Construction of Saccharomyces cerevisiae strains that accumulate relatively low concentrations of trehalose, and their application in testing the contribution of the disaccharide to stress tolerance.

Genetically related diploid strains of Saccharomyces cerevisiae that accumulate varied amounts of trehalose during starvation for nitrogen have been constructed. Strains that produced greater than 5% trehalose (dry cell weight) were more tolerant of thermal, or freeze-thaw stresses than strains that produced less than 4% trehalose. Thus trehalose appears to play a role in stress tolerance of yeast. The significance of these results is that, for the first time, a series of related, unmutated strains have been used to test the effect of trehalose on thermotolerance. Previous studies employed either heat shock treatment, or mutated strains to provide trehalose variations, and as such the contribution of the disaccharide to stress tolerance could not necessarily be separated from other factors such as heat shock proteins.

Purification, crystallization, and X-ray diffraction studies of lactotransferrin from buffalo colostrum.

Lactotransferrin is an iron-binding protein. It has been purified from buffalo colostrum. The purified lactotransferrin has been crystallized in 10% ethanol solution. The crystals are orthorhombic and the space group is P2(1)2(1)2(1) with unit cell dimensions a = 161.70 A, b = 155.75 A, c = 113.48 A. The asymmetric unit contains three molecules of the protein with a solvent content of about 59%. The crystals were stable in the X-ray beam and diffract beyond 3.5 A resolution. The native data have been collected and the structure determination is in progress.

Antimicrobial activities of the stembark of Kigelia pinnata.

Chemical investigation showed that the aqueous extracts of the stembark of Kigelia pinnata contain iridoids as major components. In the light of the traditional uses of this plant, antimicrobial activities of the aqueous extracts and two major iridoids were tested against Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Candida albicans. The crude aqueous extracts showed significant antimicrobial activity which could be partially explained by the activity of the iridoids present.

Ventilatory function in Malay Muslims during normal activity and the Ramadan fast.

Pulmonary function parameters were examined in a Malay Muslim population during normal activity and Ramadan fasting conditions. The validity of employing various lung function prediction formulae for the subjects was also assessed. Present findings indicate that the water deprivation regime and resultant dehydration during Ramadan did not cause significant changes in ventilatory functions. Although pulmonary prediction formulae based on Caucasian and African populations were inapplicable to the subjects examined, the equations derived from the neighbouring populations in Singapore could be employed.

Effects of varying dialysate calcium concentrations on the plasma calcium fractions in patients on dialysis.

The plasma-ionized calcium levels decreased during haemodialysis when a dialysate calcium concentration of 5 mg/100 ml was used. When dialysis was performed with a bath calcium concentration of 7.5 mg/100 ml, there was a significant increase in the plasm-ionized calcium levels in the post-dialysis period. These results are discussed in relation of the optimal dialysate calcium concentrations and development of dialytic bone disease.
