[The spectrum of analgesic activity of baclofen and tolibut].

Spectrum of the analgetic activity of GABA derivatives baclofen (2.5-10.0 mg/kg) and tolibut (12.5-100.0 mg/kg) was studied in experiments on albino mice and rats. Baclofen and especially tolibut showed a dose-dependent inhibition of perceptive, emotional and autonomic manifestations of nociceptive reactions at the tail's thermal and electrical stimulation and enhancement of the analgetic effect of promedol. The regressive analysis made it possible to show dependence of the analgetic effect on the level of pain sensitivity and emotional reactivity of the animals. Tolibut deepens and prolongs sombrevin anesthesia, baclofen fails to alter it.

[Hormonal aspects of cathepsin D regulation in seminal follicles].

Androgen-dependent regulation of cathepsin D activity was studied in rat seminal follicles. Cathepsin D, isolated from the follicles by means of ammonium sulfate fractionation, chromatography on Hb-Sepharose and Sephadex G-200, was similar to the enzyme from other tissues in molecular mass, Km for hemoglobin, pH optimum of activity, thermolability. Concentration of androgens in tissues affected the properties of purified cathepsin D: the enzyme activity, its sensitivity to thermoinactivation and to chymotryptic hydrolysis were increased as well as the pH optimum was expanded after orchidectomy. Testosterone restored the initial enzyme properties altered after castration.

[Evaluation of the pain-alleviating properties of a combination of analgesic and tranquilizer using multiple stepwise regression].

A method for preclinical evaluation of the optimal ratio (for analgesia) of an analgesic to a tranquilizer has been devised. It is based on the drug threshold dose decrease on the drug combined use. The multiple stepwise regression analysis of the data discovered the correlations between diazepam and promedol, optimal for pain perception decrease and nociceptive emotional responsiveness inhibition.

[Analysis of the role of various factors of the initial status of the body in the analgesic action of promedol].

A comprehensive approach is suggested to the assessment of the analgetic action of analgetics during screening. This approach lies in the study of the drug effects on the emotional, vegetative and motor manifestations of the painful reactions. Based on multiple and regression analysis the role of different factors of the initial status of the body in the development of analgesia is demonstrated. Application of the given approach is examplified by the reference analgetic promedol.

[Effect of hormonal preparations on lysosome enzyme activity in rat tissues].

Experiments on white male rats were made to study and compare the action of hormonal drugs (testosterone propionate, retabolil and chorionic gonadotropin) on lysosomal enzymes of different tissues. There were differences in the changes in the activity of acid phosphatase, ribonuclease, cathepsins and beta-galactosidase after a single administration of testosterone and after a course of drug treatment. Retabolil and chorionic gonadotropin acted on lysosomal enzymes of spermatic vesicles similarly to testosterone given in a single dose. As far as the activity of liver cathepsins and beta-galactosidase is concerned retabolil was found to produce an opposite effect as compared to that of testosterone.

[Relationship between photoinduced changes in the intercellular concentration of potassium ions and transretinal potential generation by the Muller cells of the retina].

In isolated frog retina after treatment with aspartate the photoinduced change of the extracellular potassium ions concentration [K+]0) was compared with slow PIII potential (sPIII). The time course of [K+]0 change in the outer nuclear layer was like that in sPIII one. Both the size of [K+]0 change and the sPIII amplitude were increased almost in proportion to the Log light stimulus duration in the whole range of durations used (from 13 ms to 13 s). Such similarity between these two processes suggests that there is a direct relationship between sPIII and [K+]0 change. On the basis of our data the electrical model of sPIII generation by Muller cells in response to photoinduced [K+]0 change in the photoreceptor layer is proposed.

[The cloning and expression of the phospholipase D gene from Corynebacterium pseudotuberculosis].

In a number of consecutive gene engineering operations a DNA fragment having a size of about 2.8 kb was cloned in Escherichia coli by means of Blue-script II SK+ used as vector. The insert contained pld gene coding the synthesis of phospholipase D, one of the key factors of C. pseudotuberculosis virulence. The stable and active expression of this gene in E.coli was achieved. High phospholipase A activity was accumulated in the periplasmatic space. The molecular weight of the synthesized protein was 31 kD. The product obtained by gene engineering methods was found to possess the biological activity of the natural product: it induced the hemolysis of sheep red blood cells in the presence of equi factor of Rhinococcus equi and inhibited the hemolytic activity of Staphylococcus aureus beta-hemolysin (phospholipase C). The pld gene cloned in these experiments differed from that of another C.pseudotuberculosis strain. Further research is underway with a view of searching for the limits of pls gene.
