Conduction of nonconjugative plasmids by F' lac is not necessarily associated with transposition of the gamma delta sequence.

A nonconjugative kanamycin-resistant (Kmr) recombinant plasmid, pNR5311, transferred at a low frequency from an Hfr or F' lac Escherichia coli donor to an F- lac- recipient. Among the transconjugants, two types of Kmr plasmids were found: one was indistinguishable from pNR5311 (type A), and the other was a recombinant between pNR5311 and the gamma delta sequence (type B). When the F' lac strain was used as a donor, 5% of lactose-fermenting (Lac+) and 75% of lactose-nonfermenting (Lac-) transconjugants had type A plasmids. A kinetic study revealed that type A plasmids were transferred more readily in short mating periods than were type B plasmids. Involvement of Tn903, which is present in pNR5311, in transfer of type A plasmids was unlikely since there was no discernible change in the F' lac molecule coexisting with the type A plasmid in the transconjugant cells. The non-gamma delta-associated conduction of pNR5311 by F' lac did not require the recA+ function of the donor. Conduction of pBR322 by F' lac was also carried out, and two types of plasmids with and without gamma delta were found, as with pNR5311. These findings suggest that the transfer of nonconjugative plasmids is conducted by a novel pathway which is not associated with translocation of transposable elements into either plasmid.

Spectral and perceptual evidence for CV coarticulation in devoiced /si/ and /syu/ in Japanese.

We examined the spectral and perceptual characteristics of devoiced /si/ and /syu/ syllables in Japanese. Measurements showed that the deleted vowel can color the spectrum of the [integral of] in these syllables, although the mean influence is small. Perception tests showed that Japanese listeners can use this spectral coloring as a cue to the identity of the underlying vowel, although identification was substantially worse than in syllables where the /i/ or /u/ is maintained. These results suggest that a supposedly lower-level coarticulation between the fricative and the vowel can occur before a higher-level process deletes the vowel, contradicting the order implied by traditional accounts of speech as a translation of discrete phonological units.

Orchid allergy.

A female office worker acquired contact allergy by handling Cymbidium cultivars for several years. The sensitizing properties of this ornamental orchid were demonstrated by experimental sensitization of guinea pigs. Chemical analysis of the ether extracts disclosed the presence of two quinones. The structure of the main quinone was elucidated as 2,6-dimethoxy-1,4-benzoquinone by x-ray crystallographic analysis. In the patient and in the sensitized animals, only this quinone elicited strong reactions, while tests with the second constituent were negative or gave only weak responses. Although 2,6-dimethoxy-1,4-benzoquinone is only a moderately strong sensitizer, its role as a contact allergen is emphasized, as it has been found in more than 50 different plant and wood species.

[CT findings and surgical treatment of atlanto-axial rotatory fixation: a case report].

A case of atlanto-axial rotatory fixation (AARF) was presented in a 19-year-old female who developed sudden onset of neck pain and limitation of neck movement after direct carotid angiography for seizure disorder. Neurological examination was negative except for cock-robin posture and mild hypesthesia and hypalgesia in left C2 distribution. Plain films of the cervical spine disclosed abnormal alignment of C1-C2 and possible rotational dislocation. Bilateral selective vertebral angiography showed marked anterior and posterior displacement of left and right vertebral artery, respectively, at the level of C1. On CT metrizamide myelography, there was clockwise rotation of C1 on C2 with locked facet on the left but no evidence of cord compression was found. With diagnosis of AARF, manual reduction under general anesthesia and with fluoroscopic control was first attempted without success. Therefore, the patient underwent open reduction by using high speed air-drill and posterior fusion of C1 to C3 with acryl and wire. Postoperative course was uneventful and the patient went back to work as a computer operator in three months. The etiology of AARF was described by many authors, but in our case, congenital hypogenesis of transverse and alar ligaments plus minor trauma was most suggested. For neurological manifestations of AARF, occipital neuralgia, headache, neck pain, limitation of neck movement and cock-robin posture were reported, but the cock-robin posture was most characteristic and was an important symptom for the early diagnosis. In neuroradiological findings of AARF, plain CT and CT metrizamide myelography are very useful. Because they clearly demonstrate the degree of rotation and interlocking of atlanto-axial joints, and the presence of cord compression.

Conformational studies of poly(alanine) in dichloroacetic acid by nuclear magnetic resonance.

Proton nuclear magnetic resonance spectra were studied for monodisperse L-alanine oligopeptides (the dimer, trimer, tetramer, and nonamer) containing an n-butylamide group at the C-terminal residue and for the random and block copolymers of D- and L-alanines having sharp molecular weight distributions in dichloroacetic acid (DCA). It was found that the NH signals of poly(L-alanine)s and some copoly(D,L-alanine)s were split into three peaks in DCA, suggesting that these NH peaks reflect the conformation of the polymers. These NH peaks were assigned to the terminal helix (helix-coil junction) and the random-coil and the inner helix, respectively, from the lowest field. On the basis of the above assignment, the microconformations of polycalanine)s in DCA were examined quantitatively. Moreover, it was concluded from the specific nature of the solvent that both the effects of the hydrophobic side chains of the polymers and the acidity of the solvent are most important for the formation of the helical conformation of poly(L-alanine).
