ESR analysis of the FAD in bovine liver monoamine oxidase.

Two hydrazine spin labels, 1-oxyl-2,2,5,5-tetramethylpyrroline-3-carbonyl ethyl hydrazine and 1-oxyl-2,2,6,6-tetramethylpiperidino-4-hydrazine, were synthesized as probes of the FAD binding site of monoamine oxidase. The reporter nitroxide moiety showed an ESR spectrum classified as partially immobilized which is indicative of FAD near the surface of the enzyme. Attempts to pick up flavin semiquinone or free radical intermediates during substrate oxidation with the spin traps 5,5-dimethyl-1-pyrroline-1-oxidase and phenyl-t-butylnitrone were not successful.

Hormonal manipulation of pulmonary metastases from carcinoma of the prostate.

Chest roentgenograms and medical records were reviewed on 91 consecutive patients having orchiectomy for advanced carcinoma of the prostate during a 4-year period. Our purpose was to investigate the potential of antiandrogen therapy for inducing regression of prostate pulmonary metastases and respiratory difficulty accompanying lymphangitic spread. Patients encountered during the study period who had lung metastases but refused orchiectomy and were treated with oral estrogens were also included. Antiandrogen therapy was found to be effective in inducing regression of prostatic pulmonary metastases and respiratory distress accompanying lymphangitic spread in those patients who had no prior hormonal manipulation.

The immediate hypotensive effect of oral nifedipine.

The immediate hypotensive effect of oral nifedipine was assessed in 15 patients with moderate to severe hypertension. Supine and erect blood pressure and heart rates were measured before and 30 minutes after 10 mg nifedipine. The results showed that there was a significant (p less than 0.001) reduction in both systolic and diastolic pressures. There was a slight increase in heart rate and mild postural hypotension but neither appeared to be significant. It is concluded that nifedipine given by the oral route is an effective agent for the rapid lowering of elevated blood pressure and deserves further evaluation as an agent for the management of hypertensive emergencies.

Solitary nonparasitic hepatic cyst causing obstructive jaundice.

Solitary nonparasitic hepatic cysts are rarely associated with jaundice and abnormalities of liver function have been attributed to hepatocellular disease in the absence of a separate obstructing lesion of the biliary tree. This case represents the first cholangiographic documentation of a solitary nonparasitic hepatic cyst compressing biliary radicles resulting in obstructive jaundice. The implication for treatment is discussed.

Diffuse enlargement of gastric folds as a manifestation of secondary syphilis.

A case of gastric lues with diffuse enlargement of gastric rugae is presented. Confirmation of the etiological role of the spirochete in producing this very unusual roentgen manifestation of syphilitic gastropathy was provided by recovery of the organism from the gastric mucosa and reversal of the clinical and roentgenographic aberrations by penicillin therapy alone. Diffuse hyperrugosity and erosive gastritis occurring in a patient with a reactive serologic test for syphilis should raise the possibility of luetic gastritis.

Colonic ileus. Indication for prompt decompression.

Eleven cases of colonic ileus were associated with a variety of clinical conditions. The dominant roentgenographic finding was severe cecal dilation. Its recognition and prompt management are required to prevent possible disastrous perforation.

The activity of galactose-1-phosphate uridyltransferase and galactokinase in human fetal organs.

The activity of galactose-1-phosphate uridyltransferase (transferase) and galactokinase in several organs from human fetuses 7-28 weeks old was measured by using radioactive substrates and column chromatography for product identification. The specific activity of transferase and galactokinase increased with gestational age and reached, at the 28th week, a maximal level of 30.0 and 7.9 nmol/min/mg protein (units) in liver, 4.7 and 2.5 units in kidney, 6.0 and 4.0 units in lung, 6.7 and 2.9 units in spleen, 5.2 and 2.6 units in cardiac muscle, and 4.0 and 1.4 units in skeletal muscle, respectively. The activity in brain, on the other hand, remained quite constant with 1.2 units in the case of transferase and 0.5 units in the case of galactokinase during this period. The activities of both enzymes in the liver of children were slightly lower than the highest fetal level during the period of pregnancy studied. Galactokinase activity in fetal erythrocytes was approximately 4 times higher and the transferase activity approximately 30% higher than in adults. The Km value of fetal liver transferase for galactose-1-phosphate was found to be 0.330-0.357 mM and that of galactokinase for galactose, 0.265-0.277 mM.
