Single chloride channels in endosomal vesicle preparations from rat kidney cortex.

Endocytotic vesicles from rat kidney cortex, isolated by differential centrifugation and enriched on a Percoll gradient, contain both an electrogenic H+ translocation system and a conductive chloride pathway. Using the dehydration/rehydration method, we fused vesicles of enriched endosomal vesicle preparations and thereby made them accessible to the patch-clamp technique. In the fused vesicles, we observed Cl- channels with a single-channel conductance of 73 +/- 2 pS in symmetrical 140 mM KCl solution (n = 25). The current-voltage relationship was linear in the range of -60 to +80 mV, but channel kinetic properties depended on the clamp potential. At positive potentials, two sublevels of conductance were discernible and the mean open time of the channel was 10-15 msec. At negative voltages, only one substate could be resolved and the mean open time decreased to 2-6 msec. Clamp voltages more negative than -50 mV caused reversible channel inactivation. The channel was selective for anions over cations. Ion substitution experiments revealed an anion permeability sequence of Cl- = Br- = I- greater than SO4(2-) approximately F-. Gluconate, methanesulfonate and cyclamate were impermeable. The anion channel blockers 4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid (DIDS, 1.0 mM) and 5-nitro-2-(3-phenylpropylamino)-benzoic acid (NPPB, 0.1 mM) totally inhibited channel activity. Comparisons with data obtained from radiolabeled Cl(-)-flux measurements and studies on the H+ pump activity in endocytotic vesicle suspensions suggest that the channel described here is involved in maintenance of electroneutrality during ATP-driven H+ uptake into the endosomes.

Mutated and hypermutated genes of persistent measles viruses which caused lethal human brain diseases.

Persistent measles viruses (MVs) causing lethal human brain diseases are defective, and the structure of several mutated matrix genes has been elucidated previously. The present study of four persistent MVs revealed a high number of differences from a consensus sequence also in other genes. Amino acid changes accumulated in the carboxyl terminus of the nucleocapsid protein and in the amino terminus of the phosphoprotein, but did not significantly alter these products, which are implicated in viral replication and transcription. The contrary is true for the envelope glycoproteins: In three of four cases, mutations caused partial deletion of the short intracellular domain of the fusion protein, most likely compromising efficient viral budding. Moreover, in the hemagglutinin gene of a strain showing strongly reduced hemadsorption, 20 clustered A to G mutations, resulting in 16 amino acid changes, were detected. This hypermutation might be due to unwinding modification of a part of the MV RNA genome accidentally present in a double-stranded form. Finally, we classified four lytic and seven persistent MV strains on the basis of their sequences. Surprisingly, the four lytic viruses considered belong to the same class. The persistent viruses form more loosely defined groups, which all differ from the vaccine strain Edmonston.

Silver staining of Pneumocystis carinii in the rat's lung.

The backscattered electron imaging mode of the scanning electron microscope was used to study the ontogenetic acquisition of argyrophilia in Pneumocystis carinii in rats. Silver staining continually increased from the late trophozoite to the mature cyst stage. The silver uptake began with a fine outline at the surface of the bodies of the late trophozoites; their cellular extensions, however, did not stain. The oblate precyst forms acquired the silver in heterogeneous patches. On spherical cysts the silver staining became more uniform and intense with at least one dense spot. The spherical and collapsed cysts also had short silver staining projections that may represent microvilli. Collapsed forms were paler than spherical ones and appear to be cysts that have undergone partial or complete release of sporozoites. These cell surface observations confirm and amplify previous transmission electron microscopical and histochemical studies indicating that silver staining correlates with the acquisition of the cell pellicle.

Quantitative assessment of human neutrophil chemiluminescence induced by opsonized Escherichia coli K-12.

The interaction of opsonized E. coli K-12 bacteria and polymorphonuclear leukocytes (PMN) was quantified, using luminol-enhanced chemiluminescence (CL) as a parameter of PMN stimulation. On a double-logarithmic scale light emission depended on the opsonin concentration used during pre-opsonisation. The most potent CL-inducing agent was fresh human serum, and its stimulatory activity depended on an intact complement (C) system. Both inactivation of C by heating or blocking the classical C pathway with EGTD decreased the CL-inducing potency by a factor of 8 to 16. Hypogammaglobulinemic heated serum mediated little CL. IgG for intravenous use mediated CL generation, but reduction/alkylation and sulphitolysis reduced the stimulatory power. Evidence is presented that the anti-K-12 antibodies within commercial IgG and IgM used for substitution do not improve the stimulatory power of IgG-deficient, IgM- and C-sufficient serum, unless very high Ig concentrations are substituted.

Pore formation by the Escherichia coli hemolysin: evidence for an association-dissociation equilibrium of the pore-forming aggregates.

