VIP-like and GFAP-like immunoreactivities in the chicken brain stem. I. The medulla.

An immunocytochemical analysis was performed on the chicken medulla according to the PAP-DAB procedure, to study the distribution here of the neurons reacting to anti-VIP antibodies and the gliocytes reacting to anti-GFAP antibodies. Positive and negative controls were carried out in both the immunoreactions. The findings seem to indicate that more numerous VIP-immunoreactive neurons are located in the chicken than in the rat or mouse medulla, mainly in its intermediate 1/3, that VIP-immunoreactive fibres connect the medullar group of immunoreactive neurons with at least the first three cervical neuromeres, and confirm also for the chicken medulla, as already reported in other species by various researchers, the close relation of some VIP-immunoreactive neuronal processes with the small blood vessel walls, and the distribution of the GFAP-immunoreactive gliocytes.

VIP-like and GFAP-like immunoreactivities in the chicken brain stem. II. The pons and the tegmentum of the mesencephalon.

An immunocytochemical analysis was performed on the chicken pons and mesencephalon (except the optic tectum) according to the PAP-DAB procedure, to study the distribution here of the neurons reacting to anti-VIP antibodies and the gliocytes reacting to anti-GFAP antibodies. Positive and negative controls were carried out in both the immunoreactions. The VIP-immunoreactive neurons showed a distribution essentially corresponding to that observed in other species by various Authors. They appeared scattered mainly in 3 sites: (a) the subventricular grey between pons and mesencephalon; (b) the periaqueductal grey, up to the diencephalon; (c) the rostro-ventral portion of the mesencephalic tegmentum, up to the diencephalon. Furthermore, some perivascular VIP-immunoreactive neuronal processes were seen. No differences have so far been detected as regards the GFAP-like immunoreactivity distribution, in comparison with the data reported by the authors in the chicken medulla and by others in in the brain stem of some other species.

[Histomorphogenesis of spinal ganglia developing on the chorioallantoic membrane].

The aim of the present research is to study the development of spinal ganglia implanted onto chorio-allantoic membrane (CAM), where they are supplied by 'foreign', extraembryonic vessels. Small blocks including a segment of the neural tube and 4 pairs of somites were isolated from the brachial or thoracic or lumbar region (neuromeres C13 - T1 or T3 - T6 or L1 - L4) of thirty 3-day donor chick embryos and grafted on the CAM of as many 10-day host chick embryos. At regular time intervals after grafting, the implants were fixed, embedded in paraffin, serially cross-sectioned and stained. The preliminary results are similar for the ganglia from the 3 regions: they seem to indicate that in the spinal ganglia grafted on the CAM the developing blood vessels do not exert any morphogenetic influence on the substratum; this corroborates results from previous researches (2) on the normally 'in situ' developing ganglia. It is now the author purpose to submit to a careful analysis a datum from the present research: namely, the earlier appearance of the endoganglionic vessels in the brachial and lumbar grafted ganglia than in the thoracic ones.

GABA immunoreactivity in the human cerebellar cortex: a light and electron microscopical study.

The distribution of gamma-aminobutyric acid (GABA) in surgical samples of human cerebellar cortex was studied by light and electron microscope immunocytochemistry using a polyclonal antibody generated in rabbit against GABA coupled to bovine serum albumin with glutaraldehyde. Observations by light microscopy revealed immunostained neuronal bodies and processes as well as axon terminals in all layers of the cerebellar cortex. Perikarya of stellate, basket and Golgi neurons showed evident GABA immunoreactivity. In contrast, perikarya of Purkinje neurons appeared to be negative or weakly positive. Immunoreactive tracts of longitudinally- or obliquely-sectioned neuronal processes and punctate elements, corresponding to axon terminals or cross-sectioned neuronal processes, showed a layer-specific pattern of distribution and were seen on the surface of neuronal bodies, in the neuropil and at microvessel walls. Electron microscope observations mainly focussed on the analysis of GABA-labelled axon terminals and of their relationships with neurons and microvessels. GABA-labelled terminals contained gold particles associated with pleomorphic vesicles and mitochondria and established symmetric synapses with neuronal bodies and dendrites in all cortex layers. GABA-labelled terminals associated with capillaries were seen to contact the perivascular glial processes, basal lamina and endothelial cells and to establish synapses with subendothelial unlabelled axons.

[A case of pseudosarcoma of the esophagus (author's transl)].

Pseudosarcoma of the esophagus previously has been described and documented in only 13 patients. Our case, after a successful total esophagectomy with esophagogastroplasty, is physically well without any symptoms of recurrence to date (18 months postoperative).

Mitomycin-C and lonidamine as second-line therapy for colorectal cancer: a phase II study.

AIMS AND BACKGROUND: Recent preclinical data have suggested that lonidamine may potentiate the acitivity of mitomycin C in human colon cancer cell lines LoVo and HT29. STUDY DESIGN: A phase II study was carried out in 14 patients with advanced colorectal cancer pretreated with fluorouracil and folinic acid. Treatment consisted of lonidamine, 600 mg po, followed after 2 h by mitomycin, 20 mg/m2 by iv bolus, followed by lonidamine, 150 mg tio for 5 days; the cycle was repeated every 6 weeks. RESULTS: No objective response was seen. Three patients had stable disease; the median survival for the whole group was 4 months. Although hematologic toxicity was negligible, lonidamine-related side effects were moderate to severe in most patients and mainly represented by myalgia and gastric pain. DISCUSSION: Despite a sound preclinical rationale, this schedule of lonidamine and mitomycin C was ineffective and toxic in patients with advanced colorectal cancer. More experimental data about lonidamine are needed in order to design more effective regimens based on the combination of this interesting drug with other anticancer agents.
