetic determination in some forms, its association with hypertension and atherosclerosis, approaches to prevention, prognosis, classification. Practical recommendations on detailed family history collection in patients with Itsenko-Cushing disease have been developed.

[Method of studying the opinion of patients on the operation of an ambulatory rehabilitation center].

A questionnaire has been developed to study the extent of patient's satisfaction with rehabilitative treatment under conditions of a specialized rehabilitation centre. 100 neurological and traumatological patients were surveyed by means of questionnaires. A technique for the data obtained processing has been suggested which permitted one to identify the main factors lessening the extent of satisfaction with the outcomes of treatment. A comparative analysis was made of patients' and physicians' opinion on the quality of treatment provided. The comparison of questionnaire data provided by patients and objective indicators of health institution performance is aimed at increasing the level of health care delivery.

[Methodological approaches to the evaluation of the effects of environmental pollution on morbidity of the population].

A method is proposed which is based on the comparison of calculation coefficients estimated by eliminating the actual rates of morbidity resulting in temporary disability among workers and employees of some branches of national economy residing in different territories. Calculation algorithms are provided, the use of which is illustrated by relative and specific examples. It is demonstrated that the method based on the evaluation of present information systems is fully acceptable for assessing the influence of environmental pollution on the morbidity of able-bodied population.

[Thermographic examination of patients with neurologic manifestations of lumbar osteochondrosis].

The article deals with thermography data collected for 94 patients with different clinical signs of lumbar osteochondrosis (reflex, radicular, vascular, radiculo-spinal). The features of thermograms were described as depending on the leading clinical syndrome of the disease. A lack of direct correlation between the severity of the pain syndrome and thermoasymmetry was registered. The data suggest wider use of the diagnostic capacities of IR-imaging techniques in patients with vertebrogenic pathology.

[Changes in the resting ECG in males with impaired carbohydrate tolerance of different degrees without clinical symptoms of ischemic heart disease and arterial hypertension].

The resting ECGs were analyzed in males aged 20-60 years with various tolerance to carbohydrates. There were statistically significant differences between the values characterizing the functions of automatism and conductivity on the resting ECG in patients with diabetes mellitus without any clinical symptoms of coronary heart disease (CHD), arterial hypertension, cardiac failure and other chronic diseases and in healthy males with the normal tolerance to carbohydrates. in patients with borderline impairments of carbohydrate tolerance (CT), these parameters occupied an intermediate position between the corresponding values in patients with diabetes mellitus and in subjects with the normal tolerance to carbohydrates. During the mass screening of the population the complex of the examinations conducted should include the methods of monitoring carbohydrate metabolism in order to identify individuals with CT impairments who are at risk of developing both CHD and diabetes mellitus.

[Arachidonoyl amino acids and arachidonoyl peptides: synthesis and properties].

N-Arachidonoyl (AA) derivatives of amino acids (glycine, phenylalanine, proline, valine, gamma-amino butyric acid (GABA), dihydroxyphenylalanine, tyrosine, tryptophan, and alanine) and peptides (Semax, MEHFPGP, and PGP) were synthesized in order to study the biological properties of acylamino acids. The mass spectra of all the compounds at atmospheric pressure electrospray ionization display the most intense peaks of protonated molecular ions; the detection limits for these compounds are 10 fmol per sample. AA-Gly showed the highest inhibitory activity toward fatty acid amide hydrolase from rat brain (IC50 6.5 microM) among all the acylamino acids studied. AA-Phe, AA-Tyr, and AA-GABA exhibited a weak but detectable inhibitory effect (IC50 55, 60, and 50 microM, respectively). The acylated amino acids themselves, except for AA-Gly, were stable to the hydrolysis by this enzyme. All the arachidonoylamino acids inhibited cabbage phospholipase D to various degrees; AA-GABA and AA-Phe proved to be the most active (IC50 20 and 27 microM, respectively). Attempts to detect the biosynthesis of AA-Tyr in homogenates of rat liver and nerve tissue showed no formation in vitro of either this acylamino acid or AA-dopamine and AA-Phe, the products of its metabolism. The highest contents of these metabolites were detected in liver homogenate and in the brain homogenate, respectively. Acylamino acids exert no cytotoxic effect toward the glioma C6 cells. It was shown that N-acylation of Semax with arachidonic acid results in enhancement of its hydrolytic stability and increases its affinity for the sites of specific binding in rat cerebellum membranes.
