Carcinoembryonic antigen (CEA) in plasma is useful for the detection of recurrent colonic or gastric cancer and the monitoring of plasma in patients with recurrent cancer displaying therapeutic effect. We report a sharp decline of CEA in a patient with recurrent gastric cancer by 6 months oral administration of UFT. The patient was an 81-year-old male who had undergone gastrectomy for advanced gastric cancer. Eight months post-operatively, the plasma CEA began to rise logarithmically, and recurrent tumor in the remnant stomach and liver metastasis were detected by fibergastroscope (FGS) and abdominal CT. After administration of UFT at a dose of 300 mg per day, CEA abruptly declined logarithmically and normalized in 6 months. Presently marked reduction of recurrent foci and metastases were found by FGS and abdominal CT. Therefore sequential changes in plasma CEA in this patient can be considered to reflect the effect of therapy for recurrent gastric cancer by UFT.
Lymphocytes isolated from fresh human epithelial ovarian tumors can be expanded in the presence of recombinant interleukin-2, and some CD8 antigen-positive lymphocytes can lyse autologous fresh tumor cells. We treated 7 patients with advanced or recurrent epithelial ovarian cancers, using the adoptive transfer of these tumor-infiltrating lymphocytes (TILs) following a single i.v. injection of cyclophosphamide. One case of disappearance (complete response) and 4 cases of greater than 50% decrease of tumor (partial response) were reported (14.3 and 57.1%, respectively). Regression of tumors in the ovary, liver, lung, and lymph node, both primaries and metastases, lasted for 3-5 months. Furthermore, 10 patients were treated alternately with a cisplatin-containing chemotherapeutic regimen and the adoptive transfer of TILs. Seven cases of complete regression and 2 cases of partial regression of cancer were reported. Four of the 7 patients with a complete response have had no recurrence for greater than 15 months of follow-up. It appears that this experimental technique of adoptive transfer of TILs achieves high response rates even without recombinant interleukin-2 administration and that the prospect of combined therapy using TILs and cisplatin offers hope for increasing the cure rate and long-term survival.
'Per repeat is a rodent genomic DNA fragment which is homologous to the Gly-Thr repetitive amino acid sequence of the Drosophila clock gene period'. This study examined the temporal and spatial expression of the per repeat mRNA in rat brain. Northern blot analysis showed that the level of per repeat mRNA species fluctuates under light-dark cycle conditions in rat brain. Furthermore, the fluctuation of per repeat mRNA was clearly observed in the suprachiasmatic nucleus, but not so in other regions including the hippocampus as shown by in situ hybridization. The above results suggest that the Gly-Thr repetitive sequence also plays an important role in the circadian rhythm of mammals.
We examined the correlation among preoperative serum carcinoembryonic antigen (CEA) levels, staining properties of the tumors by CEA immunohistochemistry and the tumorigenicity of their xenografts in nude mice, in 28 patients with gastric cancer. Eleven (40 per cent) of them were positive for serum CEA (greater than or equal to 2.5 ng/ml) and seven (25 per cent) of the xenografts were tumorigenic in nude mice. All the tumorigenic cases were positive for serum CEA (p less than 0.001) and the mean value of the serum CEA level in the patients with tumorigenic neoplasms was 20.8 ng/ml, being significantly higher than that (1.4 ng/ml) in the patients with non-tumorigenic neoplasms (p less than 0.001). Twenty-five of the 28 carcinomas (89 per cent) were positive for CEA staining in their cancer cells by the ABC method and CEA localization correlated with tumorigenicity (p less than 0.05). These results suggest that the serum CEA level in patients is correlated with the tumorigenicity of their gastric carcinoma xenografts in nude mice and may account for the poor prognosis of patients with high serum CEA.
A monoclonal antibody (MAb-1D10), generated by immunizing Balb/c mice with homogenized bovine retinal tissue, labeled the Müller cells in the bovine retina and the Purkinje cells of the rat cerebellum. In the neonatal rat cerebellum, immunoreactivity for MAb-1D10 appeared first in the ventral regions of vermis (lobuli I-III and IX-X) on the 11th postnatal day (P11), while in the dorsal vermis and cerebellar hemisphere, the immunoreactive Purkinje cells were detected on P13. Western blotting analysis disclosed that MAb-1D10 recognized bovine retinal polypeptides with mol.wt. of 48 kDa. Transplantation of the rat cerebellar primordia on 16th embryonic day demonstrated large and round cells with a linear arrangement in the grafts surviving well in the host rat striatum after 50 days of the grafting. These large cells were immunohistochemically labeled by MAb-1D10. No other cells in both the graft and the host striatum were immunoreactive to the MAb.
Protein kinase inhibitor H-7 and dibutyryl (dB)-cAMP were found to induce neuritic processes in mouse neuroblastoma N18TG2 cells (36). In the present study, morphological differences between the neurites induced by H-7 and those by dB-cAMP were examined using electron microscopy (TEM and SEM) and tubulin immunohistochemistry. It was observed that: 1) The neurites induced by H-7 were relatively thin and frequently had varicosities. On the other hand, the neurites induced by dB-cAMP were thick but they had few varicosities. 2) Centrioles were frequently observed in the cells treated with dB-cAMP but were not encountered in the H-7-treated cells. 3) TEM and tubulin immunohistochemistry revealed that the main shafts of the neurites induced either by H-7 or dB-cAMP were filled with microtubules, but that the varicosities induced by H-7 contained a smaller amount of microtubules. 4) The stability to colchicine was greater in the neurites induced by H-7 than in those by dB-cAMP. From these features of the neurites, it was inferred that neurite outgrowth induced by dB-cAMP is deeply related to the formation of microtubules and that the neurites induced by H-7 were involved in other processes probably including an adhesive property of cell surfaces.
