Screening for mutants temperature sensitive in protein synthesis by using methionine analogues.

Two new mutants are described which are temperature sensitive in protein synthesis. The mutants were obtained by a screening procedure using methionine analogues. The method is based on two findings: a) that in E. coli, and in other members of the Enterobacteriaceae, sensitivity to methionine analogues increases with the growth temperature, and b) that cells which are not synthesizing proteins during treatment with methionine analogues have a shorter lag in resuming growth subsequent to removal of the analogue.

[Correlation of growth hormone and aetiology of psoriasis].

Pathological alterations of capillaries, accelerated epidermopoesis, increased rate and extent of metabolism in the psoriatic lesion and first manifestation of psoriasis rarely in the first 20 years of life made us decide to investigate the human growth hormone (STH) in blood serum. In 70 patients suffering from psoriasis we found that the extention of skin lesions was correlated to the STH levels, which were determined every 2 h all over the day. The average STH level in psoriasis was three times as high as average value in the group of 40 controls. Thus, one can conclude that psoriasis is caused by a persistence of STH with individual penetrance.

The importance of ventilation in exercise-induced asthma.

The degree of post treadmill-running decrease in pulmonary function (Exercise-Induced Asthma) in 11 adult asthmatics was compared with the decrease in pulmonary function followed by resting isocapnic hyperventilation. It was checked that ventilation during the hyperventilation was kept identical to the ventilation during treadmill-running by continuous recording of respiratory frequency, minute ventilation, tidal volume and accumulated ventilation. The temperature of the inspired air was identical in the two situations and the relative humidity was 40% during treadmill-running and 15% during hyperventilation. The average accumulated ventilation during treadmill-running and hyperventilation was 411 1/6 min in both events. The decrease in peak expiratory flow after treadmill-running was 25% and after isocapnic hyperventilation 24%. It is concluded that the ventilation is of more importance for the decrease in pulmonary function after exercise, than the work load.

In vitro radiosensitivity of human leukemia cell lines.

The in vitro radiobiologic survival values (n, D0) of four tumor lines derived from human hematopoietic tumors were studied. These cell lines were HL50 (n = 1.3, D0 = 117 rad[1.17 Gy]), promyelocytic leukemia; K562 (n = 1.4, D0 = 165 rad[1.65 Gy]), erythroleukemia; 45 (n = 1.1, D0 = 147 rad[1.47 Gy]), acute lymphocyte leukemia; and 176 (n = 4.0, D0 = 76 rad[0.76 Gy]), acute monomyelogenous leukemia. More cell lines must be examined before the exact relationship between in vitro radiosensitivity and clinical radiocurability is firmly established.

Pancreatic pseudocysts. A follow-up study.

From 1968 to 1978, 37 men and 14 women, with a median age of 43 years, were operated on for a pancreatic pseudocyst. Alcohol abuse was the dominating cause in 65% of the patients. Internal drainage (medium risk patients) was carried out in 76%, external drainage (high risk patients) in 12%, and pancreatic resection (low risk patients) in 12% of the patients. The hospital mortality rate was 14%. The patients who died were significantly older than those discharged from the hospital alive. At the time of follow-up (1--11 years, median: 4 years) after operation, a further 13% had died. Thirty per cent of the alcoholic and none of the nonalcoholic patients had severe pain at follow-up examination. Evaluated by their ability to work and pain, the late results were poorer for the alcoholics who continued drinking, better for alcoholics who had stopped drinking and best for nonalcoholics.

Age-related changes in the incorporation of [35S]sulfate into two proteoglycan populations from human cartilage.

From human hyaline cartilage (processus xyphoid) preincubated in the presence of [35S]sulfate, proteoglycans were extracted by 4M guanidinium chloride and divided into 6 age groups. Fractionation of proteoglycans by gel filtration under dissociative conditions resulted in two proteoglycan fractions (a and b) with different hydrodynamic volumes. The higher molecular weight fraction a contained chondroitin sulfate, the fraction b keratan sulfate as predominant glycosaminoglycan, the chondroitin sulfate/keratan sulfate ratio decreasing with increasing age in either fraction. The relative portion of proteoglycan fraction b and its 35S-labelling increased with increasing age. From the specific 35S radioactivities of the chondroitin sulfate and keratan sulfate preparations, the occurrence of two independent proteoglycan populations is suggested. A precursor-product relationship between proteoglycan fraction a and b could be excluded.

Transient ECG changes during acute attacks of pancreatitis.

Information on the prevalence of ECG abnormalities in patients with acute pancreatitis together with pertinent simultaneous laboratory data have been missing. This prospective study was undertaken in order to clarify these points. 54 patients with 72 acute attacks of pancreatitis were examined. 31 patients (57%) had transient ECG abnormalities. The ECG changes consisted mainly of unspecific T-wave changes (25 cases) and accelerated atrial or nodal rhythms (8 cases). The ECG changes were more common in patients with biliary etiology (80%) than in patients with alcoholic etiology (49%), probably partly due to the higher age of the patients with biliary disease. The laboratory data did not give any clue to the cause of the ECG changes. The authors believe that the ECG changes may be due to underlying ischemic heart disease unmasked by the stress of acute pancreatitis, and/or imbalance of the autonomous nervous system.

The interferon-induced protein kinase PK-i from mouse L cells.

Interferon-treated L cells are characterized by an increased protein kinase activity that can selectively phosphorylate the small subunit of eukaryotic initiation factor 2. This protein kinase, PK-i, has been extensively purified and shown to be a potent inhibitor of mRNA translation. The purified PK-i contains the endogenously phosphorylated 67,000 Mr protein characteristic of interferon-treated cell extracts. PK-i can also phosphorylate arginine-rich histones. Purified PK-i can be activated by preincubation with ATP (but not adenylyl imidodiphosphate) and low concentrations of double-stranded RNA. The activation results in an increase in the first rate of eIF-2 phosphorylation. Activated PK-i becomes resistant to high concentrations of double-stranded RNA and more thermostable. A stimulator of PK-i activity, factor A, was isolated, as well as a specific phosphoprotein phosphatase that dephosphorylates the 67,000 Mr protein and eIF-2. These two factors, which are present in untreated L cells, may regulate the translation inhibitory activity of the interferon-induced and double-stranded RNA-activated protein kinase PK-i.

Linkage studies on the human Pi, Gm, GLO, and HLA genes.

Linkage data of the four gene markers Pi, Gm, GLO, and HLA exclude Gm and Pi gene assignment to the short arm of chromosome 6. The findings, however, do not disprove the following gene order: HLA--GLO--centromere--Gm--Pi.

[A computer-controlled monitoring and therapy system for shock compensation in severe burns (author's transl)].

A computer-controlled control loop for the automatic diagnosis of shock and infusion therapy is reported. External information (off-line) and vital parameters measured with serial devices (on-line) are used by the computer for the automatic control of infusion pumps. By this method the critical shock phase could be prevented in five seriously burned patients. The construction of the system also permits its use in other shock conditions.