Lipid bilayer experiments were performed in the presence of hemolysin of Escherichia coli. The toxin had a rather low activity in membranes formed of pure lipids, such as phosphatidylcholine or phosphatidylserine. In membranes from asolectin, a crude lipid mixture from soybean, hemolysin was able to increase the conductance by many orders of magnitude in a steep concentration-dependent fashion, which suggested that several hemolysin molecules could be involved in the conductive unit. Furthermore, the much higher toxin activity in asolectin membranes would be consistent with the assumption that this lipid contains a receptor needed for membrane activity of the toxin. The results of single-channel records showed that the membrane activity of hemolysin is due to the formation of ion-permeable channels with a single-channel conductance of about 500 pS in 0.15 M KCl. The hemolysin channel seemed to be formed by a toxin oligomer which showed an association-dissociation reaction and had a mean lifetime of about 2 s at small transmembrane voltages. The conductance of the hemolysin channels was only moderately dependent on the salt concentration in the aqueous phase. Zero-current membrane potential experiments showed that the hemolysin channel is cation selective. The mobility sequence of the cations in the channel was similar to their mobility sequence in the aqueous phase, which was consistent with the assumption that the hemolysin channel is wide and that the interior field strength is not very high. From the single-channel conductance, a lower limit of about 1.0 nm for the effective channel diameter could be estimated.

Pulmonary capillary density in rats given monocrotaline. A cast corrosion study.

A mechanism for monocrotaline-induced pulmonary hypertension in experimental animals may be the loss of small blood vessels, but when pulmonary arteries have been studied, the results have been controversial. In this report, the density of the entire lung microvasculature as measured by gray-scale analysis of scanning electron microscopic images of corrosion casts is reported. In the first experiment, monocrotaline, 60 mg/kg, was injected intraperitoneally into Sprague-Dawley rats. After 16 days, the pulmonary vascular tree was cast with methylmethacrylate. The vascular density index of the casts at a magnification x 100 on the alveolar surface was 0.40 for the monocrotaline-treated animals and 0.48 for the saline-treated control animals (p less than 0.01). On the pleural surface, it was 0.51 for the monocrotaline animals and 0.67 for the saline animals (p less than 0.001). A second experiment found no significant difference in vascular density index between different doses of monocrotaline. Light and scanning electron microscopy suggest that capillary and small vessel thrombosis may account for the decrease in the density of vessels in pulmonary microvascular casts that occurs in response to monocrotaline.

[CO2-stunning of swine for slaughter from the anesthesiological viewpoint].

For investigations of CO2-stunning of feeder- and slaughter-pigs parameters of behaviour, blood-gas-analyses and electroencephalograms were chosen. The following results were obtained: 1. Blood-gas-analyses proved that the CO2-stunning does not produce unconsciousness due to a lack of oxygen. 2. The criterias of general anaesthesia: unconsciousness, muscle-relaxation and analgesia with total reversibility could be confirmed. 3. The violent convulsive symptoms were evaluated as reactions identical with the stage II of GUEDEL's scheme of anaesthesia. 4. Muscular agitation, which sometimes appeared a few seconds before the stage of excitation, was judged to belong either to the start of the excitation phase or to the end of Guedel's stage of analgesia, during which the sensitivity is decreased. Neither study of behavior nor objective measurements showed, during the first 10 to 20 seconds of exposure to the CO2, any sign of pain or suffering related to the Act for Prevention of Cruelty to Animals, and accordingly such suffering should not be ascribed to the CO2 stunning method.

SR33557, an indolizinsulfone blocker of Ca2+ channels: identification of receptor sites and analysis of its mode of action.

SR33557 belongs to a new class of molecules (indolizinsulfones) that act on the same receptor complex that has been characterized for other classical calcium channel effectors. The main binding properties of SR33557 to rabbit skeletal muscle are as follows. (i) Unlabeled SR33557 completely inhibits the specific binding of all classes of calcium channel antagonists such as dihydropyridines [(+)-[3H]PN200-110], phenylalkylamines ([3H] verapamil), benzothiazepines (d-(cis)-[3H]diltiazem), and diphenybutylpiperidines ([3H]fluspirilene). In all these cases inhibition of binding is of a noncompetitive nature. (ii) [3H]SR33557 binds with high affinity to T tubule membranes (KD = 0.08 nM) and the maximum binding capacity (Bmax = 78 pmol/mg of protein) is the same as that found for other classes of Ca2+ channel antagonists. Photoaffinity labeling confirms that [3H]SR33557 associates with the same protein of Mr 165,000 that binds the classical calcium channel inhibitors. 45Ca2+ uptake experiments performed with the rat aortic cell line A7r5, the insulin-secreting cell line RINm5F, and the pheochromocytoma cell line PC12 demonstrate that SR33557 fully inhibits the 1,4-dihydropyridine-sensitive 45Ca2+ uptake elicited by depolarization. A very good correlation was found between inhibition of 45Ca2+ uptake and of [3H]dopamine release in PC12 cells and between inhibition of 45Ca2+ uptake and of L-type Ca2+ current in A7r5 cells under whole-cell patch-clamp conditions.

