Cross-linking of collagen depending on age.

The spectrum of aldimine cross-link bonds of skin collagen is dependent on the localization of the examined material. In correlation to this, there is no synchronous behaviour of cross-linkage in the aging process. The age-dependent decrease of HLNL and the increase of DHLNL in the collagen of tail tendon is only unsubstantially found in the skin of neck. It is supposed that environmental conditions or bodily activity in an area might effect the cross-link bonds of collagen.

[Results concerning the sensitivity of old patients against digitalis (author's transl)].

It could be demonstrated that there was an age-dependent increase of digitalisintoxications in correlation with digitalisconcentrations in blood. This phenomenon can be explained with the frequently observed underweight in the senium. Some patients older than 70 years showed symptoms of intoxication in spite of a normal digitalisconcentration in serum. But the presented results did not demonstrate a significant difference between young and old patients concerning the sensitivity against digitalis. Therefore it is necessary to consider the bodyweight for digitalis therapy in older patients.

[The importance of body weight in treatment with digoxin and digoxin derivatives (author's transl)].

A total of 1109 determinations of digoxin concentration in serum were performed in 317 patients with cardiac failure during oral maintenance therapy with digoxin, beta-acetyldigoxin and beta-methyldigoxin. It was shown that the optimal therapeutic serum concentration (1.21 to 1.70 ng/ml) can be obtained reliably if the dosage of digoxin and its derivatives is based on the body weight. The daily doses recommended for oral maintenance therapy are 4mug/kg for beta-methyldigoxin, 5 mug/kg for beta-acetyldigoxin, and 8 mug/kg for digoxin. For initiating cardiac therapy the double maintenance dose can be prescribed once. Digoxin derivatives should be preferred to digoxin when choosing the drug.

[Hydroxylation of skin collagen in relation to age: analyses of the human skin].

The results of human skin analysis are demonstrated. It has been pointed out, that the content of hydroxyproline and hydroxylysine, in addition to the usual variation as the other aminoacids varies extensively. The extent of hydroxylation was estimated from the relation of OH-aminoacids to (not hydroxylated) each other. The determined variation of the results is most probably due to a different collagen hydroxylation within the biological dispersion. An age-related degree of hydroxylation could not be detected.

[Age-dependent metabolism and polymerisation of collagen (author's transl)].

Investigations of collagen metabolism on the one side and of crosslinking of fibres on the other side speak for the assumption that the decrease of synthesis of collagen correlates with an increase of its crosslinking. This process can be seen in the skin of aging rats. The increase of steady crosslinks bonds represents a regulation to elongate the life-span of fibres, that means a compensation for the decreased production of new collagen.

Digitalis effect and blood concentration.

Comparing cardial and extracardial effects of digitalis with corresponding digoxin blood concentrations, we determine the therapeutic range of glycosides. These studies are based on 1109 determinations in 317 patients with heart failure. The clear intoxication limit is above 2.0 ng/ml.

[Contribution to the problem of Arthrogryposis multiplex (author's transl)].

A case of Arthrogryposis multiplex appearing after birth and of no characteristical course is described. It is shown that in respect to existing works for diagnosis of Arthrogryposis multiplex the insufficiency of the hitherto existing valid diagnostic criterions is obvious. This especially because the pathogenesis of Arthrogryposis multiplex is unknown. Trying to look for the condition of the connective tissue of our patient, it was found that the collagen of skin shows a young matter of polymerisation and a nearly correlating electron microscopical picture.

Is there a recycling of hydroxyproline?

Rats can produce glycine from hydroxyproline and vice versa. After the injection, hydroxyproline is rapidly converted into glycine and then incorporated into collagen. Later the labelled amino acids in collagen are hydroxyproline, proline, serine, threonine, alanine, and glutamic acid. We suppose that all these labelled amino acids come from glycine.

The dynamics of collagen deposition in liver damaged by CCl4.

The rates of synthesis and catabolism of collagen are determined in the livers of normal rats and of rats treated with carbon tetrachloride. Several experimental conditions are applied with the intention of investigating the dynamic processes separately, in order to determine the relevant values. The deposition of collagen in liver after damage by CCl4 is a consequence of the reduction of turnover with a simultaneous increase in collagen-production. Together with the increase in synthesis, which seems to be a consequence of a numerical increase in cells, there is a reduction in catabolism of collagen: the more severe the damage, the more marked is the decrease of specific catabolism. Collagen-deposition in the liver is thus the result of accumulation. The results indicate the possibility that collagen-catabolism is effected by enzymes produced by adjacent parenchymal cells, so that their destruction enhances the accumulation of collagen material. The findings mentioned above are discussed in the context of the general criteria of scar-formation and the repair of damaged tissues.

[Protein and amino acid composition of newborn rats under defective nutritional conditions].

The law protein nutrition of the nursing rat inhibits the growth of the new born rats. A percentage reduction of the dry substances by decrease of N2-free substances takes place. In relation to g dry substance the body protein content, collagen also, rises. Absolutely, these parts rest unaltered, just so the relation of the parenchymatous protein to collagen. The often support of view that starving provokes a collagen formation of the body, that means an exceed of the framework, can not be confirmed. Though in this paper we can give an explanation for the occurrence of this conclusion. Long interuptions of lactation periods remarkably have no effect on the growth and on the amin compartments of the body of new born rats. Only tryptophane is reduced in favour of the serine-fraction in the body of those animals. We discuss this finding as a change of the metabolisme of tryptophane depending of stress.