A monoclonal antibody (MAb-1E7), generated against bovine retinal homogenate, labeled the outer and inner segment layers of the vertebrate retina. Immuno-electron microscopic observation clearly demonstrated that antigen(s) bound by MAb-1E7 was localized in the cell membrane of the outer segment and the distal portion of the inner segment. Western blot analysis revealed that MAb-1E7 recognized 40 kD- and 27 kD-polypeptides. Mouse retina with hereditary photoreceptor degeneration (C3H/He and CBA strains) did not involve the MAb-1E7 immunoreactive structures. The present immunocytochemical observation demonstrated that MAb-1E7 was highly specific to the outer segment of the photoreceptor cells and, therefore, can be a useful marker for the cells.
Thirty-two surgical specimens and three cell lines of human gastric cancers were used for subcutaneous transplantation into nude mice, resulting in the establishment of eight (25%) xenografts from the surgical specimens and two (67%) from the cell lines. The localization of epidermal growth factor (EGF) in the surgical specimens and cell lines of the gastric cancers and their xenografts in nude mice was then investigated immunohistochemically. Epidermal growth factor was stained in the cytoplasm of the cancer cells, being detected in 16 (50%) of the 32 surgical specimens and in all of the cell lines. Seven (44%) of the sixteen EGF-positive surgical specimens and one (6%) of the 16 EGF-negative ones were tumorigenic in nude mice. All of the xenografts in nude mice were positive for EGF. The tumorigenicity of human gastric cancer xenografts in nude mice may, therefore, be correlated with the presence of EGF in cancer cells.
Neonatal administration of cytosine arabinoside (Ara-C) induced marked hypoplasia of the mouse cerebellum. In 17-20-day-old mice with neonatal Ara-C injections, the external granular layer was still preserved as an irregular cell laminae in the superficial part of the cerebellum, in contrast to its complete disappearance in normal animals at these stages. Immunohistochemical examination using anti-glial fibrillary acidic protein and monoclonal antibody-1D11 (Ono et al: Dev. Brain Res., 50:154-159, 1989) demonstrated malformation of a palisade-like arrangement of labeled glial fibers in the superficial part of the Ara-C treated mouse cerebellum. Furthermore, the immunoreactive astroglia were observed just beneath the pial membrane and their processes were oriented to the deeper part of the molecular layer.
We examined the localization of epidermal growth factor (EGF) in 185 specimens of primary human gastric cancer using the avidin-biotin peroxidase complex immunohistochemical method on formalin-fixed paraffin-embedded sections. Thirty-four per cent of the gastric cancer specimens were positive for EGF, which was mainly located in the cytoplasm of the cancer cells and occasionally in the stromal cells, but was not detected in non-cancerous gastric epithelium. Moreover, the presence of EGF in gastric cancer was correlated with gastric wall invasion and lymph node metastasis. EGF was found more often in advanced cancers than in early ones (p less than 0.01), and also more often in cancers with lymph node metastasis than in those without (p less than 0.05). The five-year survival of patients with EGF-positive tumors was worse than that of patients with EGF-negative tumors (p less than 0.05). The presence of EGF in human gastric cancer may thus represent higher malignant potential.
We established a human gastric cancer xenograft which, when inoculated into nude mice, showed a positive correlation between tumor growth and the serum level of carcinoembryonic antigen (CEA). Serum CEA levels in the mice rose continuously with increasing tumor weight after inoculation, showing a correlation coefficient of 0.96. A positive correlation was also observed between the tissue CEA level and tumor weight, the former increasing along with the latter. Furthermore, the level of serum CEA closely paralleled that of tissue CEA. The serum CEA level fell after tumor extirpation, with a half-life of approximately 86 h. These results suggest that the elevation of serum CEA is attributable to the gain in tumor weight as well as the increase of CEA production in the tumor tissue. Thus, human gastric cancer xenografts in nude mice are a good model for examining the biological role of CEA.
We investigated immunohistochemical expressions of the epidermal growth factor (EGF) and the epidermal growth factor receptor (EGFR) in 25 cases of pancreatic carcinoma, including 13 cases of metastatic lymph nodes. EGF and EGFR were stained mainly in the cytoplasm and on the surface of some pancreatic carcinoma cells, with positive rates of 18/25 (72%) and 9/25 (36%) respectively. In all cases EGFR was noted only when EGF was also detected. EGF and EGFR were stained frequently in differentiated types. In metastatic lymph nodes, EGF and EGFR were found in 54% and 15% respectively, and they were detected only when they were also noted in the same patient's primary lesions. EGF and EGFR were found more frequently in Stages III and IV than in Stage II. These results emphasize that EGF and EGFR may promote both the proliferation and the differentiation of human pancreatic carcinoma cells, as in other sites.
The effects of estradiol on the growth of six human gastric xenografts in nude mice were studied and diverse effects were found, including one case of stimulation, two of inhibition and three of unchanged condition. Neither the histological features of the original tumor nor the estrogen-binding capacity seemed to be related to the response to estradiol. It is concluded that the growth of human gastric cancer can be modulated by estradiol.