Biased hypermutation and other genetic changes in defective measles viruses in human brain infections.

We assessed the alterations of viral gene expression occurring during persistent infections by cloning full-length transcripts of measles virus (MV) genes from brain autopsies of two subacute sclerosing panencephalitis patients and one measles inclusion body encephalitis (MIBE) patient. the sequence of these MV genes revealed that, most likely, almost 2% of the nucleotides were mutated during persistence, and 35% of these differences resulted in amino acid changes. One of these nucleotide substitutions and one deletion resulted in alteration of the reading frames of two fusion genes, as confirmed by in vitro translation of synthetic mRNAs. One cluster of mutations was exceptional; in the matrix gene of the MIBE case, 50% of the U residues were changed to C, which might result from a highly biased copying event exclusively affecting this gene. We propose that the cluster of mutations in the MIBE case, and other combinations of mutations in other cases, favored propagation of MV infections in brain cells by conferring a selective advantage to the mutated genomes.

Characterization of the nucleoside-binding site inside the Tsx channel of Escherichia coli outer membrane. Reconstitution experiments with lipid bilayer membranes.

Reconstitution of purified Tsx protein from Escherichia coli into lipid bilayer membranes showed that Tsx formed small ion-permeable channels with a single-channel conductance of 10 pS in 1 M KCl. The dependence of conductance versus salt concentration was linear, suggesting that Tsx has no binding site for ions. Conductance was inhibited by the addition of 20 mM adenosine. Titration of the Tsx-mediated membrane conductance with different solutes including free bases, nucleosides, and deoxynucleosides suggested that the channel contains a binding site for nucleosides but not for sugars or amino acids, and binding increased in the following order: free base, nucleoside, and deoxynucleoside. Among the five nucleosides the stability constant for the binding increased in the order of cytidine, guanosine, uridine, adenosine, and thymidine. Control experiments revealed that the binding of the nucleosides is independent of ion concentration in the aqueous phase, i.e. there was no competition between nucleosides and ions for the binding site inside the channel. The binding of the solutes to the channel interior can be explained by a one-site two-barrier model for the Tsx channel. The advantage of a binding site inside a specific porin for the permeation of solutes is discussed with respect to the properties of a general diffusion pore.

Pore formation by pho-controlled outer-membrane proteins of various Enterobacteriaceae in lipid bilayers.

The structural genes of the PhoE porins of Klebsiella pneumonia, Enterobacter cloacae and Escherichia coli C, cloned in multicopy plasmids, were transfered into a porin-deficient E. coli K-12 strain, which was constitutive for the pho regulon, and the PhoE porins were isolated and purified. PhoE of Salmonella typhimurium could not be cloned but was isolated from a pho-constitutive strain. Reconstitution experiments with artificial lipid bilayer membranes showed that the different PhoE proteins formed pores exhibiting a single-channel conductance of about 200 pS at 0.1 M KCl. All PhoE porins formed anion-selective channels in KCl at neutral pH. The degree of the selectivity was dependent on the PhoE species. The different PhoE porins formed general diffusion pores similar to the general porins but exhibited a considerable advantage for the permeation of phosphate through the outer membrane as compared to the constitutive OmpC and OmpF porins of E. coli K-12.

Properties of structure and interaction of the receptor for omega-conotoxin, a polypeptide active on Ca2+ channels.

Binding properties of omega-conotoxin (GVIA) to avian and mammalian neuronal Ca2+ channels were investigated using a radioiodinated toxin derivative. An exceptionally high affinity of 0.6 to 2 pM was found both from equilibrium and kinetics measurements. Only one class of non-interacting binding sites was detected. In chick brain, dissucinimidyl suberate specifically cross-linked the toxin to 170 kDa component that comprises a 140 kDa peptide disulfide linked to a 30 kDa peptide, very similar to the heavily glycosylated component of the L-type DHP-sensitive Ca2+ channel. A large peptide of 210-220 kDa was labelled using the azidonitrobenzoyloxy derivative of omega-conotoxin as cross-linking reagent but not the 170/140+30 kDa component. The results suggest that the neuronal Ca2+ channel could be composed by the association of two distinct high molecular weight peptides of 220 kDa and 170/140+30 kDa.
